Identifying plant traits that promote invasiveness has been a major goal in invasion ecology. Germination plays a central role in the life cycle of plants and therefore could be a key trait in determining species invasiveness. In this study, seed germination of two confamilial, co‐occurring species that share ecological characteristics, the exotic invasive Gleditsia triacanthos L and the native Acacia aroma Gillies ex. Hook. & Arn., was compared. Seeds were obtained from individuals of three localities in the Chaco Serrano region of Córdoba, Argentina. Percent of seed germination and mean germination time were recorded in chemically and mechanically scarified seeds, and the former variable was also recorded in seeds subjected to: passage through the digestive tract of dispersers, fire simulations, fire simulation plus mechanical scarification, seed longevity, and dormancy break over time. In general, both species showed similar germination percentage. However, non‐scarified seeds of the exotic species lost physical dormancy when subjected to experiments of dormancy break over time, whereas, the native species had shorter mean germination time. The greater percentage of seed germination over time of the exotic species than of the native one might be triggering the spread of the former, whereas the shorter mean germination time might be hindering its expansion to more arid regions. The study of different mechanisms for achieving seed germination, particularly in hard seed species, could provide important information on the expansion of invasive species as well as useful knowledge for their management.  相似文献

European rabbits (Oryctolagus cuniculus), a keystone species in the Iberian Mediterranean ecosystem, are the staple prey of the Iberian lynx (Lynx pardinus) and the Spanish imperial eagle (Aquila adalberti). These predators require medium to high rabbit densities and a low degree of human disturbance. We compared rabbit abundances in areas of central-southern Spain under three levels of protection and management: protected areas, intensively managed (nonprotected) hunting estates, and other nonprotected areas. We used pellet abundance indices to estimate rabbit density in 118 surveys conducted during the summers of 2002 and 2003. We observed greater rabbit abundance in intensively managed hunting estates compared to protected areas and other nonprotected areas, perhaps because policy makers did not consider rabbit numbers when selecting priority areas. Alternatively, differences in game management practices (e.g., predator control or habitat management) may explain the higher rabbit densities observed in managed hunting estates. Our results suggest that the best feeding conditions for the Iberian lynx and the Spanish imperial eagle occur in intensively managed hunting areas, where such predators are frequently persecuted. The conservation of these endangered predators may require efforts to increase rabbit densities in protected areas.  相似文献

TGF-beta superfamily co-receptors are emerging as targets for cancer therapy, acting both directly on cells and indirectly on the tumour neovasculature. Endoglin (CD105), an accessory component of the TGF-beta receptor complex, is expressed in certain melanoma cell lines and the endothelial cells of tumour neovessels. Targeting endoglin with immunotoxins is an attractive approach for actively suppressing the blood supply to tumours. Here, we report evidence indicating that endoglin is expressed in mouse melanoma B16MEL4A5 and mouse fibroblast L929 cell lines. We prepared an immunotoxin to target endoglin by coupling the rat anti-mouse MJ7/18 (IgG2a) monoclonal antibody (mAb) to the non-toxic type 2 ribosome-inactivating protein nigrin b (Ngb) with N-succinimidyl 3-(2-pyridyldithio)-propionate (SPDP) as a linker with a molar nigrin b at a MJ7/18 stoichiometry of 2:1. The MJ7-Ngb immunotoxin generated killed both cell lines, with IC50 values of 4.2 × 10?9 M for B16MEL4A5 and 7.7 × 10?11 M for L929 cells. For in vivo assays of the immunotoxin, B16MEL4A5 cells were injected subcutaneously into the right flanks of 6-week-old C57BL/6 J mice. When the animals developed palpable solid tumours, they were subjected to treatment with the immunotoxin. While treatment with either MJ7/18 mAb or Ngb did not affect tumour development, treatment with the immunotoxin completely and steadily blocked tumour growth up to 7 days, after which some tumours re-grew. Thus, vascular-targeting therapy with this anti-vascular immunotoxin could promote the destruction of newly created tumour vessels at early stages of B16MEL4A5 tumour development and readily accessible CD105+ B16MEL4A5 melanoma cells.  相似文献

Habitat sinks can attract dispersing animals if high mortality or breeding failure are difficult to detect (e.g., when due to human hunting or pollution). Using a simple deterministic model, we explore the dynamics of such source-sink systems considering three scenarios: an avoided sink, no habitat preference, and an attractive sink. In the second two scenarios, there is a threshold proportion of sink habitat above which the whole population decreases to extinction, but this extinction threshold varies with habitat preference and the relative qualities of the two habitat types. Hence, it would be necessary to know the habitat preferences of any species in a source-sink system to interpret data on population increases and declines. In the attractive sink scenario, small changes in the proportion of sink habitat may have disproportionate effects on the population persistence. Also, small changes in growth rates at the source and the sink severely affect the threshold and the time of extinction. For some combinations of demographic parameters and proportion of habitat sink, the decline affects the source first; thus, during some time, it will be hidden to population monitoring at the sink, where numbers can even increase. The extinction threshold is also very sensitive to the initial population sizes relative to carrying capacity. Attractive sinks represent a novel aspect of source-sink dynamics with important conservation and management implications.  相似文献

Introduction

The human monoclonal antibody adalimumab is known to induce an anti-globulin response in some adalimumab-treated patients. Antibodies against adalimumab (AAA) are associated with non-response to treatment. Immunoglobulins, such as adalimumab, carry allotypes which represent slight differences in the amino acid sequences of the constant chains of an IgG molecule. Immunoglobulins with particular IgG (Gm) allotypes are racially distributed and could be immunogenic for individuals who do not express these allotypes. Therefore, we investigated whether a mismatch in IgG allotypes between adalimumab and IgG in adalimumab-treated patients is associated with the development of AAA.

Methods

This cohort study consisted of 250 adalimumab-treated rheumatoid arthritis (RA) patients. IgG allotypes were determined for adalimumab and for all patients. Anti-idiotype antibodies against adalimumab were measured with a regular radio immunoassay (RIA), and a newly developed bridging enzyme linked immunosorbent assay (ELISA) was used to measure anti-allotype antibodies against adalimumab. The association between AAA and the G1m3 and the G1m17 allotypes was determined. For differences between groups we used the independent or paired samples t-test, Mann-Whitney test or Chi square/Fisher's exact test as appropriate. To investigate the influence of confounders on the presence or absence of AAA a multiple logistic regression-analysis was used.

Results

Adalimumab carries the G1m17 allotype. No anti-allotype antibodies against adalimumab were detected. Thirty-nine out of 249 patients had anti-idiotype antibodies against adalimumab (16%). IgG allotypes of RA patients were associated with the frequency of AAA: patients homozygous for G1m17 had the highest frequency of AAA (41%), patients homozygous for G1m3 the lowest frequency (10%), and heterozygous patients' AAA frequency was 14% (P = 0.0001).

Conclusions

An allotype mismatch between adalimumab and IgG in adalimumab-treated patients did not lead to a higher frequency of AAA. On the contrary, patients who carried the same IgG allotype as present on the adalimumab IgG molecule, had the highest frequency of anti-adalimumab antibodies compared to patients whose IgG allotype differed from adalimumab. This suggests that the allotype of adalimumab may not be highly immunogenic. Furthermore, patients carrying the G1m17-allotype might be more prone to antibody responses.  相似文献

Mammalian carnivore communities affect entire ecosystem functioning and structure. However, their large spatial requirements, preferred habitats, low densities, and elusive behavior deem them difficult to study. In recent years, noninvasive techniques have become much more common as they can be used to monitor multiple carnivore species across large areas at a relatively modest cost. Hair snares have the potential to fulfill such requirements, but have rarely been tested in Europe. Our objective was to quantitatively assess the effectiveness of hair snares for surveying mesocarnivores in the Iberian Peninsula (Southwestern Europe), by comparison with camera-trapping. We used an occupancy modeling framework to assess method-specific detectability and occupancy estimates and hypothesized that detection probabilities would be influenced by season, sampling method, and habitat-related variables. A total of 163 hair samples were collected, of which 136 potentially belonged to mesocarnivores. Genetic identification success varied with diagnostic method: 25.2 % using mitochondrial CR, and 9.9 % using the IRBP nuclear gene. Naïve occupancy estimates were, in average, 5.3?±?1.2 times higher with camera-trapping than with hair-snaring, and method-specific detection probabilities revealed that camera traps were, in average, 6.7?±?1.1 times more effective in detecting target species. Overall, few site-specific covariates revealed significant effects on mesocarnivore detectability. Camera traps were a more efficient method for detecting mesocarnivores and estimating their occurrence when compared to hair snares. To improve hair snares' low detection probabilities, we suggest increasing the number of sampling occasions and the frequency at which hair snares are checked. With some refinements to increase detection rates and the success of genetic identification, hair-snaring methods may be valuable for providing deeper insights into population parameters, attained through adequate analysis of genetic information, that is not possible with camera traps.  相似文献

Background  

As there is limited information about the clinical signs of BSE and scrapie in goats, studies were conducted to describe the clinical progression of scrapie and BSE in goats and to evaluate a short clinical protocol for its use in detecting scrapie-affected goats in two herds with previously confirmed scrapie cases. Clinical assessments were carried out in five goats intracerebrally infected with the BSE agent as well as five reported scrapie suspects and 346 goats subject to cull from the two herds, 24 of which were retained for further monitoring. The brain and selected lymphoid tissue were examined by postmortem tests for disease confirmation.  相似文献

Rabbit restocking is widely used in Spain for conservation and/or hunting purposes; however, the success of rabbit restocking is generally low. Thus, many studies have assessed ways to reduce this problem, one of which is the use of a “soft-release” procedure whereby rabbits are acclimated to their release site for a variable time period prior to release. This study assessed the short-term effects of two soft-release confinement periods on the survival of rabbits during an experimental restocking program carried out in southwest Spain. The post-release survival rate of rabbits confined at the release site for six nights was significantly higher than that of rabbits confined for three nights. The longer acclimation period after rabbit translocation minimized mortality while rabbits adapted to their new environment.  相似文献
