Nonenzymatic glycation of biomacromolecules occurs due to the diabetes mellitus and ageing. A number of small molecules, known
as chemical chaperones, stabilize protein conformation against thermal and chemically induced denaturation. These compounds
are including: polyamines (e.g. spermine and spermidine), amino acids (e.g. lysine) and polyols (e.g. glycerol). In this study
the effect of spermidine (Spd), spermine (Spm), and glycerol on glycation, structure and function of lysozyme (LZ), as an
extra-cellular protein, by different techniques is investigated. LZ is incubated with or without glucose (50 or 100 mM) in
the absence or presence of Spd/Spm/glycerol at 37 °C up to 16 weeks. All the observed changes of glycated-LZ in comparison
with the native protein, including: increased fluorescence emission, alteration in the secondary and tertiary structure, and
reduced electrophoretic mobility- indicate its structural changes that are accompanied with its reduced activity. Glucose
in the presence or absence of Spd induces the protein dimerization, but glucose plus Spm induces its trimmerization. In contrast,
glycerol inhibits the LZ glycation and prevents the large changes on its structure and function. Glucose binds lysine residues,
decreases the protein positive charges and induces some alterations in its structure and activity. Polyamines also directly
bind to LZ, increase its positive charges and hence induce more glycation; more conformational changes, oligomerization and
its inactivation in the presence of glucose, but glycerol affect the protein environment and preserve protein from these harmful

Ailanthus altissima or Ailanthus glandulosa (Simaroubaceae) is known as tree of heaven. It is a dioecious plant with staminate and pistillate flowers that grow on separate trees. In recent years, A. altissima has been frequently planted in numerous areas, especially in arid and semiarid lands of Iran and also used as an ornamental tree in several Iran cities including Kerman. The aim of this research was to identify IgE-binding proteins responsible for type I hypersensitivities of A. altissima pollen extract. In this study, pollen’s proteins were extracted and analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Total protein content of pollen extracts was measured by Bradford assay. Allergenicity of pollen extract was evaluated by skin test and immunoblotting. Results showed that total protein concentration of A. altissima pollen was found to be 1.34 μg μl?1. In A. altissima pollen extract, different protein fractions were identified by SDS-PAGE mostly at 42, 53.7, 63, 87.7, 100 and 120 kDa. Skin tests showed a delayed hypersensitivity. Immunoblotting of A. altissima pollen extract with pooled subject’s sera detected a major IgE-binding component of 42 kDa. Moreover, these results will provide a platform for cloning cDNA encoding allergenic protein from A. altissima pollen. 相似文献

Viruses play diverse and important roles in ecosystems. In recent years, trade-offs between host and virus traits have gained increasing attention in viral ecology and evolution. However, microbial organism traits, and viral population parameters in particular, are challenging to monitor. Mathematical and individual-based models are useful tools for predicting virus-host dynamics. We have developed an individual-based evolutionary model to study ecological interactions and evolution between bacteria and viruses, with emphasis on the impacts of trade-offs between competitive and defensive host traits on bacteria-phage population dynamics and trait diversification. Host dynamics are validated with lab results for different initial virus to host ratios (VHR). We show that trade-off based, as opposed to random bacteria-virus interactions, result in biologically plausible evolutionary outcomes, thus highlighting the importance of trade-offs in shaping biodiversity. The effects of nutrient concentration and other environmental and organismal parameters on the virus-host dynamics are also investigated. Despite its simplicity, our model serves as a powerful tool to study bacteria-phage interactions and mechanisms for evolutionary diversification under various environmental conditions. 相似文献

Crocus sativus L., mostly famous as saffron, has gained more attention due to its crocin (crocetin ester) pigment responsible for its extensive pharmaceutical properties. In this study, we established two different callus cultures from corm and style explants of saffron to find out the best explant as a suitable source for crocin production. Comparative analyses of total phenolic, flavonoid, carotenoid and anthocyanin contents were also performed in the two callus cultures. For callus induction, different combinations of MS medium with name thidiazuron (TDZ), benzylaminopurine (BA), 1-naphthaleneacetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D) alone or in combination were tested. Of the used media, all the combinations containing TDZ and NAA gave 100% callus induction. HPLC-DAD and HPLC–ESI-MS analysis were used for identification of crocin esters in established callus cultures. The highest amount of 0.35 mg g?1 DW crocin was detected in style originated calli grown on the medium containing 3 mg L?1 NAA?+?1 mg L?1 TDZ while the corm calli showed the most abundant total carotenoid (0.73 mg g?1 DW), phenolic (15.04 mg gallic acid equivalent g?1 DW) and flavonoid (0.76 mg rutin equivalent g?1 DW) contents. In general, style-derived calli showed longer time survival with a fine texture and good quality compared to corm-derived calli. 相似文献

Colletotrichum truncatum conidia produced from a one week-old culture in a liquid semi-defined medium with a C:N ratio of 5:1 were more tolerant of desiccation than those harvested from two or three week-old cultures. Conidia washed with 20% (w/v) sucrose germinated better than unwashed conidia or those washed in 10% (w/v) sucrose, 10 and 20% (w/v) glucose or fructose, 0.1% (w/v) soluble starch, 0.9% (w/v) NaCl or deionized water. Washing with sucrose (20% w/v) also resulted in significantly longer germ tubes than those produced by unwashed conidia or conidia washed with deionized water or NaCl (0.9% w/v). Conidia washed twice in sucrose showed greater desiccation tolerance during storage at 15% relative humidity (RH) and 15°C than at 30% RH and 15 or 25°C or at 15% RH and 25, 5 or -10°C. 相似文献
