Long-term (1987–2015) monitoring data from Lake Lugano (Switzerland/Italy) were used to examine the effects of winter weather, solar radiation and mixing regime on the recovery of the trophic state of a lake undergoing nutrient management. Main hypotheses were that (H1) trends in trophic state were influenced not only by nutrient management, but also by winter weather and solar radiation, and (H2) the effects of management were more pronounced in the holomictic South basin than in the meromictic North basin of the lake. External loadings of phosphorus were strongly reduced during the study period, but key indicators of trophic state, including phosphorous concentration, primary production, chlorophyll a and deep-water oxygenation, showed inconsistent responses. Supporting H1, winter weather (parameterized using an index of the East Atlantic pattern) influenced temporal variation in phosphorus concentration and primary production in the North basin. Supporting H2, the effects of management on trophic state were clearer in the South basin, where most trophic-state indicators declined. Meromixis affected the restoration of the North basin lake by transmitting effects of climatic variation on trophic state. The added variability obscured the effects of restoration and caused sudden deteriorations in water quality, indicating that the restoration of meromictic lakes presents unique challenges. 相似文献

investigate the effect of immobilization and culture conditions on MnP production in cultures of Phanerochaete chrysosporium grown on polyurethane foam. Higher concentrations of foam and lower levels of spore inoculums resulted in the formation of
scattered mycelial pellets, increased autolysis of chlamydospore-like cells (a reservoir of MnP), and a higher activity of
MnP. Even though MnP was a secondary metabolite, the addition of 5 times more glucose and diammonium tartrate, as carbon and
nitrogen sources, resulted in a 4 fold increase in the dry cell mass. However, MnP activity decreased under these conditions

Histone acetylation plays a critical role in controlling chromatin structure, and reactive oxygen species (ROS) are involved in cell cycle progression. To study the relationship between histone acetylation and cell cycle progression in plants, sodium butyrate (NaB), a histone deacetylase (HDAC) inhibitor that can cause a significant increase in histone acetylation in both mammal and plant genomes, was applied to treat maize seedlings. The results showed that NaB had significant inhibition effects on different root zones at the tissue level and caused cell cycle arrest at preprophase in the root meristem zones. This effect was accompanied by a dramatic increase in the total level of acetylated lysine 9 on histone H3 (H3K9ac) and acetylated lysine 5 on histone H4 (H4K5ac). The exposure of maize roots in NaB led to a continuous rise of intracellular ROS concentration, accompanied by a higher electrolyte leakage ratio and malondialdehyde (MDA) relative value. The NaB-treated group displayed negative results in both TdT-mediated dUTP nick end labelling (TUNEL) and γ-H2AX immunostaining assays. The expression of topoisomerase genes was reduced after treatment with NaB. These results suggested that NaB increased the levels of H3K9ac and H4K5ac and could cause preprophase arrest accompanied with ROS formation leading to the inhibition of DNA topoisomerase. 相似文献

QUESTION
A 6-second spirometry test is easier than full exhalations. We compared the reliability of the ratio of the Forced expiratory volume in 1 second/Forced expiratory volume in 6 seconds (FEV1/FEV6) to the ratio of the FEV1/Forced vital capacity (FEV1/FVC) for the detection of airway obstruction.METHODS
The PLATINO population-based survey in individuals aged 40 years and over designed to estimate the prevalence of post-Bronchodilator airway obstruction repeated for the same study participants after 5–9 years in three Latin-American cities.RESULTS
Using the FEV1/FVC<Lower limit of normal (LLN) index, COPD prevalence apparently changed from 9.8 to 13.2% in Montevideo, from 9.7 to 6.0% in São Paulo and from 8.5 to 6.6% in Santiago, despite only slight declines in smoking prevalence (from 30.8% to 24.3%). These changes were associated with differences in Forced expiratory time (FET) between the two surveys. In contrast, by using the FEV1/FEV6 to define airway obstruction, the changes in prevalence were smaller: 9.7 to 10.6% in Montevideo, 8.6 to 9.0% in São Paulo, and 7.5 to 7.9% in Santiago. Changes in the prevalence of COPD with criteria based on FEV1/FVC correlated strongly with changes in the FET of the tests (R2 0.92) unlike the prevalence based on a low FEV1/FEV6 (R2 = 0.40).CONCLUSION
The FEV1/FEV6 is a more reliable index than FEV1/FVC because FVC varies with the duration of the forced exhalation. Reporting FET and FEV1/FEV6<LLN helps to understand differences in prevalence of COPD obtained from FEV1/FVC-derived indices. 相似文献947.
The spatial heterogeneity of DTPA-extractable zinc in the cultivated soils of Shenyang suburbs in Liaoning Province of China was investigated, and its map was drawn by the methods of geostatistics combined with geographic information system. The data of soil DTPA-extractable zinc fitted normal distribution after logarithm transformation, and its semivariogram fitted a spherical model. The semivariogram indicated that the spatial dependence of soil DTPA-extractable zinc content was moderate, with the spatial dependence range of 1.69 km and the fractal dimension of 1.96. Stochastic factors contributed to 49.9% of the spatial variability, while structural factors contributed to 50.1% of it. The spatial heterogeneity of soil DTPA-extractable zinc shown by a kriged interpolation map was deeply influenced by stochastic factors such as city pollution, land use pattern and crop distributions. For example, the average content of Zn in vegetable garden soils was 2.5–4 times as much as in their originated soils, and was lower in paddy soils than in their originated soils. The areas with a higher content of soil DTPA-extractable zinc appeared in the near suburbs and the riverside along Hunhe River and the wastewater drainage of Xihe River, and the extremely high values in the near suburb of the city’s residential area were a striking feature, indicating the key role of city pollution in the spatial heterogeneity of soil DTPA-extractable zinc. When recorded in the form of a soil pollution map, the results of such a survey make it possible to identify the unusually polluted areas, and to provide more information for precise agriculture and environmental control. 相似文献

A DNA barcoding study was conducted to determine the optimal combination of loci needed for successful species‐level molecular identification in three extant cycad genera—Ceratozamia, Dioon, and Zamia—that occur in Mexico. Based on conclusions of a previous multigene study in representative species of all genera in the Cycadales, we tested the DNA barcoding performance of seven chloroplast coding (matK, rpoB, rpoC1, and rbcL) and non‐coding (atpF/H, psbK/I, and trnH‐psbA) regions, plus sequences of the nuclear internal transcribed spacer. We analysed data under the assumptions of the “character attributes organization system” (CAOS), a character‐based approach in which species are identified through the presence of ‘DNA diagnostics’. In Ceratozamia, four chloroplast regions and one nuclear region were needed to achieve > 70% unique species identification. In contrast, the two‐gene combination atpF/H + psbK/I and the four‐gene combination atpF/H + psbK/I + rpoC1 + ITS2 were needed to reach 79% and 75% unique species identification in Dioon and Zamia, respectively. The combinations atpF/H + psbK/I and atpF/H + psbK/I + rpoC1 + ITS2 include loci previously considered by the international DNA barcoding community. However, none of the three combinations of potential DNA barcoding loci found to be optimal with a character‐based approach in the Mexican cycads coincides with the ‘core barcode’ of chloroplast markers (matK + rbcL) recently proposed for universal use in the plant kingdom. 相似文献

In the black chafer Holotrichia kiotonensis Brenske (Coleoptera: Scarabaeidae), mating behavior was observed between 1940 and 2010 hours at < 0.1 lx in both the laboratory and the field. In the laboratory, an ether extract of female abdominal glands induced a series of pre-mating behaviors such as short-distance orientation and abdominal bending. When the extract was fractionated by silica gel column chromatography, the active fraction was eluted with 50% ether in hexane and 100% ether. Gas chromatography–mass spectrometry analysis revealed that both active fractions contained anthranilic acid (2-amino-benzoic acid) as a major compound. The amount of this compound was determined to be ca. 600 ng/female by high performance liquid chromatography analysis with a fluorescence detector. In the field, male chafers were observed to land on cotton balls impregnated with 10 mg of authentic anthranilic acid. When a white ball treated with anthranilic acid was placed 2–10 cm away from an untreated black ball, males were observed to land significantly more frequently on the latter. These results suggest that males could recognize white balls below 0.1 lx and landed on black balls. When a treated black ball was placed beside an untreated black ball, more males landed on the former. The difference was significant when the distance between the two lures was 5 or 10 cm, but not significant when it was 2 cm. These observations demonstrated that anthranilic acid was the sex-attractant pheromone for the black chafer H. kiotonensis and that the males located and landed on a pheromone source by using olfaction in conjunction with visual orientation. The importance of visual orientation in this nocturnal species is discussed in comparison with the congeneric diurnal species Holotrichia loochooana loochooana. 相似文献
