Pollen of 16 species of Parkia (ca. 30 spp.) was examined in the light, scanning and transmission electron microscopes (LM, SEM and TEM). Pollen is shed as large (61-236 μm), globose, 16–32-grained polyads that remain united following acetolysis. Thin sections reveal that polyad cohesion is maintained through fusion and/or adhesion and/or appression of adjacent ektexines in lateral and/or proximal and/or distal regions. In lateral and proximal regions the ektexine is often embedded in a dense granular endexinous matrix. Individual grains of the polyad are clearly outlined in most species but are completely obscured in several New World species. Sculpturing varies both in pattern and type of element. Nonuniformly sculptured polyads, in which the sculpturing of individual grains differs between the peripheral and central portions of the distal face, occur only among Old World species. Uniformly sculptured polyads occur among both New and Old World taxa. Two main sculpturing types are present among Parkia species: verrucate and tectate-perforate, the latter particularly well developed in the reticulate polyads of the neotropical P. ulei and P. multijuga p.p. Ultrastructurally, all Parkia polyads exhibit ektexine and endexine continuous around the individual grain. Distal ektexine is prominent and clearly stratified, usually exhibiting a thick, perforate tectum, columellate interstitium, and a thin, locally discontinuous foot layer. Lateral and proximal ektexine is significantly thinner and less structured. Individual grains within the polyad possess three to four peripheral-distal apertures usually at the points of contact between adjacent grains. Proximal and central grain apertures occur in most, but not all, species. Polyad characters suggest a particularly close relationship among the New World species P. decussata, P. discolor, P. gigantocarpa, P. igneiflora, and P. nitida. Parkia ulei, though sculpturally closest to P. multijuga p.p., is ultrastructurally most similar to the African P. biglobosa. The large tectate-perforate-fossulate polyads of P. pendula and P. platyparkia are unique in the New World.  相似文献

Coxsackievirus B3 (CVB3) is a common human pathogen that has been associated with serious diseases including myocarditis and pancreatitis. To better understand the effect of cytotoxic T-lymphocyte (CTL) responses in controlling CVB3 infection, we have inserted well-characterized CTL epitopes into the CVB3 genome. Constructs were made by placing the epitope of interest upstream of the open reading frame encoding the CVB3 polyprotein, separated by a poly-glycine linker and an artificial 3Cpro/3CDpro cleavage site. This strategy results in the foreign protein being translated at the amino- terminus of the viral polyprotein, from which it is cleaved prior to viral assembly. In this study, we cloned major histocompatibility complex class I-restricted CTL epitopes from lymphocytic choriomeningitis virus (LCMV) into recombinant CVB3 (rCVB3). In vitro, rCVB3 growth kinetics showed a 1- to 2-h lag period before exponential growth was initiated, and peak titers were approximately 1 log unit lower than for wild-type virus. rCVB3 replicated to high titers in vivo and caused severe pancreatitis but minimal myocarditis. Despite the high virus titers, rCVB3 infection of naive mice failed to induce a strong CD8+ T-cell response to the encoded epitope; this has implications for the proposed role of "cross-priming" during virus infection and for the utility of recombinant picornaviruses as vaccine vectors. In contrast, rCVB3 infection of LCMV-immune mice resulted in direct ex vivo cytotoxic activity against target cells coated with the epitope peptide, demonstrating that the rCVB3-encoded LCMV-specific epitope was expressed and presented in vivo. The preexisting CD8+ memory T cells could limit rCVB replication; compared to naive mice, infection of LCMV-immune mice with rCVB3 resulted in approximately 50-fold-lower virus titers in the heart and approximately 6-fold-lower virus titers in the pancreas. Although the inserted CTL epitope was retained by rCVB3 through several passages in tissue culture, it was lost in an organ-specific manner in vivo; a substantial proportion of viruses from the pancreas retained the insert, compared to only 0 to 1.8% of myocardial viruses. Together, these results show that expression of heterologous viral proteins by recombinant CVB3 provides a useful model for determining the mechanisms underlying the immune response to this viral pathogen.  相似文献

The family Picornaviridae contains some notable members, including rhinovirus, which infects humans more frequently than any other virus; poliovirus, which has paralysed or killed millions over the years; and foot-and-mouth-disease virus, which led to the creation of dedicated institutes throughout the world. Despite their profound impact on human and animal health, the factors that regulate pathogenesis and tissue tropism are poorly understood. In this article, we review the clinical and economic challenges that these agents pose, summarize current knowledge of host-pathogen interactions and highlight a few of the many outstanding questions that remain to be answered.  相似文献

We propose a new Poisson method to estimate the variance for prevalence estimates obtained by the counting method described by Gail et al. (1999, Biometrics 55, 1137-1144) and to construct a confidence interval for the prevalence. We evaluate both the Poisson procedure and the procedure based on the bootstrap proposed by Gail et al. in simulated samples generated by resampling real data. These studies show that both variance estimators usually perform well and yield coverages of confidence intervals at nominal levels. When the number of disease survivors is very small, however, confidence intervals based on the Poisson method have supranominal coverage, whereas those based on the procedure of Gail et al. tend to have below-nominal coverage. For these reasons, we recommend the Poisson method, which also reduces the computational burden considerably.  相似文献

The anticonflict properties of the anxiolytic diazepam and of the dipeptide Litoralon gamma-L-glutamyl-taurine and two of its analogues SZJ 3388, gamma-aminobutyryl-ethanolamine phosphate, and SZJ 3361, D-1-aminoisobutyrylethanolamine phosphate have been investigated in a "time-to-emerge" conflict paradigm in non-deprived rats. Diazepam, Litoralon and compound SZJ 3388 significantly decreased the "time-to-emerge" latency (TTE latency) in a dose-dependent manner versus saline- or vehicle-treated controls in doses between 0.05 and 0.50 mg/kg intraperitoneally. The analogue SZJ 3361 was inactive as regards the TTE latency, while the anti-histaminic promethazine lengthened the TTE latency in a dose-dependent manner. The number of irresolute responses was significantly decreased following administration of diazepam, SZJ 3388 and Litoralon and was positively correlated with the TEE latency-decreasing activity of these compounds. The data are discussed in terms of the benzodiazepine-like anxiolytic or anti-conflict properties of these dipeptides.  相似文献

To evaluate the receiver operating characteristics (ROC) to determine the cutoffs of waist circumference as a potential population directed screening tool for hypercholesterolaemia (≥6.5 mmol/L), low high density lipoprotein cholesterol (<0.9 mmol/L), and hypertension (treated and/or systolic ≥160 and/or diastolic blood pressure ≥95 mmHg), in 2183 men and 2698 women aged 20 to 59 years selected at random from Dutch civil registries. Main outcome measures: Height, weight, body mass index (BMI), waist circumference, total plasma cholesterol and high density lipoprotein cholesterol concentrations, and blood pressure. Results: ROC curves showed that sensitivity equalled specificity at waist circumferences between 93–95 cm in men and 81–84 cm in women for identifying individual risk factors, and 92 cm in men and 81 cm in women for identifying those with at least one risk factor. Sensitivity and specificity were equal at levels between 61% to 69% for identifying individual risk factors, with positive predictions (56.8% in men and 37.8% in women) within 2% of those using previously defined ‘Action Level 1’ of waist circumference 94 cm in men and 80 cm in women (58.8% in men and 37.4% in women). Risk prediction by anthropometric methods was relatively low: ROC areas for identifying each risk factor by waist varied from 55% to 60%, and reached about 65% for identifying at least one risk factor. Height accounted for less than 03% of variance in waist circumference. Using BMI at 25 kg/m2 gave similar prediction to waist, but its combination with waist did not improve predictive values. Conclusions: Measurement of waist circumference ‘Action Level 1’ at 94 cm (37 inches) in men and 80 cm (32 inches) in women could be adopted as a simpler valid alternative to BMI for health promotion, to alert those at risk of cardiovascular disease, and as a guide to risk avoidance by self-weight management  相似文献

We present x-ray diffraction results of dipalmitoylphosphatidylcholine (DPPC) multilayers in three structural phases. Using pure DPPC, precision temperature control, and high angular resolution methods, we have discovered splitting of the first diffraction order due to multilayering in the Pβ phase. This splitting permits us to calculate the amplitude of ripples in this phase. The amplitude is large enough to suggest a structural mechanism for rippling.  相似文献

Fifteen Eurasian and Australian species of Viscum L. were examined by light, scanning, and transmission electron microscopy. Pollen characters divide the species into two groups, each correlated with differences in habit and inflorescence structure: Group I (12 spp.) characterized by psilate or slightly sculptured exines and a non-uniform ektexine pattern and Group II (3 spp.) possessing highly sculptured (echinate, rodlet) surfaces and uniform ektexine patterns. Within each of the groups, pollen characters divide the species into several subgroups. Among Group I, species V. nepalense, V. heyneanum and V. ovalifolium are particularly close. The Group I species, V. trilobatum, is placed in its own subgroup primarily because of its uniform ektexine pattern—a unique feature among Asian and Australian Viscum. Of the three Group II species, V. album and V. alniformosanae are palynologically almost indistinguishable. Pollen of the Group II V. cruciatum, though exomorphologically similar to V. album, is closest ultrastructurally to the Indian V. trilobatum. Overall, the most common and probably basic pollen characters among the Eurasian and Australian species include: subprolate, rounded convex almost spherical shapes; tricolporate apertures, and non-uniform sculpturing and ektexine patterns. Oblate-spheroidal or prolate-spheroidal shapes, prominent sculpturing, and a uniform ektexine pattern are derived characters largely restricted to the Eurasian and Asian Group II species (V. album, V. alniformosanae, V. cruciatum).  相似文献

Aims

The readily available global rock phosphate (P) reserves may be depleted within the next 50–130 years warranting careful use of this finite resource. We develop a model that allows us to assess a range of P fertiliser and soil management strategies for Barley in order to find which one maximises plant P uptake under certain climate conditions.

Methods

Our model describes the development of the P and water profiles within the soil. Current cultivation techniques such as ploughing and reduced till gradient are simulated along with fertiliser options to feed the top soil or the soil right below the seed.

Results

Our model was able to fit data from two barley field trials, achieving a good fit at early growth stages but a poor fit at late growth stages, where the model underestimated plant P uptake. A well-mixed soil (inverted and 25 cm ploughing) is important for optimal plant P uptake and provides the best environment for the root system.

Conclusions

The model is sensitive to the initial state of P and its distribution within the soil profile; experimental parameters which are sparsely measured. The combination of modelling and experimental data provides useful agricultural predictions for site specific locations.
