Growth and sporulation of Oidium begoniae (begonia powdery mildew) on detached leaves was observed daily under a stereomicroscope. A mathematical model of growth and sporulation was constructed based on the patterns observed. The essential components of the model are three genetically fixed components of colony growth and sporulation and six components which changed with environmental and physiological conditions. The genetically fixed components were: the ultimate number of germ tubes, the hyphal growth unit (HGU or mean length supporting one branch), and the diurnal periodicity of sporulation. These components were shown to have approximately the same values at 15 and 21 °C. Components which varied with temperature were: the latent periods of germination, branching and sporulation, the time taken to reach the ultimate germ tube number, and the number of conidiophores per HGU, The derived equations were tested against observed sporulation and resulted in highly significant correlations. Similar results were obtained with published observations for Erysiphe graminis f. sp. hordei. This colony-level model can be used as units to construct models of disease progress and suggests that it is theoretically possible to model disease in the field from laboratory studies. The model can be used in computer simulations of colony growth as demonstrated by a computer program in the Appendix.  相似文献

The objective was to evaluate the effects of growth hormone (GH) on the survival, growth, maturation, and fertilization of oocytes derived from caprine preantral ovarian follicles cultured in vitro. Preantral follicles were isolated from the cortex of caprine ovaries and individually cultured for 18 d in the absence (control) or presence of bovine GH at concentrations of 10 or 50 ng/mL (GH10 and GH50, respectively). Follicle development was evaluated on the basis of survival, antral cavity formation, diameter increase, and the presence of healthy cumulus-oocyte complexes and mature oocytes. After culture, oocytes were subjected to in vitro maturation (IVM) and in vitro fertilization (IVF). The rate of antrum formation after Day 6 of culture was higher in both GH10 and GH50 than in the control (81.0, 92.7, and 47.6%, respectively, P < 0.05). Percentages of grown oocytes that were acceptable for IVM were also higher (P < 0.05) in GH-treated groups than in the control (54.8, 48.8, and 11.9% for GH10, GH50, and Control). A higher percentage of oocytes in the GH50 treatment underwent meiotic resumption (50.0%), produced mature oocytes, and enabled production of an embryo after IVF than in the control group (0.0%; P < 0.05). In conclusion, GH promoted in vitro growth and maturation of goat preantral follicle oocytes and enabled production of an embryo. Furthermore, this study was apparently the first to produce a caprine embryo by in vitro fertilization of oocytes derived from preantral follicles grown in vitro.  相似文献

Abstract: Artificial nesting islands, or rafts, are often deployed in common loon (Gavia immer) breeding territories to decrease negative impacts of mammalian predation and water-level fluctuations on nesting success. The management value of rafts has been demonstrated in other studies; however, no published studies have quantified the use or associated reproductive benefits of rafts on lakes exhibiting water-level fluctuations. These lakes constitute a major portion of loon nesting habitat in New England and the Midwest. We used long-term data sets from loon survey and raft management efforts on lakes with stable (SWL) and fluctuating water levels (FWL) in New Hampshire and Maine, USA, to compare raft-use patterns on both types of lakes. We then modeled the influence of percentage of nesting attempts on rafts, lake fluctuation type, and human development index on nesting success as a function of the number of nesting attempts. Loons used 76% of all rafts for nesting, and initial use patterns were similar between SWL and FWL lakes. Half (51%) of rafts used for nesting were first used during the initial year of deployment and 90% of those used were used by the third year. Based on our model, we would expect to see an 8.6% increase in nesting success associated with each successive categorical increase in raft use (0–33%, 33–60%, 60–100%). Nesting success varied with lake fluctuation type, increasing by 21.4% from FWL to SWL types. Our model estimated a 12.8% decrease in nesting success associated with an increasing human development index. Naturally nesting loons on FWL lakes are likely to display mean nesting success levels lower than those needed to sustain populations. We suggest that natural nesting habitat on lakes with fluctuating water levels during the loon nesting season may constitute an ecological trap warranting consideration of raft management. Findings in this study are germane for managing breeding loon populations, particularly those on reservoirs requiring permits from the Federal Energy Regulatory Commission.  相似文献

The nitroheterocyclic drugs nifurtimox and benznidazole are first-line drugs available to treat Chagas disease; however, they have limitations, including long treatment courses and toxicity. Strategies to overcome these limitations include the identification of new drugs with specific target profiles, re-dosing regimens for the current drugs, drug repositioning and combination therapy. In this work, we evaluated combination therapy as an approach for optimization of the current therapeutic regimen for Chagas disease. The curative action of benznidazole/itraconazole combinations was explored in an established infection of the mice model with the T. cruzi Y strain. The activities of the benznidazole/itraconazole combinations were compared with the results from those receiving the same dosage of each individual drug. The administration of benznidazole/itraconazole in combination eliminated parasites from the blood more efficiently than each drug alone. Here, there was a significant reduction of the number of treatment days (number of doses) necessary to induce parasitemia suppression with the benznidazole/itraconazole combination, as compared to each compound administered alone. These results clearly indicate the enhanced effects of these drugs in combination, particularly at the dose of 75 mg/kg, as the effects observed with the drug combinations were four times more effective than those of each drug used alone. Moreover, benznidazole/itraconazole treatment was shown to prevent or decrease the typical lesions associated with chronic experimental Chagas disease, as illustrated by similar levels of inflammatory cells and fibrosis in the cardiac muscle tissue of healthy and treated mice. These results emphasize the importance of exploring the potential of combination treatments with currently available compounds to specifically treat Chagas disease.  相似文献

Abstract: The widespread and ongoing declines of North American bird populations that have affinities for grassland and grass-shrub habitats (hereafter referred to as grassland birds) are on track to become a prominent wildlife conservation crisis of the 21st century. There is no single cause responsible for the declines of grassland birds. Rather, a cumulative set of factors such as afforestation in the eastern United States, fragmentation and replacement of prairie vegetation with a modern agricultural landscape, and large-scale deterioration of western U.S. rangelands are the major causes for these declines. The North American Bird Conservation Initiative (NABCI) is a set of comprehensive and coordinated strategic actions modeled on the Joint Venture initiatives that were used to successfully implement the North American Waterfowl Management Plan. The NABCI is emerging as a potential broad-scale solution for conserving populations of grassland birds. Coordinating grassland bird conservation efforts with initiatives to stabilize and increase upland game birds that have strong affinities for grassland habitats—such as quail and prairie grouse—presents additional opportunities to leverage funding and resources that will positively impact virtually all species of North American grassland birds.  相似文献

Models of Mobile Animal Populations (MAP models) simulate long-term land use changes, population trends and patterns of biological diversity on landscapes of 103-105 ha. MAP models can incorporate information about past land-use patterns and management practices and can project future patterns based on management plans. We illustrate this approach with an example of how implementation of a U.S. Forest Service management plan at the Savannah River Site in South Carolina, U.S.A., might influence population trends of Bachman's Sparrow Aimophila aestivalis, a relatively rare and declining species in southeastern pine forests. In this case, a management plan, largely designed to improve conditions for an endangered species, Red-cockaded Woodpecker Picoides borealis, may have a negative impact, at least in the short term, on another species of management concern, Bachman's Sparrow.
In a parallel processing version of the MAP models, a single landscape that would ordinarily be too large or detailed to be simulated on a single computer is subdivided into a number of smaller landscapes, and each landscape is simulated in parallel, either on a single multi-tasking machine or on a group of networked machines. With this approach we are attempting to determine just how large a landscape must be before the dynamics of a population within it are more or less independent of factors beyond the landscape boundaries.  相似文献

Upon adhesion to laminin-111, aortic smooth muscle cells initially form membrane protrusions with an average diameter of 2.9μm. We identified these protrusions also as subcellular areas of increased redox potential and protein oxidation by detecting cysteine sulfenic acid groups with dimedone. Hence, we termed these areas oxidative hot spots. They are spatially and temporally transient during an early stage of adhesion and depend on the activity of the H(2)O(2)-generating NADPH oxidase 4. Presumably located on cellular protrusions, integrin α7β1 mediates adhesion and migration of vascular smooth muscle cells to laminins of their surrounding basement membrane. Using protein chemistry and mass spectrometry, two specific oxidation sites within the integrin α7 subunit were identified: one located in its genu region and another within its calf 2 domain. Upon H(2)O(2) treatment, two cysteine residues are oxidized thereby unlocking a disulfide bridge. The genu region is a hinge, around which the integrin domains pivot between a bent/inactive and an upright/active conformation. Also, cysteine oxidation within the calf 2 domain permits conformational changes related to integrin activation. H(2)O(2) treatment of α7β1 integrin in concentrations of up to 100μM increases integrin binding activity to laminin-111, suggesting a physiological redox regulation of α7β1 integrin.  相似文献

In this work the parameters of Low Temperature Conversion - LTC were applied in a centrifuged sludge from a sewage treatment plant located in Rio de Janeiro, Brazil. Before the conversion, the sludge was dried and analyzed by TGA to observe its behavior with increasing temperature. The chemical composition of the crude pyrolysis oil was analyzed by FTIR, 1H NMR and GC-MS. The results showed that the oil is a mixture of hydrocarbons, oxygenated and nitrogenated compounds. Using a catalytic treatment it was possible to fractionate the oil where the predominant constituents were hydrocarbons showing that the cracking was effective. An important result was the difference between the calorific value of dry sludge (10 MJ kg−1), the pyrolysis oil (36 MJ kg−1) and one of the fractions separated by catalytic cracking (40 MJ kg−1) when compared with commercial diesel (45 MJ kg−1).  相似文献
