Multiple pathways participate in the AMPA receptor trafficking that underlies long-term potentiation (LTP) of synaptic transmission. Here we demonstrate that protein SUMOylation is required for insertion of the GluA1 AMPAR subunit following transient glycine-evoked increase in AMPA receptor surface expression (ChemLTP) in dispersed neuronal cultures. ChemLTP increases co-localisation of SUMO-1 and the SUMO conjugating enzyme Ubc9 and with PSD95 consistent with the recruitment of SUMOylated proteins to dendritic spines. In addition, we show that ChemLTP increases dendritic levels of SUMO-1 and Ubc9 mRNA. Consistent with activity dependent translocation of these mRNAs to sites near synapses, levels of the mRNA binding and dendritic transport protein CPEB are also increased by ChemLTP. Importantly, reducing the extent of substrate protein SUMOylation by overexpressing the deSUMOylating enzyme SENP-1 or inhibiting SUMOylation by expressing dominant negative Ubc9 prevent the ChemLTP-induced increase in both AMPAR surface expression and dendritic SUMO-1 mRNA. Taken together these data demonstrate that SUMOylation of synaptic protein(s) involved in AMPA receptor trafficking is necessary for activity-dependent increases in AMPAR surface expression. 相似文献

Bacillus subtilis is an aerobic endospore forming bacterium widely spread in different environments. Because it represents a biological agent of some health relevance, its rapid detection and identification is highly desirable. By using FT-IR spectroscopy for this purpose slightly different characteristics were obtained from cell mass grown in differently composed cultural media, and harvested in different phases of bacterial growth. If cultivated uniformly, i.e., 24h at 30 degrees C in a minimum-strength nutrient broth, cell mass of B. subtilis delivered a well differentiated spectrum with major absorption bands of nucleic acid structures at 3300cm(-1), cell wall constituents at 3000-2800cm(-1), proteinaceous structures at 1660, 1544 and 1235cm(-1), and some aliphatic structural units at 1080cm(-1). Attenuated total reflectance, and absorption/transmission scanning techniques, delivered structurally identical spectra but those obtained by the former technique were more expressed. 相似文献

Background
Malignant mesothelioma cells have an epithelioid or sarcomatoid morphology, both of which may be present in the same tumor. The sarcomatoid phenotype is associated with worse prognosis and heterogeneity of mesothelioma cells may contribute to therapy resistance, which is often seen in mesothelioma. This study aimed to investigate differences in sensitivity between mesothelioma cell lines to anti-cancer drugs. We studied two novel drugs, selenite and bortezomib and compared their effect to four conventional drugs. We also investigated the immunoreactivity of potential predictive markers for drug sensitivity; Pgp, MRP-1, ERCC1, RRM1, TS, xCT and proteasome 20S subunit.Materials and methods
We treated six mesothelioma cell lines with selenite, bortezomib, carboplatin, pemetrexed, doxorubicin or gemcitabine as single agents and in combinations. Viability was measured after 24 and 48 hours. Immunocytochemistry was used to detect predictive markers.Results
As a single agent, selenite was effective on four out of six cell lines, and in combination with bortezomib yielded the greatest response in the studied mesothelioma cell lines. Cells with an epithelioid phenotype were generally more sensitive to the different drugs than the sarcomatoid cells. Extensive S-phase arrest was seen in pemetrexed-sensitive cell lines. MRP-1 predicted sensitivity of cell lines to treatment with carboplatin and xCT predicted pemetrexed effect.Conclusions
The observed heterogeneity in sensitivity of mesothelioma cell lines with different morphology highlights the need for more individualized therapy, requiring development of methods to predict drug sensitivity of individual tumors. Selenite and bortezomib showed a superior effect compared to conventional drugs, motivating clinical testing of these agents as future treatment regime components for patients with malignant mesothelioma. 相似文献994.

In the work presented here, changes in haematopoiesis of mice (B6129SF2/J) were studied 1 year after their whole-body exposure
to a dose of 7 Gy (72% of mice survived). The irradiated mice were compared with non-irradiated younger (4 months of age)
and older (16 months of age) mice. There was a significant increase in the relative abundance of primitive stem cells with
long-term capability of the haematopoiesis recovery lin−/Sca-1+/CD117+/CD34− in the bone marrow of mice aged 16 months (irradiated and non-irradiated) compared with those aged 4 months. In terms of
the ability to respond to further whole-body irradiation at a dose of 1 Gy, the presence of γH2A.X foci was studied in lin− bone marrow cells. There was a considerable number of persisting foci in lin− stem cells isolated from the bone marrow of the older irradiated mice. In the blood count from the peripheral blood of the
older mice (both non-irradiated and irradiated at 7 Gy), there was a significant increase in granulocytes. In the group exposed
to 7 Gy, the numbers of thrombocytes significantly increased, and on the contrary, the numbers of erythrocytes, the amount

The authors used museomics to reconstruct the mitochondrial genome from two individuals of the Moroccan, endemic and extinct trout, Salmo pallaryi. They further obtained partial data from 21 nuclear genes previously used for trout phylogenetic analyses. Phylogenetic analyses, including publicly available data from the mitochondrial control region and the cytochrome b gene, and the 21 nuclear genes, place S. pallaryi among other North African trouts. mtDNA places S. pallaryi close to Salmo macrostigma within a single North African clade. Although the nuclear coverage of the genome was low, both specimens were independently positioned as sisters to one of two distantly related North African clades, viz. the Atlas clade with the Dades trout, Salmo multipunctatus. Phylogenetic discordance between mtDNA and nuclear DNA phylogenies is briefly discussed. As several specimens that were extracted failed to produce DNA of sufficient quality, the authors discuss potential reasons for the failure. They suggest that museum specimens in poor physical condition may be better for DNA extraction compared to better-preserved ones, possibly related to the innovation of formalin as a fixative before ethanol storage in the early 20th century. 相似文献

Stream nitrogen (N) export and nitrate concentration were measured at 14 forested watersheds (GEOMON network) in the Czech Republic between 1994 and 2005. In the
last several decades, emissions of sulfur (S) and N compounds have decreased throughout much of Europe. In the Czech Republic,
atmospheric deposition of S has decreased substantially since the beginning of 1990s, whereas N deposition remains largely
unchanged at most sites. The mean dissolved inorganic nitrogen (DIN) streamwater export ranged from 0.2 to 12.2 kg ha−1 y−1 at the GEOMON sites. Despite decades of elevated N deposition, 44–98% of DIN inputs to these watersheds were retained or
denitrified, and many watersheds showed seasonal variation in nitrate concentrations. Dissolved organic N export was quantified
in 1 year only and ranged from 0.05 to 3.5 kg ha−1 y−1. Spatial variability in DIN export among watersheds was best explained by spatial variability in average acidic deposition,
particularly S deposition (R
2 = 0.81, P < 0.001); DIN input and forest floor carbon:nitrogen (C/N) also provided significant explanatory power. DIN export was strongly
influenced by the forest floor C/N ratio and depth of the forest floor soils (R
2 = 0.72, P < 0.001). The only variable that predicted variations in forest floor C/N (R
2 = 0.32, P < 0.05) among watersheds was S deposition. Forest floor depth was also related to deposition variables, with S deposition
providing the most explanatory power (R
2 = 0.50, P < 0.01). Variation in forest floor depth was also associated with climatic factors (precipitation and temperature). Temporal
variability in DIN export was primarily associated with changes in acidic deposition over time; S deposition explained 41%
of variability in DIN exports among all watersheds and years. Extensive acidification of forested watersheds was associated
with the extraordinarily high S inputs to much of the Czech Republic during earlier decades. We hypothesize that recovery
from acidification has led to improved tree health as well as enhanced microbial activity in the forest floor. As these watersheds

The O-polysaccharide of Proteus vulgaris O44, strain PrK 67/57 was studied by 1H and 13C NMR spectroscopy, including 2D COSY, TOCSY, ROESY, H-detected 1H, 13C HMQC, HMQC-TOCSY and HMBC experiments. The polysaccharide was found to contain an amide of D-glucuronic acid with L-alanine [D-GlcA6(L-Ala)], and the following structure of the linear pentasaccharide repeating unit was established: [structure: see text]. The structural data of the O-polysaccharide and the results of serological studies with P. vulgaris O44 O-antiserum showed that the strain studied is unique among Proteus bacteria, which is in agreement with its classification in a separate Proteus serogroup, O44. 相似文献

Seed size is a crucial life‐history trait determining the amount of reserves that are available to establishing seedlings. The most frequently observed patterns in seed size distribution are a higher frequency of large‐seeded species in shaded habitats and a positive correlation of seed size with plant size. We analysed to what extent realised niche dimensions, as expressed by Ellenberg indicator values and plant functional traits such as plant height and life form, explained seed mass variation in the central European flora. By including information on phylogenetic relatedness of the species, not only contemporary ecology but also the evolutionary history of plant species could be taken into account. Seed mass evolution was slow and was best explained by selection‐inertia models with multiple adaptive peaks as a function of either habitat or life form. The highest seed mass optima were observed in the deciduous forest and saltwater and seashore habitats, and in phanerophytes in case of models with optima as a function of life form. The analyses showed that Ellenberg values were more important than habitat and life form in explaining seed mass distribution in the central European flora. The often observed relation between shade and large seeds was also evident in our study, but we found an equally important relation between large seeds and drought and a positive relation between seed mass and salinity. Our results indicate that not only plant size and competition for light but a complex set of factors influence the ecology of seed size, and that a more precise delineation of species’ niches improves the understanding of seed size evolution. 相似文献
