Soil-Atmosphere Methane Exchange in Undisturbed and Burned Mediterranean Shrubland of Southern Italy
Soils represent the primary biotic sink for atmospheric methane (CH4). Uncertainty is associated, however, with global soil CH4 consumption because of the few data available from many areas and, in particular, from Mediterranean-type ecosystems. In this study, soil-atmosphere CH4 exchange was measured for one year in a coastal Italian shrubland (maquis), from both undisturbed areas and areas treated with experimental fire. Although fire represents one of the most frequent disturbance factors in seasonally dry environments, very few studies in these ecosystems have focused on its effect on soil CH4 fluxes. Significant differences in soil ammonium content, water content, and temperature were measured between burned and unburned plots, however, no statistical differences were observed for CH4 fluxes. CH4 fluxes varied between −0.39 and −16.1 mg CH4 m−2 day−1 and temporal variations were mainly driven by variations in soil water content and temperature. The highest CH4 oxidation rates were measured during the driest and warmest period. Low gravimetric soil water content in the top 10 cm, as well as high NH4+ concentration, did not seem to reduce methanotrophic activity, suggesting that maximal CH4 oxidation activity might take place deeper in the soil profile, at least during part of the year. 相似文献

In this work, we describe four new micromammal sites in the northern side of the Gormaget ravine, in the Alcoy Basin (Spain): AF-1’06, AF-1’07, AF-1A and AF-2. Based on the study of the faunal remains from these localities, we infer a latest Turolian-earliest Ruscinian age for AF-1’06 and AF-1A, and an earliest Ruscinian age for AF-1’07 and AF-2. The species assemblage of the locality AF-1’06, the only one which have yielded a sufficient number of remains to perform a palaecological analysis, shows warm and dry conditions near the Mio-Pliocene boundary in the Alcoy Basin. These data show a reduction of dry and warm indicators from older to younger localities in the Alcoy Basin, suggesting a change to colder and more humid conditions during the Early Pliocene. 相似文献

Whether to commit limited cellular resources toward growth and proliferation, or toward survival and stress responses, is an essential determination made by Target of Rapamycin Complex 1 (TORC1) for a eukaryotic cell in response to favorable or adverse conditions. Loss of TORC1 function is lethal. The TORC1 inhibitor rapamycin that targets the highly conserved Tor kinase domain kills fungal pathogens like Candida albicans, but is also severely toxic to human cells. The least conserved region of fungal and human Tor kinases are the N-terminal HEAT domains. We examined the role of the 8 most N-terminal HEAT repeats of C. albicans Tor1. We compared nutritional- and stress responses of cells that express a message for N-terminally truncated Tor1 from repressible tetO, with cells expressing wild type TOR1 from tetO or from the native promoter. Some but not all stress responses were significantly impaired by loss of Tor1 N-terminal HEAT repeats, including those to oxidative-, cell wall-, and heat stress; in contrast, plasma membrane stress and antifungal agents that disrupt plasma membrane function were tolerated by cells lacking this Tor1 region. Translation was inappropriately upregulated during oxidative stress in cells lacking N-terminal Tor1 HEAT repeats despite simultaneously elevated Gcn2 activity, while activation of the oxidative stress response MAP kinase Hog1 was weak. Conversely, these cells were unable to take advantage of favorable nutritional conditions by accelerating their growth. Consuming oxygen more slowly than cells containing wild type TOR1 alleles during growth in glucose, cells lacking N-terminal Tor1 HEAT repeats additionally were incapable of utilizing non-fermentable carbon sources. They were also hypersensitive to inhibitors of specific complexes within the respiratory electron transport chain, suggesting that inefficient ATP generation and a resulting dearth of nucleotide sugar building blocks for cell wall polysaccharides causes cell wall integrity defects in these mutants. Genome-wide expression analysis of cells lacking N-terminal HEAT repeats showed dysregulation of carbon metabolism, cell wall biosynthetic enzymes, translational machinery biosynthesis, oxidative stress responses, and hyphal- as well as white-opaque cell type-associated genes. Targeting fungal-specific Tor1 N-terminal HEAT repeats with small molecules might selectively abrogate fungal viability, especially when during infection multiple stresses are imposed by the host immune system. 相似文献

以山西省吉县蔡家川流域为对象,研究了植被覆盖类型对流域水文的影响.结果表明:不同植被覆盖的流域年径流系数分别为:林地流域1.6%~2.3%,以农、牧为主的流域3.1%~3.9%;各流域基流系数差异显著,人工林流域为零,次生林为主的流域1.0%~1.5%,以农、牧为主的流域2.5%~2.8%;在雨季人工林流域的径流总量是次生林流域的3.37倍、农地流域的1.9倍,而农地流域的基流量是次生林流域的2.2倍;短历时高强度降雨条件下,人工林流域、次生林流域地表径流量分别是农地流域的10.8倍和2.2倍;在历时较长的暴雨条件下,人工林流域单位面积上的洪峰流量是农地流域的3.4倍,次生林流域的6.9倍;在长历时、大雨量条件下,农地流域的径流量是次生林流域的1.8倍.水平梯田的水源涵养功能与次生林植被相当,次生林植被的水源涵养功能远好于人工植被,在水资源短缺的黄土高原应提倡植被的自然恢复. 相似文献

Gene and domain duplication in the chordate Otx gene family: insights from amphioxus Otx 总被引:5,自引:1,他引:5
We report the genomic organization and deduced protein sequence of a
cephalochordate member of the Otx homeobox gene family (AmphiOtx) and show
its probable single-copy state in the genome. We also present molecular
phylogenetic analysis indicating that there was single ancestral Otx gene
in the first chordates which was duplicated in the vertebrate lineage after
it had split from the lineage leading to the cephalochordates. Duplication
of a C-terminal protein domain has occurred specifically in the vertebrate
lineage, strengthening the case for a single Otx gene in an ancestral
chordate whose gene structure has been retained in an extant
cephalochordate. Comparative analysis of protein sequences and published
gene expression patterns suggest that the ancestral chordate Otx gene had
roles in patterning the anterior mesendoderm and central nervous system.
These roles were elaborated following Otx gene duplication in vertebrates,
accompanied by regulatory and structural divergence, particularly of Otx1
descendant genes.

The reactivity of human cord blood lymphocytes was assessed against a panel of monoclonal antibodies (MoAb). The mean proportion of OKT3+ cells (pan-T) was significantly lower in cord blood (52 +/- 13.8%; mean +/- SD) compared with that of adult blood (75 +/- 8.9%) and paralleled well with the E-rosette-forming capacity (50 +/- 16.3%). Both the proportions of OKT4+ cells (helper/inducer phenotype) and of OKT8+ cells (suppressor/cytotoxic phenotype) were significantly reduced in cord blood (43 +/- 11.8% vs 50.3 +/- 7.4% and 20 +/- 10.3% vs 25.6 +/- 6.0%, respectively), while the overall OKT4/OKT8 ratio was increased compared with adult blood (2.87 +/- 1.83 vs 2.04 +/- 0.61). Unlike adult blood, in 30 of the 35 samples of cord blood an overlap was observed between the total proportion of OKT4+ and OKT8+ cells (65 +/- 15.2%) and that of OKT3+ cells (52 +/- 14.3%). Although small numbers of cells coexpressing both antigens were occasionally found, double-staining analysis showed that the overlap in cord blood was mostly due to an expanded proportion of OKT3 (Leu-4)-/OKT8 (Leu-2)+ cells. Relevant proportions of OKT6+ (common thymocyte antigen) and OKT10+ (thymocytes, activated T cells, precursor cells) cells were found in cord blood as opposed to adult blood (10.8 +/- 8.6% vs 0.6 +/- 0.6% and 67 +/- 18.0% vs 8 +/- 2.1%, respectively), while terminal deoxynucleotidyl transferase-positive cells were observed only in two samples of cord blood. A small proportion of T cells (E-rosette+) reacted with the MoAb OKIa1 (HLA-DR). Finally, the proportion of cord blood cells recognized by the MoAb Leu-7 (HNK-1 clone) was almost negligible compared with adult blood (2.8 +/- 2.4% vs 15 +/- 7.5%). These data confirm the immaturity and heterogeneity of cord blood lymphocytes and demonstrate the presence at birth of circulating lymphocytes which express a surface phenotype reminiscent of that found in the late stages of intrathymic differentiation and in some human T-cell leukemias. Human cord blood may thus represent a suitable model for the study of the differentiation pathway of normal and pathological T-cells in humans. 相似文献

The selective migration of na?ve, effector, memory and regulatory T-cells is a multiple step process regulated by a specific arrangement of cytokines, chemokines and adhesion receptors that guide these cells to specific locations. Recent research has outlined two major pathways of lymphocyte trafficking under homeostatic and inflammatory conditions, one concerning tropism to cutaneous tissue and a second one related to mucosal-associated sites. Ischemic pain--examples include the chest pain of a heart attack and the leg pain of a 30 s sprint--occurs when muscle gets too little oxygen for its metabolic need. Lactic acid cannot act alone to trigger ischemic pain because the pH change is so small. Here, we show that another compound released from ischemic muscle, adenosine tri-phosphate (ATP), works together with acid by increasing the pH sensitivity of acid-sensing ion channel number 3 (ASIC3), the molecule used by sensory neurons to detect lactic acidosis. Our data argue that ATP acts by binding to P2X receptors that form a molecular complex with ASICs; the receptor on sensory neurons appears to be P2X5, an electrically quiet ion channel. Coincident detection of acid and ATP should confer sensory selectivity for ischemia over other conditions of acidosis. 相似文献
