Microcystins are cyanotoxins that occur in ground water and thus pose a potential health risk. Microcystin-LR (microcystin-leucine-arginine) is a potent hepatotoxin, and is suspected of being a tumour promoter. Poisoning with this toxin causes several dysfunctions in hepatocytes by inhibiting protein phosphatases 1 and 2A, and notably produces oxidative stress, disrupts the cytoskeleton, and deregulates mitogen-activated protein kinase pathway. Medaka fish (Oryzias latipes) was chosen as a model for studying the effects of this cyanotoxin on liver proteins using a gel-free approach, iTRAQ. Fish were gavaged with microcystin-LR. Two hours later, 325 proteins could be identified by Scaffold Q+ and 32 proteins revealed statistically significant variations above a ∣0.2∣ threshold of log(2) ratio by comparison with control. These proteins are mostly involved in the translation and maturation of proteins, lipid metabolism and detoxification. This study demonstrates that under subtropical environmental conditions the circulating levels of triiodothyronine (T 3) and thyroxine (T 4) in adult male roseringed parakeets undergo parallel changes according to a diurnal cycle with a trough in the early morning and a peak in the evening. Chronic afternoon administration of melatonin (25 µg/ 100 g body wt./ day for 30 consecutive days) abolishes time-bound changes in the concentrations of T 3 by suppressing its evening peak, but does not abolish the diurnal rhythm of T 4. Exogenous melatonin, however, causes a significant increase in the evening value of serum T 4 and a decrease in its value at midnight resulting in phase-shifting of the diurnal fluctuations of T 4 compared to that in the control birds. It appears that exogenous melatonin can alter the diurnal profiles of serum thyroid hormones in a wild psittacine bird. 相似文献

Harbour seals are active at night and during the day and see well in both air and water. Polarised light, which is a well-known visual cue for orientation, navigation and foraging, is richly available in harbour seal habitats, both above and below the water surface. We hypothesised that an ability to detect and use polarised light could be valuable for seals, and thus tested if they are able to see this property of light. We performed two behavioural experiments, one involving object discrimination and the other involving object detection. These objects were presented to the seals as two-dimensional stimuli on a specially modified liquid crystal display that generated objects whose contrast was purely defined in terms of polarisation (i.e. objects lacked luminance contrast). In both experiments, the seals’ performance did not deviate significantly from chance. In contrast, the seals showed a high baseline performance when presented with objects on a non-modified display (whose contrast was purely defined in terms of luminance). We conclude that harbour seals are unable to use polarised light in our experimental context. It remains for future work to elucidate if they are polarisation insensitive per se. 相似文献

During the years 2015–2016, we studied the fauna of epigeon in eight similar habitats in Slovakia and Serbia. The study material of Opiliones was obtained by a pitfall trap method. We focused on the comparison of the incidence of harvestmen in floodplain forests and ecotones alongside the Danube River in Slovakia and along the Tisa and Begej River in Serbia. We examined their dependence on anthropogenic impact (frequency of visits), cover of vegetation layers (herbs, shrubs and trees), plant diversity of vegetation layers, area of forest stands, distance to the forest edge, circumference of forest stands, age of forest stands, thickness of litter layer and physico-chemical properties of soil and litter (pH, conductivity, P, N, C, H). Total epigeic activity of harvestman showed significant positive relationship with proportion of N in litter. Community equitability was positively related with species richness of plant communities in shrub layer and negatively related with H content of soil. We did not find any significant correlations between diversity of harvestman communities and measured environmental characteristics. 相似文献

Two new ternary tetrazolate Eu(III) complexes with phosphine oxide co‐ligands Eu(PTO)3·(P1/P2) [PTO = 5‐(2‐pyridyl‐1‐oxide)tetrazole, P1 = diphenylphosphorylamino‐phenylphosphoryl‐benzene, P2 = diphenylphosphorylpyridine)‐bis‐isobutyricphosphoryl] were synthesized and characterized using UV, fluorescence, IR and 1H NMR spectroscopic techniques. The analytical data prove that the complexes are mononuclear in nature and the central Eu(III) ion is coordinated by three N and three O atoms of tetrazolate, and two O atoms of the corresponding bidentate phosphine oxide ligands. The ancillary ligand increased the photoluminescence efficiency of Eu(PTO)3·P1 (complex 3) by twofold compared with our previously reported Eu(PTO)3 complex (complex 1). In 2005, the International Society of Urological Pathology (ISUP) introduced several modifications to the original Gleason system that were intended to enhance the prognostic power of Gleason score (GS). The objective of this study was to clinically validate the 2005 ISUP Gleason grading system for its ability to detect metastasis. We queried our institutional RP database for men with NCCN clinically localized intermediate to high-risk disease undergoing radical prostatectomy (RP) between 1992 and 2010 with no additional treatment until the time of metastatic progression. A case-cohort design was utilized. A total of 333 available RP samples were re-reviewed and GS was reassigned per the 2005 ISUP Gleason system. Cumulative incidence of metastasis was 0%, 8.4%, 24.5% and 44.4% among specimens that were downgraded, unchanged, had one point GS increase and two point GS increase, respectively. The hazard ratio for metastasis raised in GS 8 and 9 compared to GS 7 from 2.77 and 5.91 to 3.49 and 9.31, respectively. The survival c-index of GS increased from 0.70 to 0.80 when samples were re-graded at 5 years post RP. The c-index of the reassigned GS was higher than the original GS (0.77 vs 0.64) for predicting PCSM at 10 years post RP. The regraded GS improved the prediction of metastasis and PCSM. This validates the updated Gleason grading system using an unambiguous clinical endpoint and highlights the need for reassignment of Gleason grading according to 2005 ISUP system when considering comparisons of novel biomarkers to clinicopathological variables in archival cohorts. 相似文献

Inflammation and oxidative and dicarbonyl stress play important roles in the pathogenesis of type 2 diabetes. Metformin is the first-line drug of choice for the treatment of type 2 diabetes because it effectively suppresses gluconeogenesis in the liver. However, its “pleiotropic” effects remain controversial. In the current study, we tested the effects of metformin on inflammation, oxidative and dicarbonyl stress in an animal model of inflammation and metabolic syndrome, using spontaneously hypertensive rats that transgenically express human C-reactive protein (SHR-CRP). We treated 8-month-old male transgenic SHR-CRP rats with metformin (5 mg/kg/day) mixed as part of a standard diet for 4 weeks. A corresponding untreated control group of male transgenic SHR-CRP rats were fed a standard diet without metformin. In a similar fashion, we studied a group of nontransgenic SHR treated with metformin and an untreated group of nontransgenic SHR controls. In each group, we studied 6 animals. Parameters of glucose and lipid metabolism and oxidative and dicarbonyl stress were measured using standard methods. Gene expression profiles were determined using Affymetrix GeneChip Arrays. Statistical significance was evaluated by two-way ANOVA. In the SHR-CRP transgenic strain, we found that metformin treatment decreased circulating levels of inflammatory response marker IL-6, TNFα and MCP-1 while levels of human CRP remained unchanged. Metformin significantly reduced oxidative stress (levels of conjugated dienes and TBARS) and dicarbonyl stress (levels of methylglyoxal) in left ventricles, but not in kidneys. No significant effects of metformin on oxidative and dicarbonyl stress were observed in SHR controls. In addition, metformin treatment reduced adipose tissue lipolysis associated with human CRP. Possible molecular mechanisms of metformin action–studied by gene expression profiling in the liver–revealed deregulated genes from inflammatory and insulin signaling, AMP-activated protein kinase (AMPK) signaling and gluconeogenesis pathways. It can be concluded that in the presence of high levels of human CRP, metformin protects against inflammation and oxidative and dicarbonyl stress in the heart, but not in the kidney. Accordingly, these cardioprotective effects of metformin might be especially effective in diabetic patients with high levels of CRP. 相似文献

Small transmembrane proteins of the tetraspanin superfamily are believed to function as the main structural blocks of specialized membrane microdomains (referred to as tetraspanin-enriched microdomains, TERM or TEM). Through a multitude of homotypic and heterotypic interactions, tetraspanins regulate lateral clustering and, consequently, signalling involving adhesion and growth factor receptors as well as costimulatory proteins. The presence of major histocompatibility complex (MHC) I and MHCII molecules in TERM led to suggestion of tetraspanins' involvement in antigen presentation. In addition, certain tetraspanins function as viral co-receptors and may be important for viral egress from infected cells. It has recently become apparent that in addition to their purely structural function as organizers of TERM, tetraspanins also regulate various aspects of trafficking and biosynthetic processing of associated receptors.
