通过放射性标记DNA探针筛选和PCR克隆筛选 ,从皖南尖吻蝮蛇毒腺的λgt11cDNA文库中 ,克隆得到了 11个编码C型凝集素类似蛋白亚单位的cDNA .其中 2个克隆分别含有编码an tithrombinA的A链和B链的序列 .另外 6个则含有编码如下肽链的序列 :antithrombinC的A链和B链、agkisacutacin的A链和B链、抗凝血因素的A链和B链 ,其它 3个克隆含有编码未知蛋白质的序列 .有趣的是 ,在编码AntithrombinC的序列中 ,每条链中除了含有 7个与所有C型凝集素类似蛋白位置一致的Cys外 ,在B链上还含有一个额外的Cys3;2条B链上的Cys3可能形成一对新的链间二硫键 .氨基酸序列的比较及进化分析显示 ,C型凝集素类似蛋白B链起源于一个单系类群 ,A链至少有 4个进化起源  相似文献

The ocean is a natural habitat for antibiotic-producing bacteria, and marine aquaculture introduces antibiotics into the ocean to treat infections and improve aquaculture production. Studies have shown that the ocean is an important reservoir of antibiotic resistance genes. However, there is a lack of understanding and knowledge about the clinical importance of the ocean resistome. We investigated the relationship between the ocean bacterial resistome and pathogenic resistome. We applied high-throughput sequencing and metagenomic analyses to explore the resistance genes in bacterial plasmids from marine sediments. Numerous putative resistance determinants were detected among the resistance genes in the sediment bacteria. We also found that several contigs shared high identity with transposons or plasmids from human pathogens, indicating that the sediment bacteria recently contributed or acquired resistance genes from pathogens. Marine sediment bacteria could play an important role in the global exchange of antibiotic resistance.  相似文献

Human serum albumin (HSA) and human parathyroid hormone (1-34) [PTH (1-34)] fusion protein [HSA/PTH (1-34)] is a promising long-acting form of PTH (1-34) for osteoporosis treatment. Secretory expression of intact HSA/PTH (1-34) in Pichia pastoris GS115 was accompanied by two degradation fragments, with molecular weights around 66 kDa, in addition to the well-known ~45 kDa HSA-truncated fragment, resulting in a low yield of intact protein. In this study, two internal cleavage sites were identified in the PTH (1-34) portion of the fusion protein by Western Blot analysis. To minimize proteolytic cleavages, several protease genes including PEP4 (encoding proteinase A), PRB1 (proteinase B) and seven YPSs genes (yapsin family members) were knocked out respectively by disruption of the individual genes and the selective combinations. Reduced degradation was observed by single disruption of either PEP4 gene or YPS1 gene, and the lowest level of degradation was observed in a pep4yps1△ double disruptant. After 72 h of induction, more than 80 % of the HSA/PTH (1-34) secreted by the pep4yps1△ double disruptant remained intact, in comparison to only 30 % with the wild-type strain.  相似文献

四种冬季水生植物组合对富营养化水体的净化效果   总被引:3,自引:0,他引:3  
选择10种耐低温的水生植物构建4种植物组合,研究了冬季低温环境下不同水生植物组合对富营养化水体的净化效果.结果表明:组合1[常绿水生鸢尾(Iris hexagonus Hybrid)*羊蹄(Rumex japonicus)+金叶“金钱蒲”(Acorus gramineus“Ogan”)+反曲灯心草“蓝箭”(Juncus inflexus“Blue Arrows”)]4种植物均能在试验富营养化水体中茂盛生长,且对TN、NOx-N、NH4-N和TP的去除率分别为47.8%、52.2%、32.4%和70.1%;组合2[常绿水生鸢尾+羊蹄+金叶“金钱蒲”+大苞萱草(Hemerocallis middendorfii)]4种植物也都能在试验富营养化水体中存活,并且有一定量的生长,对TN、NOx-N、NH4-N和TP的去除率分别为44.2%、58.5%、34.6%和67.8%;而未种植物的对照对TN、NOx-N、NH4-N和TP的去除率分别为40.0%、25.9%、27.3%和64.5%;组合1和2对富营养化水体有较好的净化效果.组合3和组合4中由于吊兰(Chlorophytum comosum)和三穗薹草(Carex tristachya)等植物长势较差,仅对NOx-N具有较明显去除能力,对其他指标去除效果不明显.通过这些水生植物在富营养化水体中生长特性和对营养元素的去除能力,发现冬季组合1和2的净化效果较好,是低温条件下适宜的浮床植物组合形式.  相似文献

Syngas fermentation is a promising route for resource recovery. Acetate is an important industrial chemical product and also an attractive precursor for liquid biofuels production. This study demonstrated high fraction acetate production from syngas (H2 and CO2) in a hollow-fiber membrane biofilm reactor, in which the hydrogen utilizing efficiency reached 100 % during the operational period. The maximum concentration of acetate in batch mode was 12.5 g/L, while the acetate concentration in continuous mode with a hydraulic retention time of 9 days was 3.6?±?0.1 g/L. Since butyrate concentration was rather low and below 0.1 g/L, the acetate fraction was higher than 99 % in both batch and continuous modes. Microbial community analysis showed that the biofilm was dominated by Clostridium spp., such as Clostridium ljungdahlii and Clostridium drakei, the percentage of which was 70.5 %. This study demonstrates a potential technology for the in situ utilization of syngas and valuable chemical production.  相似文献

以山梨猕猴桃(Actinidia rufa(Siebold and Zuccarini)Planchon ex Miquel)品种‘63101’和中华猕猴桃(Actinidia chinensis var.chinensis C. F. Liang)品种‘磨山雄7号’杂交产生的400余株F1代个体为材料,连续2~3年对其果实品质和外观特点的9个性状进行测定,分析它们的遗传规律。结果显示,子代群体单果质量连续3年低于双亲中值,遗传传递力连续3年低于100%,优势率连续3年均为负值;可溶性固形物子代中值除2015年略低外,其余2年均高于亲中值12.3%;优势率均为正值,遗传传递力均大于100%;子代果实形状出现分离,与母本圆柱形相同的比例占82%;果皮毛被中,偏离母本的有毛比例为60%;果皮颜色出现绿色、绿褐色、褐色的分离,比例为1∶1.5∶1;果实肩部形状多与母本的方形相似,占比为63%;果肉颜色也多与母本的绿色相似,占比为86%;果实风味中,与父本和母本相似的表型占比均为32%,口感酸的占比18%,超低亲极酸占比1%,超高亲浓甜占比9%。说明杂交子代单果质量和可溶...  相似文献

The purpose of this study was to investigate the biological effect of miR‐16 on myocarditis and the underlying molecular mechanism. H9c2 cells were treated with 10 µg/mL lipopolysaccharide (LPS) for 12 hours to form a myocarditis injury model. We observed that LPS treatment distinctly decreased the level of miR‐16 in H9c2 cells. Upregulation of miR‐16 increased cell proliferation and reduced cell apoptosis. Then, CD40 was predicted and verified as a target gene of miR‐16 by TargetScan and luciferase reporter assay, respectively. Furthermore, the messenger RNA and protein expression of CD40 are negatively regulated by miR‐16. The relative expression of inflammatory factors was dramatically decreased by the miR‐16 mimic. Cells cotransfected with miR‐16 mimic and si‐CD40 could significantly abolish the injury of cardiomyocytes caused by myocarditis. Our study illustrated that the upregulation of miR‐16 has a protective effect on LPS‐damaged H9c2 cells, which may be achieved by regulating CD40 and the nuclear factor kappa B pathway.  相似文献

Single‐layered organic solar cells (OSCs) using nonfullerene acceptors have reached 16% efficiency. Such a breakthrough has inspired new sparks for the development of the next generation of OSC materials. In addition to the optimization of electronic structure, it is important to investigate the essential solid‐state structure that guides the high efficiency of bulk heterojunction blends, which provides insight in understanding how to pair an efficient donor–acceptor mixture and refine film morphology. In this study, a thorough analysis is executed to reveal morphology details, and the results demonstrate that Y6 can form a unique 2D packing with a polymer‐like conjugated backbone oriented normal to the substrate, controlled by the processing solvent and thermal annealing conditions. Such morphology provides improved carrier transport and ultrafast hole and electron transfer, leading to improved device performance, and the best optimized device shows a power conversion efficiency of 16.88% (16.4% certified). This work reveals the importance of film morphology and the mechanism by which it affects device performance. A full set of analytical methods and processing conditions are executed to achieve high efficiency solar cells from materials design to device optimization, which will be useful in future OSC technology development.  相似文献

生态系统水分利用效率(Water Use Efficiency,WUE)是衡量碳水循环耦合程度的重要指标,估算安徽省农田生态系统的WUE并分析其时空变异规律,探究其影响因素,对该区域农田水资源的科学配置和合理利用有着重要的意义。基于MODIS遥感数据和气象数据,分析安徽省近15年农田WUE的年际、年内时空变化特征以及与气候因子的关系,研究表明:(1) 2000—2014年间安徽省农田WUE的变化范围为1.38—1.66 g C mm~(-1)m~(-2),多年均值为1.54 g C mm~(-1)m~(-2),整体上农田WUE年际呈现增长变化趋势,变化率为0.011 g C mm~(-1)m~(-2);具有较强的空间分异性规律,整体上呈现北高南低趋势,淮河以北各市的农田WUE较高,高于全省多年均值,淮河以南地区的农田WUE则低于全省水平。(2)安徽省农田WUE的年内变化呈现双峰型变化格局,具有明显的季节性差异,表现为春季秋季夏季冬季;以淮河为界,南北各市变化趋势略有不同,淮河以南地区的最大值在4月份,淮河以北则以3月份为最高。(3)安徽省农田WUE动态变化受到气候因子降雨影响的区域占比17.14%;气温影响的区域占比0.73%;降雨和气温综合影响所占0.71%,而农田WUE与气温和降雨影响均不显著的区域占比为81.42%;因此,气候因素中降雨在安徽省农田WUE的变化中起主导作用,由于人为因素的干预,非气候因素对农田WUE变化的影响更大。  相似文献
