Background

Salsolinol (SALSO), a product from the reaction of dopamine (DA) with acetaldehyde, is found increased in dopaminergic neurons of Parkinson's disease (PD) patients. The administration of SALSO in rats causes myenteric neurodegeneration followed by the formation of deposits of the protein α-synuclein (aS), whose aggregation is intimately associated to PD.

Methods

NMR, isothermal titration calorimetry and MS were used to evaluate the interaction of SALSO with aS. The toxicity of SALSO and in vitro-produced aS-SALSO species was evaluated on mesencephalic primary neurons from mice.

Results

SALSO, under oxidative conditions, stabilizes the monomeric state besides a minor population of oligomers of aS, resulting in a strong inhibition of the fibrillation process. SALSO does not promote any chemical modification of the protein. Instead, the interaction of SALSO with aS seems to occur via hydrophobic effect, likely mediated by the NAC (non-amyloid component) domain of the protein. aS-SALSO species were found to be innocuous on primary neurons, while SALSO alone induces apoptosis via caspase-3 activation. Importantly, exogenous aS monomer was capable of protecting neurons against SALSO toxicity irrespective whether the protein was co-administered with SALSO or added until 2?h after SALSO, as evidenced by DAPI and cleaved-caspase 3 assays. Similar protective action of aS was found by pre-incubating neurons with aS before the administration of SALSO.

Conclusions

Interaction of SALSO with aS leads to the formation of fibril-incompetent and innocuous adducts. SALSO toxicity is attenuated by aS monomer.

Significance

aS could exhibit a protective role against the neurotoxic effects of SALSO in dopaminergic neuron.  相似文献

Crude methanolic extracts and fractions from the aerial parts of seven species of Hypericum (H. caprifoliatum Cham. and Schltdl., H. carinatum Griseb., H. connatum Lam., H. ternum A. St.-Hil., H. myrianthum Cham. and Schltdl., H. piriai Arechav. and H. polyanthemum Klotzsch ex Reichardt) growing in southern Brazil were analyzed for their in vitro antifungal activity against a panel of standardized and clinical opportunistic pathogenic yeasts and filamentous fungi, including dermatophytes, by the agar dilution method. Chloroform and hexane extracts of H. ternum showed the greatest activity among extracts tested.  相似文献

The fraction of cellular volume occupied by the cytoplasm has been assessed by point-counting volumetry in 0.5 micrometer araldite sections. Measurement of the transection radius of the nuclei of acinar cells allowed the assessment of the mean nuclear volume according to the method of Bach. With these data, the cytoplasm volume was calculated in cubic micrometers. Ultrastructural morphometric data were obtained by means of superimposing on the electron photomicrographies (x 21,000 or x 28,500) a test system of 84 evenly spaced segments according to Weibel et al. The analysis of the values obtained for the pancreas and parotid gland allowed for the supposition that the process of maturation in the rat, either for the pancreas or the parotid gland, occurs simultaneously with the mitotic process, this last being held as the predominating one in the first 3--4 postnatal weeks. The cytodifferentiation process becomes more marked in the last phases studied and on the 40th day it already shows parameters held as similar to those of the adult animal.  相似文献

Cytokine Receptor-Like Factor 2 (CRLF2) overexpression occurs in 5-15% of B-cell precursor acute lymphoblastic leukaemia (B-ALL). In ∼50% of these cases, the mechanisms underlying this dysregulation are unknown. IKAROS Family Zinc Finger 1 (IKZF1) is a possible candidate to play a role in this dysregulation since it binds to the CRLF2 promoter region and suppresses its expression. We hypothesised that IKZF1 loss of function, caused by deletions or its short isoforms expression, could be associated with CRLF2 overexpression in B-ALL. A total of 131 paediatric and adult patients and 7 B-ALL cell lines were analysed to investigate the presence of IKZF1 deletions and its splicing isoforms expression levels, the presence of CRLF2 rearrangements or mutations, CRLF2 expression and JAK2 mutations. Overall survival analyses were performed according to the CRLF2 and IKZF1 subgroups. Our analyses showed that 25.2% of patients exhibited CRLF2 overexpression (CRLF2-high). CRLF2-high was associated with the presence of IKZF1 deletions (IKZF1del, p = 0.001), particularly with those resulting in dominant-negative isoforms (p = 0.006). Moreover, CRLF2 expression was higher in paediatric samples with high loads of the short isoform IK4 (p = 0.011). It was also associated with the occurrence of the IKZF1 plus subgroup (p = 0.004). Furthermore, patients with CRLF2-high/IKZF1del had a poorer prognosis in the RELLA05 protocol (p = 0.067, 36.1 months, 95%CI 0.0-85.9) and adult cohort (p = 0.094, 29.7 months, 95%CI 11.8–47.5). In this study, we show that IKZF1 status is associated with CRLF2-high and dismal outcomes in B-ALL patients regardless of age.  相似文献

Question: Do traits of liana regeneration differ among secondary forest types of varying land‐use history and primary forest? Location: Eighty kilometers north of Manaus, Brazil. Methods: We compared plant functional traits and growth rates of liana regeneration (<1.7‐m length) among two secondary forest types and primary forest. Secondary forest types were: Vismia (on land formerly clear‐cut, used for pasture and intensively burned) and Cecropia (no pasture usage or intensive fires after clear‐cut). Results: A principal components analysis indicated that most of the primary forest species exhibited a similar habit and were characterized by short shoots and small, round leaves with low specific leaf area, whereas secondary forest species had a broad range of trait values. At the plot level, primary and secondary forest communities were separated mainly by plant length and leaf size. Plant size varied more within secondary than within primary forest plots. The two secondary forest types could not be separated based on the traits of liana regeneration. Relative growth rate (RGR) did not correlate significantly with any measured plant trait, except for a negative relation to initial length. RGR increased with decreasing canopy cover and was highest in Vismia forest plots. Conclusion: Plant functional traits of liana regeneration were more similar in the primary forest and differed substantially from secondary forests, yet canopy cover only partly explained the observed differences.  相似文献

Bovine papillomavirus (BPV) DNA sequences were detected in different tissues, in addition to epithelium. Cytogenetic abnormalities were observed in blood lymphocytes. The presence of more than one virus in a single tissue is a difficult aspect to evaluate, especially when the DNA sequences are detected in tissues that are not specifically targeted by the virus. BPV and bovine leukemia virus (BLV) are clastogenic, causing chromosome aberrations in peripheral blood lymphocytes. In the present study, we investigated the simultaneous presence of DNA sequences of both viruses and the possibility of vertical transmission and compared the types of chromosome aberrations related to viral action. BPV 1, 2, and 4 DNA sequences were found in three females of the herd and in their offspring. BLV DNA sequences were not detected in their progeny. A newborn calf that was negative for BLV infection showed specific chromosome rearrangements possibly related to the effect of infection with BPV.  相似文献

The kinetic α-secondary deuterium isotope effect, kHkD, for the pH-independent hydrolysis of nicotinamide riboside, yielding nicotinamide and ribose, in water at 25 ° is 1.14, establishing that this reaction proceeds with unimolecular substrate decomposition to yield a carboxonium ion, or related species, in the rate-determining step. Surprisingly, the corresponding isotope effect for the base-catalyzed decomposition of the same substrate is 1.12, a value indicating considerable sp2 character at the Cl′ position in the transition state for this reaction. A similar result, kHkD = 1.15, was obtained for base-catalyzed hydrolysis of NAD+. The kinetic alpha deuterium isotope effect for the pig brain NAD glycohydrolasecatalyzed hydrolysis of nicotinamide riboside is 1.08. This value suggests that CN bond cleavage to form an intermediate carboxonium ion, or structurally related species, is at least partially rate-determining. In contrast, the corresponding value for the hydrolysis of this substrate catalyzed by Escherichia coli nicotinamide ribonucleotide glycohydrolase is very near unity, a result consistent with several interpretations including a rate-determining enzyme isomerization reaction.  相似文献

ABSTRACT: BACKGROUND: The parathyroid hormone (PTH)-family consists of a group of structurally related factors that regulate calcium and bone homeostasis and are also involved in development of organs such as the heart, mammary gland and immune system. They interact with specific members of family 2 B1 G-protein coupled receptors (GPCRs), which have been characterised in teleosts and mammals. Two PTH/PTHrP receptors, PTH1R and PTH2R exist in mammals and in teleost fish a further receptor PTH3R has also been identified. Recently in chicken, PTHfamily members involved in calcium transport were characterized and specific PTHRs are suggested to exist although they have not yet been isolated or functionally characterized. The aim of this study is to further explore the evolution and function of the vertebrate PTH/PTHrP system through the isolation, phylogenetic analysis and functional characterization of the chicken receptors. RESULTS: Two PTHRs were isolated in chicken and sequence comparison and phylogenetic analysis indicate that the chicken receptors correspond to PTH1R and PTH3R, which emerged prior to the teleost/tetrapod divergence since they are present in cartilaginous fish. The vertebrate PTH2R receptor and its ligand TIP39 have been lost from bird genomes. Chicken PTH1R and PTH3R have a divergent and widespread tissue expression and are also evident in very early embryonic stages of development. Receptor stimulation studies using HEK293 cells stably expressing the chicken PTH1R and PTH3R and monitoring cAMP production revealed they are activated by chicken 1-34 N-terminal PTH-family peptides in a dose dependent manner. PTH-L and PTHrP were the most effective peptides in activating PTH1R (EC50 = 7.7 nM and EC50 = 22.7 nM, respectively). In contrast, PTH-L (100 nM) produced a small cAMP accumulation on activation of PTH3R but PTHrP and PTH (EC50 = 2.5 nM and EC50 = 22.1 nM, respectively) readily activated the receptor. PTHrP also stimulated intracellular Ca2+ accumulation on activation of PTH1R but not PTH3R. CONCLUSION: Two PTHR homologues of the vertebrate PTH1R and PTH3R were isolated and functionally characterized in chicken. Their distinct pattern of expression during embryo development and in adult tissues, together with their ligand preference, suggests that they have acquired specific functions, which have contributed to their maintenance in the genome. PTH2R and its activating ligand, TIP39, are absent from bird genomes. Nonetheless identification of putative PTH2R and TIP39 in the genome of an ancient agnathan, lamprey, suggests the PTH/PTHrP ligand and receptor family was already present in an early basal paraphyletic group of vertebrates and during the vertebrate radiation diverged via gene/genome duplication and deletion events. Knowledge of the role PTH/PTHrP system in early vertebrates will help to establish evolution of function.  相似文献

Many species of grasshopper have an XX/XO sex chromosome system, including Tropidacris cristata grandis (23, XX/XO). The X chromosome behaves differently from the autosomes, but little is known about its origin and molecular composition. To better understand the genomic composition and evolutionary processes involved in the origin of the sex chromosomes, we undertook an analysis of its meiotic behavior, heterochromatin distribution and microdissection in T. c. grandis. Analysis of meiotic cells revealed a difference in the behavior of the X chromosome compared to the autosomes, with different patterns of condensation and cellular arrangement. Heterochromatic terminal blocks were predominant. The chromosome painting revealed a bright block in the centromeric/pericentromeric region of the X chromosome and slight markings in the other regions. In the autosomes, the X chromosome probe hybridized in the centromeric/pericentromeric region, and hybridization signals on terminal regions corresponding to the heterochromatic regions were also observed. The results showed that the X chromosome contains a significant amount of repetitive DNA. Based on the hybridization pattern, it is possible that the autosomes and sex chromosomes of T. c. grandis have a similar composition of repetitive DNAs, which could mean that the X chromosome has an autosomal origin.  相似文献
