Loss of tumorigenic potential by human lung tumor cells in the presence of antisense RNA specific to the ectopically synthesized alpha subunit of human chorionic gonadotropin

A clonal strain of human lung tumor cells in culture (ChaGo), derived from a bronchogenic carcinoma, synthesizes and secretes large amounts of alpha (alpha) and a comparatively lower level of beta (beta) subunit of the glycoprotein hormone, human chorionic gonadotropin (HCG). ChaGo cells lost their characteristic anchorage-independent growth phenotype in the presence of anti-alpha-HCG antibody. The effect of the antibody was partially reversed by addition of alpha-HCG to the culture medium. ChaGo cells were transfected with an expression vector (pRSV-anti-alpha-HCG), that directs synthesis of RNA complementary to alpha-HCG mRNA. The transfectants produced alpha-HCG antisense RNA which was associated with the reduced level of alpha-HCG. Transfectants also displayed several altered phenotypic properties, including altered morphology, less mitosis, reduced growth rate, loss of anchorage-independent growth, and loss of tumorigenicity in nude mice. Treatment of transfectants with 8,bromo-cAMP resulted in increased accumulation of alpha-HCG mRNA, no change in the level of alpha-HCG antisense RNA, release of the inhibition of [3H]thymidine incorporation, and restoration of anchorage-independent growth phenotype. The overexpression of c-myc, observed in ChaGo cells, was unaffected by the reduced level of alpha-HCG. These results suggest that ectopic synthesis of the alpha subunit of HCG plays a functional role in the transformation of these human lung cells.

自花授粉作物杂交育种组合潜势的预测和比较

在黄花烟草Nicotiana russica中,开花期这一性状用V_5作亲本可能产生较大变异,无论是选择早开花的还是选择迟开花的,都有较大机会得到理想目标株系;对于株高这一性状,含有V(?)的组合可望有较大机会产生高于标准品种的株系。通过组合间育种潜势的比较,能了解各亲本中基因分布的基本情况,进而进行客观评价并对杂交组合做出取舍。试验证明,用一个组合的早期世代的参数m和D来预测高世代或纯系的育种潜势是可行的。     

反复电针对慢性痛的累加治疗作用及其机制研究

本研究从基础和临床两方面观察了反复电针对慢性痛的累加治疗作用,并结合疼痛患者及慢性痛动物模型中几种神经肽的放射免疫测定及相应受体拮抗剂的药理学研究结果,探讨了产生累加效应的可能机制。结果表明,在临床脊髓损伤性痉挛患者,１００Ｈｚ穴位体表电刺激有效地缓解痉挛并有累加效应;在临床慢性痛患者,２／１５Ｈｚ变频ＴＥＮＳ刺激有效地治疗疼痛并具有累加效应。在关节炎模型大鼠,电针刺激能产生明显的镇痛并具有累加效     

Isolation and identification of a non-specific tandemly repeated DNA sequence in Oryza species

A tandemly repeated DNA sequence (RRS7) was isolated from Oryza alta (CCDD). RRS7-related sequences were also found tandemly arrayed in genomes AA, BB, BBCC, CC, and EE, and a small amount of RRS7-related sequences were detected in genome FF and the Oryza species with unknown genomes. DNA sequence analysis of the 1844-bp insert of RRS7 revealed that it contained six tandemly repeated units, of which five were 155 bp in length and one was 194 bp in length and contained an imperfect internal 39-bp duplication. Southern blot analysis showed that the boundary sequence contained in RRS7 is a single-copy sequence. A 155-bp consensus sequence derived from the six monomeric repeats contained no internal repeat and showed no significant homology to other currently known sequences. The results of Southern blot and sequence analysis revealed that there are at least two subfamilies present in the RRS7 family; these are represented by the DraI site and the MspI site, respectively. Restriction digestion with two pairs of isoschizomers MboI/Sau3A and MspI/HpaII demonstrated that most of the C residues in the GATC sites and the internal C in the CCGG sites of the RRS7 family in O. Alta were methylated. The usefulness of the RRS7 family in determining the evolutionary relationship of the genome DD and other Oryza genomes is discussed.

控制大豆白花亲本籽粒脐色的基因有带Ｒ与ｒ之分,带Ｒ基因的白花产本与紫花亲本杂交,Ｆ１代籽料出现蓝脐性状,其基因型为Ｉ－Ｒ－Ｗ１－ｔｔ。当控制脐色的基因有两对相差时（Ｒ、ｒ;Ｗ１、ｗ１）Ｆ２代籽粒脐色分离蓝脐与无色脐之比为９∶７。     

长白山劲松林场植物群落的分类和排序

将长白山白河林业局劲松林场的５４块样地用聚类分析法划分成６个植被类群,再用ＰＣ－ＶＴＡＢ程序中经过改进的Ｂｒａｕｎ－Ｂｌａｎｑｕｅｔ学派的植被排表分析法进行综合,产生了鉴别概要表,为各个等级的植被类群筛选出诊断种。此外,还用鉴别种地样地记录进行主成分分析,以验证诊断种的有效性,结果表明,ＰＣ－ＶＴＡＢ中的植被排表分析法是筛选鉴别种的有效方法,而鉴别种以显著地提高植被分析和排序的质量。     

人鼻咽癌细胞基因组中瘤基因Ha－ras－1的Dnase Ⅰ超敏感位点的初步研究

ＤＮａｓｅⅠ超敏感位点的研究能够发现潜在的调控基因转录活化的位点,比较正常人外周血有核细胞,淋巴瘤细胞株Ｐ３ＨＲ１和人鼻咽癌低分化磷癌细胞株ＨＯｎＥ１和ＨＮＥ２中Ｈａ－ｒａｓ－１瘤基因的ＤＮａｓｅⅠ超敏感位点发现,只有ＨＯＮＥ１和ＨＮＥ２细胞基因组中存在一个ＤＮａｓｅⅠ超敏感位点,位于第一个外显子上游０．３７ｋｂ处,上述结果提示正常白细胞和Ｐ３ＨＲ１细胞中Ｈａ－ｒａｓ－１基因处于失活状态,而在鼻咽癌细胞基因组中则处于活化状态,它的活化可能与０．３７ｋｂ处的ＤＮＡ序列有密切的关系。

通过对两种肥力条件下的“8455”小麦植株化学成分与麦长管蚜和麦二叉蚜种群消长关系的研究与分析,结果表明可溶性糖、含钾量、含水量是影响麦二叉蚜种群消长的主要因子,胱氨酸是影响麦长管蚜种群消长的主要因子。
