Interactions between competing species may be intensified when they are restricted to small patches of remnant habitat, potentially increasing physiological stress in individuals. The effects of interspecific competition on stress in wildlife remain largely unexplored. In Australia, remnant woodlands are often dominated by aggressive honeyeaters, especially the noisy miner (Manorina melanocephala). Harassment of smaller birds by miners may result in their exclusion from suitable woodland habitat. We tested whether the presence of noisy miners is also associated with elevated stress in a model species of small passerine bird, the superb fairy‐wren (Malurus cyaneus). We sampled wrens from six sites, three remnant woodlands with noisy miners and three larger fragments of reserved habitat without noisy miners. Differential white blood cell counts were used to infer levels of chronic stress. We also assessed variation in body condition and the prevalence of blood parasites (Haemoproteus spp.) to test for associations between stress and parasitemia. The mean heterophil‐to‐lymphocyte (H:L) ratio was 1.8 × higher among superb fairy‐wrens living in miner‐dominated woodlands, suggesting higher levels of chronic stress. Individuals with higher stress appeared to be in poorer condition, as indicated by fat scores and residual body mass. Prevalence of blood parasites was generally high and was highest in reserved habitat (59%) where miners were absent. Birds with blood parasites living in these habitats had higher H:L ratios but the intensity of infection and H:L ratio was inversely related. Our results suggest that birds persisting in the presence of noisy miners might experience chronic stress, but further study is necessary to separate the relative importance of noisy miner aggression from other potential stressors in small patches of degraded woodland. Stress induced by interspecific aggression should be considered in future studies of wildlife living in remnant vegetation.  相似文献

In tropical soils, diversity and biotechnological potential of symbiotic diazotrophic bacteria are high. However, the phylogenetic relationships of prominent strains are still poorly understood. In addition, in countries such as Brazil, despite the broad use of rhizobial inoculants, molecular methods are rarely used in the analysis of strains or determination of inoculant performance. In this study, both rep-PCR (BOX) fingerprintings and the DNA sequences of the 16S rRNA gene were obtained for 54 rhizobial strains officially authorized for the production of commercial inoculants in Brazil. BOX-PCR has proven to be a reliable fingerprinting tool, reinforcing the suggestion of its applicability to track rhizobial strains in culture collections and for quality control of commercial inoculants. On the other hand, the method is not adequate for grouping or defining species or even genera. Nine strains differed in more than 1.03% (15) nucleotides of the 16S rRNA gene in relation to the closest type strain, strongly indicative of new species. Those strains were distributed across the genera Burkholderia, Rhizobium, and Bradyrhizobium.  相似文献

One-electron quinone reduction and two-electron carbonyl reduction convert the anticancer anthracycline doxorubicin to reactive oxygen species (ROS) or a secondary alcohol metabolite that contributes to inducing a severe form of cardiotoxicity. The closely related analogue epirubicin induces less cardiotoxicity, but the determinants of its different behavior have not been elucidated. We developed a translational model of the human heart and characterized whether epirubicin exhibited a defective conversion to ROS and secondary alcohol metabolites. Small myocardial samples from cardiac surgery patients were reconstituted in plasma that contained clinically relevant concentrations of doxorubicin or epirubicin. In this model only doxorubicin formed ROS, as detected by fluorescent probes or aconitase inactivation. Experiments with cell-free systems and confocal laser scanning microscopy studies of H9c2 cardiomyocytes suggested that epirubicin could not form ROS because of its protonation-dependent sequestration in cytoplasmic acidic organelles and the consequent limited localization to mitochondrial one-electron quinone reductases. Accordingly, blocking the protonation-sequestration mechanism with the vacuolar H+-ATPase inhibitor bafilomycin A1 relocalized epirubicin to mitochondria and increased its conversion to ROS in human myocardial samples. Epirubicin also formed approximately 60% less alcohol metabolites than doxorubicin, but this was caused primarily by its higher Km and lower Vmax values for two-electron carbonyl reduction by aldo/keto-reductases of human cardiac cytosol. Thus, vesicular sequestration and impaired efficiency of electron addition have separate roles in determining a defective bioactivation of epirubicin to ROS or secondary alcohol metabolites in the human heart. These results uncover the molecular determinants of the reduced cardiotoxicity of epirubicin and serve mechanism-based guidelines to improving antitumor therapies.  相似文献

Bradyrhizobium comprises most tropical symbiotic nitrogen-fixing strains, but the correlation between symbiotic and core genes with host specificity is still unclear. In this study, the phylogenies of the nodY/K and nifH genes of 45 Bradyrhizobium strains isolated from legumes of economic and environmental importance in Brazil (Arachis hypogaea, Acacia auriculiformis, Glycine max, Lespedeza striata, Lupinus albus, Stylosanthes sp. and Vigna unguiculata) were compared to 16S rRNA gene phylogeny and genetic diversity by rep-PCR. In the 16S rRNA tree, strains were distributed into two superclades—B. japonicum and B. elkanii—with several strains being very similar within each clade. The rep-PCR analysis also revealed high intra-species diversity. Clustering of strains in the nodY/K and nifH trees was identical: 39 strains isolated from soybean grouped with Bradyrhizobium type species symbionts of soybean, whereas five others occupied isolated positions. Only one strain isolated from Stylosanthes sp. showed similar nodY/K and nifH sequences to soybean strains, and it also nodulated soybean. Twenty-one representative strains of the 16S rRNA phylogram were selected and taxonomically classified using a concatenated glnII-recA phylogeny; nodC sequences were also compared and revealed the same clusters as observed in the nodY/K and nifH phylograms. The analyses of symbiotic genes indicated that a large group of strains from the B. elkanii superclade comprised the novel symbiovar sojae, whereas for another group, including B. pachyrhizi, the symbiovar pachyrhizi could be proposed. Other potential new symbiovars were also detected. The co-evolution hypotheses is discussed and it is suggested that nodY/K analysis would be useful for investigating the symbiotic diversity of the genus Bradyrhizobium.  相似文献

Trypanosoma cruzi is a hemoflagellate protozoan that causes Chagas’ disease. The life cycle of T. cruzi is complex and involves different evolutive forms that have to encounter different environmental conditions provided by the host. Herein, we performed a functional assessment of mitochondrial metabolism in the following two distinct evolutive forms of T. cruzi: the insect stage epimastigote and the freshly isolated bloodstream trypomastigote. We observed that in comparison to epimastigotes, bloodstream trypomastigotes facilitate the entry of electrons into the electron transport chain by increasing complex II-III activity. Interestingly, cytochrome c oxidase (CCO) activity and the expression of CCO subunit IV were reduced in bloodstream forms, creating an “electron bottleneck” that favored an increase in electron leakage and H2O2 formation. We propose that the oxidative preconditioning provided by this mechanism confers protection to bloodstream trypomastigotes against the host immune system. In this scenario, mitochondrial remodeling during the T. cruzi life cycle may represent a key metabolic adaptation for parasite survival in different hosts.  相似文献

Purpose

Sustainable development aims to enhance the quality of life by improving the social, economic and environmental conditions for present and future generations. A sustainable engineering decision-making strategy for design and assessment of construction works (i.e., civil engineering and buildings) should take into account considerations regarding the society, the economy and the environment. This study presents a novel approach for the life cycle assessment (LCA) of a case-study building subjected to seismic actions during its service life, accounting for structural reliability.

Methods

A methodology is presented that evaluates the time-dependent probability of exceeding a limit state considering the uncertainty in the representation of seismic action. By employing this methodology, the earthquake-induced damages are related to the environmental and social losses caused by the occurrence of the earthquake. A LCA of a case-study building accounting for the time-dependent seismic reliability is conducted using a damage-oriented LCA approach.

Results and discussion

The contributions of the different life cycle phases to the total environmental impact related to the building lifetime are in agreement with previous results in this field of study. However, the LCA results revealed significant risk-based contributions for the rehabilitation phase due to the induced damage resulting in seismic events. Particularly, the rehabilitation phase is expected to contribute to the total environmental impact with around the 25 % of the initial environmental impact load (related to the pre-use phase) as a consequence of seismic damage.

Conclusions and recommendations

The probability of occurrence of seismic events affects the LCA results for various life cycle phases of a building in terms of all the indicators adopted in the analysis. The time-dependent probability of collapse in a year can represent a benchmark indicator for human safety in the context of social sustainability for the building sector. The proposed approach can be implemented in a sustainable decision-making tool for design and assessment.  相似文献

Actin‐based remodelling underlies spine structural changes occurring during synaptic plasticity, the process that constantly reshapes the circuitry of the adult brain in response to external stimuli, leading to learning and memory formation. A positive correlation exists between spine shape and synaptic strength and, consistently, abnormalities in spine number and morphology have been described in a number of neurological disorders. In the present study, we demonstrate that the actin‐regulating protein, Eps8, is recruited to the spine head during chemically induced long‐term potentiation in culture and that inhibition of its actin‐capping activity impairs spine enlargement and plasticity. Accordingly, mice lacking Eps8 display immature spines, which are unable to undergo potentiation, and are impaired in cognitive functions. Additionally, we found that reduction in the levels of Eps8 occurs in brains of patients affected by autism compared to controls. Our data reveal the key role of Eps8 actin‐capping activity in spine morphogenesis and plasticity and indicate that reductions in actin‐capping proteins may characterize forms of intellectual disabilities associated with spine defects.  相似文献

BACKGROUND: The neurotrophins, which include nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT-3), NT-4/5 and NT-6, are a family of proteins that play fundamental roles in the differentiation, survival and maintenance of peripheral and central neurons. Much research has focused on the role of neurotrophins as target-derived, retrogradely transported trophic molecules. Although there is recent evidence that BDNF and NT-3 can be transported in an anterograde direction along peripheral and central axons, there is as yet no conclusive evidence that these anterograde factors have direct post-synaptic actions. RESULTS: We report that BDNF travels in an anterograde direction along the optic nerve. The anterogradely transported BDNF had rapid effects on retinal target neurons in the superior colliculus and lateral geniculate nucleus of the brain. When endogenous BDNF within the developing superior colliculus was neutralised, the rate of programmed neuronal death increased. Conversely, provision of an afferent supply of BDNF prevented the degeneration of geniculate neurons after removal of their cortical target. CONCLUSIONS: BDNF released from retinal ganglion cells acts as a survival factor for post-synaptic neurons in retinal target fields.  相似文献
