The objective was to identify postpartum risk factors between nutritional imbalance and health disorders affecting first-service conception risk (FSCR) in 21 commercial Holstein herds in Reunion Island. Multivariate logistic-regression models including herd as a random effect were used to analyze the relationship between FSCR and energy status, nitrogen status, hepatic function, mineral deficiencies, and postpartum health disorders. Two models (A and B) were built on two subsets of data (n=446 and n=863) with risk indicators measured during the first month of lactation and around time of first service, respectively, adjusted for season, breed, parity, origin, milk yield, calving to first service interval (CS1), and type of estrus (spontaneous vs. induced). The averaged conception risk was 0.266+/-0.015 (n=913) (mean+/-S.E.M.). In both models, FSCR was decreased by CS1 < or = 60 d and induced estrus. In model A, FSCR was decreased (p<0.05) for cows with mean cumulative 100 d daily milk yield < or =23 kg/d and >27 kg/d, with losses of body condition score >1.5, and with retained placenta. In model B, FSCR was decreased (p<0.05) for cows inseminated during wet season, previously raised out of the farm as nulliparous, with blood magnesium concentration < or =0.9 mmol/L, and for high-yielding cows (100 d milk yield > 27 kg/d) with glutamate deshydrogenase>17 UI/L. Hence, high-body-lipid mobilization during the first month of lactation was a strong nutritional predictor of low FSCR together with liver damage in high-yielding cows. Interestingly, our models revealed that infertility is better related to nutritional factors than to postpartum health disorders occurrence.  相似文献

Hepcidin is a liver produced cysteine-rich peptide hormone that acts as the central regulator of body iron metabolism. Hepcidin is synthesized under the form of a precursor, prohepcidin, which is processed to produce the biologically active mature 25 amino acid peptide. This peptide is secreted and acts by controlling the concentration of the membrane iron exporter ferroportin on intestinal enterocytes and macrophages. Hepcidin binds to ferroportin, inducing its internalization and degradation, thus regulating the export of iron from cells to plasma. The aim of the present study was to develop a novel method to produce human and mouse recombinant hepcidins, and to compare their biological activity towards their natural receptor ferroportin. Hepcidins were expressed in Escherichia coli as thioredoxin fusion proteins. The corresponding peptides, purified after cleavage from thioredoxin, were properly folded and contained the expected four-disulfide bridges without the need of any renaturation or oxidation steps. Human and mouse hepcidins were found to be biologically active, promoting ferroportin degradation in macrophages. Importantly, biologically inactive aggregated forms of hepcidin were observed depending on purification and storage conditions, but such forms were unrelated to disulfide bridge formation.  相似文献

Monoclonal antibodies play an increasingly important role in structural biology. In this report, we develop the use of phage display technology for the isolation of an antibody that binds to a specific subunit of a macromolecular assembly. Antibodies that bind to the intact complex are selected from a phage display library and screened with a high-density Western blot to identify a subunit-specific binder. Conventional Western blotting and competition ELISA are then used to confirm the identity of the target subunit and that the antibody binds to the native protein complex and not to an epitope that is only revealed when the antibody is immobilized for phage selection. Using this technique, monoclonal scFv and Fab fragments have been produced that bind to the 51-kDa subunit of bovine complex I, a large integral membrane protein complex from mitochondria.  相似文献

The flowers of mangrove Rhizophoraceae (tribe Rhizophoreae) are adapted to three different pollination mechanisms. Floral development of representative species of all four genera suggests that the ancestral flower of the tribe was unspecialized, with successively initiated whorls of separate sepals, petals, antisepalous stamens, and antipetalous stamens; at its inception, the gynoecium had a united, half-inferior ovary and separate stigmatic lobes. This developmental pattern is found in Rhizophora mangle (wind-pollinated) and Ceriops decandra (insect-pollinated). In Kandelia, all floral organs distal to the sepals are initiated simultaneously, and there has apparently been an evolutionary amplification in the number of stamens to about six times the number of petals. Explosive pollen release evolved independently in C. tagal and in Bruguiera. In the former, all stamens belong to one whorl and arise simultaneously upon a very weakly differentiated androecial ring primordium. In Bruguiera, the androecial ring is pronounced, and two whorls of stamens arise upon it; the primordia of the antisepalous whorl arise first but are closer to the center of the apex than the antipetalous stamen primordia. The antisepalous stamens bend toward and are enclosed by the petals early in development. In all genera, the inferior ovary develops by zonal growth of receptacular tissue; additional intercalary growth above the placenta occurs in Bruguiera. In general, floral specialization is accompanied by an increase in the width of the floral apex compared to the size of the primordia, increasing fusion of the stylar primordia, and decreasing prominence of the superior portion of the ovary. Apparent specializations of petal appendages for water storage, including the presence of sub-terminal hydathodes (previously unreported in any angiosperm), were found in two species in which flowers remain open during the day but were absent from two species normally pollinated at night or at dawn. Distinctive tribal characteristics that may aid in phylogenetic analysis include the mode of development of the inferior ovary; the aristate, bifid, usually fringed petals that individually enclose one or more stamens; the intrastaminal floral disc; and the initially subepidermal laticiferous cell layer in the sepals and ovary.  相似文献

The substitution of alanines for the two cysteines which form thioether linkages to the haem group in cytochrome c(552) from Hydogenobacter thermophilus destabilises the native protein fold. The holo form of this variant slowly converts into a partially folded apo state that over prolonged periods of time aggregates into fibrillar structures. Characterisation of these structures by electron microscopy and thioflavin-T binding assays shows that they are amyloid fibrils. The data demonstrate that when the native state of this cytochrome is destabilised by loss of haem, even this highly alpha-helical protein can form beta-sheet structures of the type most commonly associated with protein deposition diseases.  相似文献

Plant-based expression systems are attractive for the large-scale production of pharmaceutical proteins. However, glycoproteins require particular attention as inherent differences in the N-glycosylation pathways of plants and mammals result in the production of glycoproteins bearing core-xylose and core-alpha(1,3)-fucose glyco-epitopes. For treatments requiring large quantities of repeatedly administered glycoproteins, the immunological properties of these non-mammalian glycans are a concern. Recombinant glycoproteins could be retained within the endoplasmic reticulum (ER) to prevent such glycan modifications occurring in the late Golgi compartment. Therefore, we analysed cPIPP, a mouse/human chimeric IgG1 antibody binding to the beta-subunit of human chorionic gonadotropin (hCG), fused to a C-terminal KDEL sequence, to investigate the efficiency of ER retrieval and the consequences in terms of N-glycosylation. The KDEL-tagged cPIPP antibody was expressed in transgenic tobacco plants or Agrobacterium-infiltrated tobacco and winter cherry leaves. N-Glycan analysis showed that the resulting plantibodies contained only high-mannose (Man)-type Man-6 to Man-9 oligosaccharides. In contrast, the cPIPP antibody lacking the KDEL sequence was found to carry complex N-glycans containing core-xylose and core-alpha(1,3)-fucose, thereby demonstrating the secretion competence of the antibody. Furthermore, fusion of KDEL to the diabody derivative of PIPP, which contains an N-glycosylation site within the heavy chain variable domain, also resulted in a molecule lacking complex glycans. The complete absence of xylose and fucose residues clearly shows that the KDEL-mediated ER retrieval of cPIPP or its diabody derivative is efficient in preventing the formation of non-mammalian complex oligosaccharides.  相似文献

The functional relationships and properties of different subtypes of dendritic cells (DC) remain largely undefined. To better characterize these cells, we used global gene analysis to determine gene expression patterns among murine CD11c(high) DC subsets. CD4(+), CD8alpha(+), and CD8alpha(-) CD4(-) (double negative (DN)) DC were purified from spleens of normal C57/BL6 mice and analyzed using Affymetrix microarrays. The CD4(+) and CD8alpha(+) DC subsets showed distinct basal expression profiles differing by >200 individual genes. These included known DC subset markers as well as previously unrecognized, differentially expressed CD Ags such as CD1d, CD5, CD22, and CD72. Flow cytometric analysis confirmed differential expression in nine of nine cases, thereby validating the microarray analysis. Interestingly, the microarray expression profiles for DN cells strongly resembled those of CD4(+) DC, differing from them by <25 genes. This suggests that CD4(+) and DN DC are closely related phylogenetically, whereas CD8alpha(+) DC represent a more distant lineage, supporting the historical distinction between CD8alpha(+) and CD8alpha(-) DC. However, staining patterns revealed that in contrast to CD4(+) DC, the DN subset is heterogeneous and comprises at least two subpopulations. Gene Ontology and literature mining analyses of genes expressed differentially among DC subsets indicated strong associations with immune response parameters as well as cell differentiation and signaling. Such associations offer clues to possible unique functions of the CD11c(high) DC subsets that to date have been difficult to define as rigid distinctions.  相似文献

Carnitine palmitoyltransferase 1A (CPT1A) is the key regulatory enzyme of hepatic long-chain fatty acid beta-oxidation. Human CPT1A deficiency is characterized by recurrent attacks of hypoketotic hypoglycemia. We presently analyzed at both the functional and structural levels five missense mutations identified in three CPT1A-deficient patients, namely A275T, A414V, Y498C, G709E, and G710E. Heterologous expression in Saccharomyces cerevisiae permitted to validate them as disease-causing mutations. To gain further insights into their deleterious effects, we localized these mutated residues into a three-dimensional structure model of the human CPT1A created from the crystal structure of the mouse carnitine acetyltransferase. This study demonstrated for the first time that disease-causing CPT1A mutations can be divided into two categories depending on whether they affect directly (functional determinant) or indirectly the active site of the enzyme (structural determinant). Mutations A275T, A414V, and Y498C, which exhibit decreased catalytic efficiency, clearly belong to the second class. They are located more than 20 A away from the active site and mostly affect the stability of the protein itself and/or of the enzyme-substrate complex. By contrast, mutations G709E and G710E, which abolish CPT1A activity, belong to the first category. They affect Gly residues that are essential not only for the structure of the hydrophobic core in the catalytic site, but also for the chain-length specificity of CPT isoforms. This study provides novel insights into the functionality of CPT1A that may contribute to the design of drugs for the treatment of lipid disorders.  相似文献
