Mitotic chromosomes with sister chromatids bearing differentially active ribosomal gene clusters were recovered from human lymphocytes exposed to 5-azacytidine. The hypothesis was that the differential activity was determined by the hypomethylation of one of the two sister chromatids. The verification was carried out by labeling the 5-azacytidine-substituted chromatid with BUdR, and then checking the location of active clusters by specific staining techniques. Data obtained confirmed that the chromatid bearing the active cluster was indeed the 5-azacytidine-substituted one.  相似文献

Feedback lighting (LDFB), which illuminates an animal cage in response to active wheel running, exposes only the photosensitive portion of the phase-response curve to light. In the hamster, the photoinducible zone of the circadian rhythm of photoperiodic photosensitivity occurs during the interval of active wheel running. Since LDFB exposes the photoinducible zone almost as much as constant light (LL), we predicted that LDFB would maintain gonadal function just as LL does. Surprisingly, 10 male hamsters exposed to 1-sec pulses of LDFB for 8 wk had regressed testes similar to those of hamsters in continuous darkness (DD) and significantly smaller than hamsters exposed to LL (P less than 0.01). Two of 5 male hamsters exposed to 2-min pulses of LDFB underwent complete testicular regression and two had partially regressed testes. All females exposed to LDFB or to DD ceased showing cyclic signs of ovulation within 20 days, whereas most hamsters exposed to LL continued to show signs of cyclic ovulation. Six of the 8 hamsters exposed to LL had ova in their oviducts at autopsy, and also had significantly larger uteri (P less than 0.01) than hamsters exposed to DD or LDFB. None of the latter two groups (n = 6 and 9, respectively) had oviductal ova at autopsy. These results demonstrate that considerable exposure of the photoinducible zone to light does not necessarily maintain gonadal function. Light delivered to the photoinducible zone by LDFB may disrupt the normal alignment (internal coincidence) of circadian rhythms, thereby causing gonadal regression. Gonadal induction can occur when the photoinducible zone is exposed to light; however, it may not be the light itself, but rather the action of the light to alter the phase relationships of several oscillators, that causes induction and maintenance of the gonads.  相似文献

The present paper analyzes literature data dealing with the use of aminopolycarboxylates for chemical washing of heavy metals contaminated soils. The aim is to rationalize and organize the up-to-date knowledge of process dependence on different parameters, in order to give a contribution to the development of washing protocols applicable for process optimization. Process efficiency is found to be dependent on i) parameters related to soil composition, ii) parameters related to contaminant characteristics and iii) operating parameters. The effect that each of them, according to the reviewed experimental studies, has on process efficiency, is highlighted, and the interdependence among them is presented as a key point to understand the overall removal mechanism and therefore to calibrate its application case-by-case.  相似文献

Emerging Evidence on the Effectiveness of Tropical Forest Conservation   总被引:1,自引:0,他引:1  
The PLOS ONE Collection “Measuring forest conservation effectiveness” brings together a series of studies that evaluate the effectiveness of tropical forest conservation policies and programs with the goal of measuring conservation success and associated co-benefits. This overview piece describes the geographic and methodological scope of these studies, as well as the policy instruments covered in the Collection as of June 2016. Focusing on forest cover change, we systematically compare the conservation effects estimated by the studies and discuss them in the light of previous findings in the literature. Nine studies estimated that annual conservation impacts on forest cover were below one percent, with two exceptions in Mexico and Indonesia. Differences in effect sizes are not only driven by the choice of conservation measures. One key lesson from the studies is the need to move beyond the current scientific focus of estimating average effects of undifferentiated conservation programs. The specific elements of the program design and the implementation context are equally important factors for understanding the effectiveness of conservation programs. Particularly critical will be a better understanding of the causal mechanisms through which conservation programs have impacts. To achieve this understanding we need advances in both theory and methods.  相似文献

The subcellular distribution of the alpha 2-adrenergic receptor, pertussis-toxin substrates (Gi, the inhibitory G-protein) and adenylate cyclase was determined in human platelets. The alpha 2-adrenergic receptor and pertussis-toxin substrate activity codistribute with surface membranes identified by a novel fluorescent-lectin method. The platelet granule fractions did not contain detectable Gi. Only 2-4% of the total pertussis-toxin substrate activity appears in soluble fractions, and this amount was not increased upon addition of purified beta gamma units or after pretreatment of platelets with adrenaline. There is no evidence for compartmentation of the alpha 2-adrenergic receptor or Gi to account for the low-affinity component of agonist binding to the alpha 2-adrenergic receptor in human platelet membranes. Translocation of Gi from plasma membrane to platelet cytosol or granules does not appear to play any significant role in the mechanism of alpha 2-receptor-mediated platelet activation.  相似文献

Summary The non-histone chromatin proteins (NHCp) from pig liver and kidney have been partially fractionated in non-denaturing conditions by the use of histone H3 immobilized on agarose and the fractions obtained have been analysed by SDS-polyacrylamide gel electrophoresis and amino acid analysis. At least six different fractions have been obtained by successive increases of the ionic strength of the medium. Few NHCp have been evidentiated with subunit molecular weights 55,000 and less than 30,000, which display a remarkably high affinity for histone H3, and require 5 M urea to be displaced from the immobilized histone. The elution patterns of the NHCp from liver and kidney, although very similar, reveal some significant differences between the two tissues, which are undetectable by SDS-electrophoresis and which are most likely due to tissue specific proteins. This histone-affinity chromatography appears to be a promising approach for the analysis of specific histone-NHCp interactions, and as a first step for NHCp purification.  相似文献

The effects of pH on the binding of hematoporphyrin derivative (HpD) to monolayer and bilayer membranes have been studied. Absorption spectra of HpD bound to phosphatidylcholine (PC) liposomes indicate that there is greater binding of HpD to lipid films at acidic, tumoricidal pH conditions than at normal tissue pH. These results were found to correlate with surface pressure measurements of monolayer films formed under similar conditions. Surface potential measurements in conjunction with surface pressure measurements from monolayer films suggest that at low pH (i.e. less than or equal to 6.6) porphyrin intercalates within the lipid film to reach relatively high concentrations, while at higher pH (i.e. greater than or equal to 7.4) the porphyrin preferably adsorbs to the lipid film at the monolayer/water interface.  相似文献

We measured the effects of four weeks of dietary lithium treatment and of status epilepticus induced by administration of pilocarpine to lithium-treated rats on the concentrations of amino acids in four regions of rat brain: cerebral cortex, hippocampus, striatum, and substantia nigra. To ensure accurate quantitation of the amino acids, animals were sacrificed by focussed beam microwave irradiation and amino acids were measured using a fully validated triple-column ion-exchanged amino acid analyzer with post-column o-phthalaldehyde derivatization and fluorometric detection. The concentrations of four amino acids, threonine, methionine, lysine and tyrosine, were increased significantly in two to four brain regions by chronic lithium treatment. Their concentrations remained elevated, or were further increased, during status epilepticus. The concentrations of eight amino acids and ammonia were not altered by lithium treatment but increased in concentration during status epilepticus in some brain regions. Glycine, serine, arginine and citrulline were decreased by chronic lithium treatment. Status epilepticus increased the concentrations of these four amino acids above that found in the lithium-treated samples in some of the brain regions that were examined. Six amino acids and glutathione were generally unaltered by both treatments. These results are related to the effects of lithium treatment and are compared with changes reported by others following treatment with a variety of convulsive stimuli.  相似文献

The present article reviews our recent biochemical and microdialysis studies showing the evidence for an antagonistic CCK(B)/D(2) receptor interaction in the regulation of dopaminergic transmission in the nucleus accumbens and GABAergic transmission in the ipsilateral ventral pallidum. Since the nucleus accumbens plays a crucial role in regulating the output from the limbic system and consequently motivation, it may be speculated that a dysregulation of this receptor interaction may have consequences in a wide range of central nervous system disorders.  相似文献
