The nematomorph species Chordodes festae Camerano, 1897 and C. peraccae (Camerano, 1894) are redescribed by using scanning electron microscopy (SEM). C. festae has a cuticle with four different areolar types, the crown areoles being the most noticeable with long spiniform processes. The terminal end in the male specimen has two short lobe-like structures and a ventral groove. C. peraccae has three areolar types in which, as in C. festae, some areoles form groups surrounding the crown areoles. Intraspecific variations were found in body length and body colour in both species and, in C. peraccae, also in the cuticle. 相似文献

The objective of this study was to determine the viscosity of the residual unfrozen solution that cells are exposed to during freezing in the presence of glycerol and use this to interpret some key aspects of cryopreservation. The viscosity of the glycerol-water binary system exceeded 1000 cP at -40 degrees C, whilst the viscosity of the ternary system, glycerol-water-NaCl, exceeded 100,000 cP at -55 degrees C. The effect of these high viscosities on the diffusion of water at a constant temperature during freezing and during cooling at different linear rates has been estimated. At rates of cooling faster than 100 degrees C min(-1) the diffusion distance during freezing was calculated to be less than 15 microm. Validation of the diffusion calculations was confirmed by examination of the ultrastructure of the freeze concentrated matrix in samples prepared at a range of cooling rates. At a critical rate of cooling, water diffusion becomes limited by the high viscosity and two phenomena, of relevance to cryobiology, occur: (1) the composition of the freeze concentrated matrix around cells deviates from that of the equilibrium phase diagram; and (2) the osmotic loss of water from cells is restricted. These factors are of particular relevance to an understanding of the response of cells such as spermatozoa, red blood cells, and bacteria cooled rapidly with glycerol as cryoprotectant. 相似文献

We have investigated the direct effect of arachidonic acid on cholesterol transport in intact cells or isolated mitochondria from steroidogenic cells and the effect of cyclic-AMP on the specific release of this fatty acid inside the mitochondria. We show for the first time that cyclic-AMP can regulate the release of arachidonic acid in a specialized compartment of MA-10 Leydig cells, e.g. the mitochondria, and that the fatty acid induces cholesterol transport through a mechanism different from the classical pathway. Arachidonic acid and arachidonoyl-CoA can stimulate cholesterol transport in isolated mitochondria from nonstimulated cells. The effect of arachidonoyl-CoA is inhibited by the reduction in the expression or in the activity of a mitochondrial thioesterase that uses arachidonoyl-CoA as a substrate to release arachidonic acid. cAMP-induced arachidonic acid accumulation into the mitochondria is also reduced when the mitochondrial thioesterase activity or expression is blocked. This new feature in the regulation of cholesterol transport by arachidonic acid and the release of arachidonic acid in specialized compartment of the cells could offer novel means for understanding the regulation of steroid synthesis but also would be important in other situations such as neuropathological disorders or oncology disorders, where cholesterol transport plays an important role. 相似文献

Bumblebees are widely distributed across the world and have great economic and ecological importance as pollinators in the forest as well as in agriculture. The insect midgut consists of three cell types, which play various important roles in digestion, absorption, and hormone production. The present study characterized the anterior and posterior midgut regions of the bumblebee, Bombus morio. The digestive, regenerative and endocrine cells in the midgut showed regional differences in their number, nuclear size, as well as the size of the striated border. Ultrastructurally, the digestive cells contained many mitochondria and long microvilli; however, in the anterior midgut region, these cells showed dilated basal labyrinths with a few openings for the hemocoel, whereas the labyrinths of the basal posterior region remained inverse characteristics. Thus, the characterization of the midgut of B. morio supported an ecto-endoperitrophic circulation, contributing to a better understanding of the digestive process in this bee. 相似文献

Complexes of general formula [{CuX}2(YNC10H14O)] (X = Cl; Y = NHMe, NH2 or X = Br; Y = NH2) were synthesised from camphor hydrazone ligands (YNC10H14O) by reaction with the suitable copper(I) halide. Structural analysis by X-rays performed on a red crystal of [{CuCl}2(Me2NNC10H14O)] revealed that the complex is a one-dimensional copper polymer formed by two rather different copper units bridged by chloride. One of the copper units displays a tetrahedral geometry while the other is linear. Although the geometries and neighbourhoods of the two copper units are very different the oxidation state of the metal is the same, i.e. Cu(I) as corroborated by magnetic and electrochemical measurements.The ability of [{CuCl}2(Me2NNC10H14O)] to promote the activation of 4-pentyn-1-ol towards cyclization was studied under homogeneous or heterogeneous experimental conditions. Studies investigating the impacts of biodiversity loss on ecosystem processes have often reached different conclusions, probably
because insufficient attention has been paid to some aspects including (1) which biodiversity measure (e.g., species number,
species identity or trait) better explains ecosystem functioning, (2) the mechanisms underpinning biodiversity effects, and
(3) how can environmental context modulates biodiversity effects. Here, we investigated how species number (one to three species)
and traits of aquatic fungal decomposers (by replacement of a functional type from an unpolluted site by another from a metal-polluted
site) affect fungal production (biomass acumulation) and plant litter decomposition in the presence and absence of metal stress.
To examine the putative mechanisms that explain biodiversity effects, we determined the contribution of each fungal species
to the total biomass produced in multicultures by real-time PCR. In the absence of metal, positive diversity effects were
observed for fungal production and leaf decomposition as a result of species complementarity. Metal stress decreased diversity
effects on leaf decomposition in assemblages containing the functional type from the unpolluted site, probably due to competitive
interactions between fungi. However, dominance effect maintained positive diversity effects under metal stress in assemblages
containing the functional type from the metal-polluted site. These findings emphasize the importance of intraspecific diversity
in modulating diversity effects under metal stress, providing evidence that trait-based diversity measures should be incorporated
