A fluorescence-labeled wheat germ agglutinin staining technique (R. K. Sizemore et al., Appl. Environ. Microbiol. 56:2245-2247, 1990) was modified and found to be effective for staining gram-positive, acidophilic mining bacteria. Bacteria identified by others as being gram positive through 16S rRNA sequence analyses, yet clustering near the divergence of that group, stained weakly. Gram-negative bacteria did not stain. Background staining of environmental samples was negligible, and pyrite and soil particles in the samples did not interfere with the staining procedure.  相似文献

Objective

The purpose was to examine differences between taekwondo kicks and boxing punches in resultant linear head acceleration (RLA), head injury criterion (HIC15), peak head velocity, and peak foot and fist velocities. Data from two existing publications on boxing punches and taekwondo kicks were compared.

Methods

For taekwondo head impacts a Hybrid II Crash Dummy (Hybrid II) head was instrumented with a tri-axial accelerometer mounted inside the Hybrid II head. The Hybrid II was fixed to a height-adjustable frame and fitted with a protective taekwondo helmet. For boxing testing, a Hybrid III Crash Dummy head was instrumented with an array of tri-axial accelerometers mounted at the head centre of gravity.

Results

Differences in RLA between the roundhouse kick (130.11±51.67 g) and hook punch (71.23±32.19 g, d = 1.39) and in HIC15 (clench axe kick: 162.63±104.10; uppercut: 24.10±12.54, d = 2.29) were observed.

Conclusions

Taekwondo kicks demonstrated significantly larger magnitudes than boxing punches for both RLA and HIC.  相似文献

Summary Eight hitherto unrecognizedArizona serotypes (5∶13, 15; 5∶27∶28; 13∶1, 6, 7; 19∶29∶25; 22∶1, 2, 5; 22∶23∶31; 26∶24∶25; and 30∶22∶21) are described. Maintenance of an optimal redox environment is critical for appropriate functioning of cellular processes and cell survival. Despite the importance of maintaining redox homeostasis, it is not clear how the optimal redox potential is sensed and set, and the processes that impact redox on a cellular/organellar level are poorly understood. The genetic bases of cellular redox homeostasis were investigated using a green fluorescent protein (GFP) based redox probe, roGFP2 and a pH sensitive GFP-based probe, pHluorin. The use of roGFP2, in conjunction with pHluorin, enabled determination of pH-adjusted sub-cellular redox potential in a non-invasive and real-time manner. A genome-wide screen using both the non-essential and essential gene collections was carried out in Saccharomyces cerevisiae using cytosolic-roGFP2 to identify factors essential for maintenance of cytosolic redox state under steady-state conditions. 102 genes of diverse function were identified that are required for maintenance of cytosolic redox state. Mutations in these genes led to shifts in the half-cell glutathione redox potential by 75-10 mV. Interestingly, some specific oxidative stress-response processes were identified as over-represented in the data set. Further investigation of the role of oxidative stress-responsive systems in sub-cellular redox homeostasis was conducted using roGFP2 constructs targeted to the mitochondrial matrix and peroxisome and E(GSH) was measured in cells in exponential and stationary phase. Analyses allowed for the identification of key redox systems on a sub-cellular level and the identification of novel genes involved in the regulation of cellular redox homeostasis.  相似文献

Roots of sugar beets grown in liquid culture excrete substances that stimulate egg hatch and emergence of larvae from cysts of Heterodera schachtii. Their hatching effect is comparable to that of sugar beet root diffusate leached from soil-grown sugar beet plants. Consequently, liquid culture provides a way of obtaining H. schachtii hatch-stimulant free of contaminants from soil. Root diffusate, concentrated 50-fold or dried by vacuum distillation, retained hatching activity. The active principle of diffusate is dialyzable with a diffusion rate between those of inorganic salts and compounds with molecular weights greater than 15,000.  相似文献

The effect of nitrogen fertilisation on growth, foliar nutrients and water relations of four families of radiata pine (Pinus radiata, D. Don) currently in the Australian breeding program was examined from age six to 11 years. At this stage, the stand was ready for commercial thinning. The annual rainfall at the site varied from 563 to 733 mm.Application of nitrogen fertiliser resulted in stem wood volume at age nine years of 178 m3 ha-1 in the controls, compared with 228 m3 ha-1 in plots treated with 600 kg N ha-1. Pre-dawn needle water potential () measured in three consecutive summers (when rainfall ranged from 53 to 106 mm) were consistently higher (less water stress) in nitrogen fertilised than in control trees. Similarly, the water stress integral (S) decreased consistently with increasing levels of nitrogen, although total water use in fertilised trees would have been substantially higher because fertiliser application increased the leaf area index. The relationship between S and basal area was strong and paralleled that of foliar nitrogen concentration and basal area growth. Therefore, nitrogen application increased growth rates of trees by improving the nutrient status of trees and lowering the water stress on trees in summer.Families showed markedly different responses of basal area growth to nitrogen, ranging from an increase of 9.4% over three years for the least responsive family to 99.0% for the most responsive. There was no nitrogen × family interaction on or S suggesting that the large genetic variation in the growth response to nitrogen is mediated by factors other than water relations. These results have implications for managing highly productive plantations grown in an environment where rainfall is low compared to potential evapotranspiration.  相似文献
