Surficial soil development was studied in four wetland basins created on the floodplain of the Des Plaines River near Chicago, Illinois, USA. These studies determined changes in the spatial distribution of plant-available nutrients as a result of establishing two different wetland hydrologic regimes. Three wetland basins had mineral soils and one an organic soil. A geostatistical analysis including kriging of collected data indicated that all soil parameters showed significant changes in their spatial structure as a result of the water inputs and unidirectional flows. The degree of spatial variability as indicated by autocorrelation in the soil data (i.e., points closer to one another are more similar than points further apart due to the influence of landscape processes) declined for all parameters except Mg+2. Temporal changes in the spatial patterns of extractable phosphorus (P) and percent organic carbon (OC) tended to be inverse; P declined in areas where OC increased and vice versa. The spatial pattern of these changes was dissimilar in the mineral soils as compared to the organic soil and was related to patterns of primary productivity. Zones of P uptake and OC accumulation were also related to wetland hydrology and primary productivity. Changes in the distribution of nutrients, particularly P, may be viewed as a result of nutrient spirals within the wetlands. By comparison, the reorganization in the concentrations of K+ and Ca+2 appear to have been mediated by cation exchange processes. The formation of new concentration gradients was strongly related to both flow pathways and the different water inflow rates. The formation of concentration gradients in exchangeable cations was not reflected in the average concentrations within each basin. Mean values changed significantly in only a few instances. Reducing data in this way missed important biogeochemical changes occurring within the experimental wetland basins. Targeted alteration of the genome lies at the heart of the exploitation of S. pombe as a model system. The rate of analysis is often determined by the efficiency with which a target locus can be manipulated. For most loci this is not a problem, however for some loci, such as fin1 +, rates of gene targeting below 5% can limit the scope and scale of manipulations that are feasible within a reasonable time frame. We now describe a simple modification of transformation procedure for directing integration of genomic sequences that leads to a 5-fold increase in the transformation efficiency when antibiotic based dominant selection markers are used. We also show that removal of the pku70 + and pku80 + genes, which encode DNA end binding proteins required for the non-homologous end joining DNA repair pathway, increases the efficiency of gene targeting at fin1 + to around 75–80% (a 16-fold increase). We describe how a natMX6/rpl42 + cassette can be used for positive and negative selection for integration at a targeted locus. To facilitate the evaluation of the impact of a series of mutations on the function of a gene of interest we have generated three vector series that rely upon different selectable markers to direct the expression of tagged/untagged molecules from distinct genomic integration sites. pINTL and pINTK vectors use ura4 + selection to direct disruptive integration of leu1 + and lys1 + respectively, while pINTH vectors exploit nourseothricin resistance to detect the targeted disruption of a hygromycin B resistance conferring hphMX6 cassette that has been integrated on chromosome III. Finally, we have generated a series of multi-copy expression vectors that use resistance to nourseothricin or kanamycin/G418 to select for propagation in prototrophic hosts. Collectively these protocol modifications and vectors extend the versatility of this key model system.  相似文献

Data extracted from video recordings of individual estuarine flocs near the estuary bed during the advance and retreat of the salt intrusion show changes in size and settling velocity distributions. The recordings were taken using INSSEV —IN Situ SEttling Velocity instrument. Size coupled with effective density variations due to both changes in floc structure and ambient salinity result in changes in the settling velocity during the tidal cycle. In particular, just after high water slack, the appearance of high settling velocity medium size flocs and individual particles suggest that the lower density flocs have been broken up by the intense vertical shear in the currents caused by the salt wedge intrusion. Current shear is shown to have a significant influence on floc effective density.  相似文献

Gonadotropin releasing-hormone analogue (buserelin) challenges were carried out every 8 weeks from 4 to 14 months of age on thoroughbred colts born in the spring (n = 6) or autumn (n = 5) to define the onset of puberty. In all colts, luteinizing hormone (LH) secretion followed a seasonal pattern, with high baseline and maximal concentrations in the spring and summer and low concentrations in the winter. Testosterone concentrations were undetectable before spring and, thus, autumn-born colts were younger than spring-born colts when a testosterone response to buserelin was first observed. Mean weights of the autumn-born colts were 300 kg (282-327 kg) at the time of the first detectable testosterone response in the following spring (October). Spring-born colts had reached this weight in the winter (May and June, before day length had increased) but did not exhibit a significant testosterone response until the spring at a mean weight of 352 kg (327-403 kg). It is proposed that colts must achieve a threshold body weight concurrently with stimulatory photoperiod for onset of puberty to occur.  相似文献

Thornicroft's giraffe, Giraffa camelopardalis thornicrofti, is a geographically isolated subspecies of giraffe found only in north‐east Zambia. The population only occurs in Zambia's South Luangwa Valley, an area which interestingly places it between the current distribution of Masai (G. c. tippelskirchi) giraffe to the north, and the Angolan (G. c. angolensis) and South African (G. c. giraffa) giraffe in the south‐west and south, respectively. Specific studies have been undertaken on the ecology of this subspecies, but their population genetics remains unknown. We studied 34 individuals from the South Luangwa National Park and adjacent Lupande Game Management Area and seven individuals from northern Botswana. The complete cytochrome b and control region sequences of the mitochondrial genome were sequenced and analysed together with database data by maximum likelihood tree reconstruction and maximum parsimony network analyses. The giraffe from Zambia's South Luangwa Valley are most closely related to the subspecies G. c. tippelskirchi and part of their radiation. However, they form a unique population that would benefit from increased research and conservation management.  相似文献

Two geographically distinct populations of giraffe (Giraffa camelopardalis) were sampled for this study, the northern Namib Desert and Etosha National Park. Population genetic parameters and relationships within subpopulations were estimated to better understand the genetic architecture of this isolated subspecies. Gene flow between the geographically separated populations can be attributed to recent translocation of giraffe between the two populations. Inbreeding estimates in the six subpopulations studied were low though we found evidence that genetic drift may be affecting the genetic diversity of the isolated populations in northern Namibia. Population dynamics of the sampling locations was inferred with relationship coefficient analyses. Recent molecular systematics of the Namibian giraffe populations indicates that they are distinct from the subspecies Giraffa camelopardalis giraffa and classified as G. c. angolensis. Based on genetic analyses, these giraffe populations of northern Namibia, the desert‐dwelling giraffe and those protected in Etosha National Park, are a distinct subspecies from that previously assumed; thus we add data on G. c. angolensis to our scientific knowledge of this giraffe of southern Africa.  相似文献

Measures of trophic position (TP) are critical for understanding food web interactions and human‐mediated ecosystem disturbance. Nitrogen stable isotopes (δ15N) provide a powerful tool to estimate TP but are limited by a pragmatic assumption that isotope discrimination is constant (change in δ15N between predator and prey, Δ15N = 3.4‰), resulting in an additive framework that omits known Δ15N variation. Through meta‐analysis, we determine narrowing discrimination from an empirical linear relationship between experimental Δ15N and δ15N values of prey consumed. The resulting scaled Δ15N framework estimated reliable TPs of zooplanktivores to tertiary piscivores congruent with known feeding relationships that radically alters the conventional structure of marine food webs. Apex predator TP estimates were markedly higher than currently assumed by whole‐ecosystem models, indicating perceived food webs have been truncated and species‐interactions over simplified. The scaled Δ15N framework will greatly improve the accuracy of trophic estimates widely used in ecosystem‐based management.  相似文献

In a flock of highly prolific Romney ewes obtained from industry flocks, one ewe (A281), with a production record of 33 lambs born in 11 lambings, produced a number of female descendants with high ovulation rates. The mode of inheritance of this trait was determined in a series of four progeny tests of male descendants of this ewe. The first progeny test produced strong evidence for a new major gene affecting ovulation rate in this family line; this finding was supported by two subsequent progeny tests. The fourth progeny test was designed to test the hypothesis that this gene is carried on the X chromosome. The results showed that six sons of a carrier ram did not inherit the gene, but it was passed on to three of his five maternal grandsons. This finding, together with evidence of genetic segregation in the progeny of carrier females, demonstrates for the first time the presence of a major gene for prolificacy specifically located on the X chromosome. The effect of the gene is to increase ovulation rate by about one additional egg per ewe.  相似文献
