Chromosome numbers and allozyme variations were surveyed in 74 polyploid populations ofTaraxacum hondoense, in northern Honshu, Japan. Most of the populations (94.4%) consisted of triploid (2n=24), indicating the predominance of this ploidy level. Approximately 42.6% were found to contain tetraploid (2n=32), and a few plants were pentaploid (2n=40). Electrophoretic analysis at6 Pgdh-1 revealed twelve phenotypes with four alleles (including one putative null allele). The triploids showed excessive heterozygosity (82.4%) and all of the tetraploids and pentaploids were heterozygote. Phenotype IV was the most frequent and widely distributed in northern Honshu. Forty five percent of the populations were found to contain multiple phenotypes at 6Pgdh-1. A total of 21 clones were distinguished using three polymorphic loci (6Pgdh-1, Got andMdh), and a considerable amount of clonal diversity was detected both within and among polyploid populations ofT. hondoense. Factors causing multiclonality in agamospermous polyploids are discussed.  相似文献

Zebrafish was an annual species, with the main period of reproduction commencing just before the onset of the monsoon season. Brain stem noradrenergic cell groups mediating autonomic responses to stress project to airway-related vagal preganglionic neurons (AVPNs). In ferrets, their activation produces withdrawal of cholinergic outflow to the airways via release of norepinephrine and activation of alpha(2A)-adrenergic receptors (alpha(2A)-AR) expressed by AVPNs. In these studies, we examined the effects of allergen exposure of the airway (AE) with ovalbumin on noradrenergic transmission regulating the activity of AVPNs and, consequently, airway smooth muscle tone. Experiments were performed in vehicle control (Con) and AE ferrets. Microperfusion of an alpha(2A)-AR agonist (guanabenz) in close proximity to AVPNs elicited more pronounced effects in Con than AE ferrets, including a decrease in unit activity and reflexly evoked responses of putative AVPN neurons with a corresponding decrease in cholinergic outflow to the airways. Although no differences were found in the extent of noradrenergic innervation of the AVPNs, RT-PCR and Western blot studies demonstrated that AE and repeated exposure to antigen significantly reduced expression of alpha(2A)-ARs at message and protein levels. These findings indicate that, in an animal model of allergic asthma, sensitization and repeated challenges with a specific allergen diminish central inhibitory noradrenergic modulation of AVPNs, possibly via downregulation of alpha(2A)-AR expression by these neurons.  相似文献

Recent advances in hematopoietic stem cells (HSCs) expansion by growth factors including angiopoietin-like proteins (Angptls) have opened up the possibility to use HSCs in regenerative medicine. However, the unavailability of true in vitro HSCs expansion by these growth factors has limited the understanding of the cellular and molecular mechanism of HSCs expansion. Here, we report the functional role of mouse Angptls 1, 2, 3, 4, 6 and 7 and growth factors SCF, TPO, IGF-2 and FGF-1 on purified mouse bone-marrow (BM) Lineage-Sca-1+(Lin-Sca-1+) HSCs. The recombinant retroviral transduced- CHO-S cells that secrete Angptls in serum-free medium were used alone or in combination with growth factors (SCF, TPO, IGF-2 and FGF-1). None of the Angptls stimulated HSC proliferation, enhanced or inhibited HSCs colony formation, but they did support the survival of HSCs. By contrast, any of the six Angptls together with saturating levels of growth factors dramatically stimulated a 3- to 4.5-fold net expansion of HSCs compared to stimulation with a combination of those growth factors alone. These findings lead to an understanding of the basic function of Angptls on signaling pathways for the survival as well as expansion of HSCs in the bone marrow niche.  相似文献

The human relaxin family comprises seven peptide hormones with various biological functions mediated through interactions with G-protein-coupled receptors. Interestingly, among the hitherto characterized receptors there is no absolute selectivity toward their primary ligand. The most striking example of this is the relaxin family ancestor, relaxin-3, which is an agonist for three of the four currently known relaxin receptors: GPCR135, GPCR142, and LGR7. Relaxin-3 and its endogenous receptor GPCR135 are both expressed predominantly in the brain and have been linked to regulation of stress and feeding. However, to fully understand the role of relaxin-3 in neurological signaling, the development of selective GPCR135 agonists and antagonists for in vivo studies is crucial. Recent reports have demonstrated that such selective ligands can be achieved by making chimeric peptides comprising the relaxin-3 B-chain combined with the INSL5 A-chain. To obtain structural insights into the consequences of combining A- and B-chains from different relaxins we have determined the NMR solution structure of a human relaxin-3/INSL5 chimeric peptide. The structure reveals that the INSL5 A-chain adopts a conformation similar to the relaxin-3 A-chain, and thus has the ability to structurally support a native-like conformation of the relaxin-3 B-chain. These findings suggest that the decrease in activity at the LGR7 receptor seen for this peptide is a result of the removal of a secondary LGR7 binding site present in the relaxin-3 A-chain, rather than conformational changes in the primary B-chain receptor binding site.  相似文献

The relaxin peptides are a family of hormones that share a structural fold characterized by two chains, A and B, that are cross-braced by three disulfide bonds. Relaxins signal through two different classes of G-protein-coupled receptors (GPCRs), leucine-rich repeat-containing GPCRs LGR7 and LGR8 together with GPCR135 and GPCR142, now referred to as the relaxin family peptide (RXFP) receptors 1-4, respectively. Although key binding residues have been identified in the B-chain of the relaxin peptides, the role of the A-chain in their activity is currently unknown. A recent study showed that INSL3 can be truncated at the N terminus of its A-chain by up to 9 residues without affecting the binding affinity to its receptor RXFP2 while becoming a high affinity antagonist. This suggests that the N terminus of the INSL3 A-chain contains residues essential for RXFP2 activation. In this study, we have synthesized A-chain truncated human relaxin-2 and -3 (H2 and H3) relaxin peptides, characterized their structure by both CD and NMR spectroscopy, and tested their binding and cAMP activities on RXFP1, RXFP2, and RXFP3. In stark contrast to INSL3, A-chain-truncated H2 relaxin peptides lost RXFP1 and RXFP2 binding affinity and concurrently cAMP-stimulatory activity. H3 relaxin A-chain-truncated peptides displayed similar properties on RXFP1, highlighting a similar binding mechanism for H2 and H3 relaxin. In contrast, A-chain-truncated H3 relaxin peptides showed identical activity on RXFP3, highlighting that the B-chain is the sole determinant of the H3 relaxin-RXFP3 interaction. Our results provide new insights into the action of relaxins and demonstrate that the role of the A-chain for relaxin activity is both peptide- and receptor-dependent.  相似文献

Rinderpest is an acute and highly contagious viral disease of ruminants, often resulting in greater than 90% mortality. We have constructed a recombinant vaccinia virus vaccine (v2RVFH) that expresses both the fusion (F) and hemagglutinin (H) genes of rinderpest virus (RPV) under strong synthetic vaccinia virus promoters. v2RVFH-infected cells express high levels of the F and H glycoproteins and show extensive syncytium formation. Cattle vaccinated intramuscularly with as little as 10(3) PFU of v2RVFH and challenged 1 month later with a lethal dose of RPV were completely protected from clinical disease; the 50% protective dose was determined to be 10(2) PFU. Animals vaccinated with v2RVFH did not develop pock lesions and did not transmit the recombinant vaccinia virus to contact animals. Intramuscular vaccination of cattle with 10(8) PFU of v2RVFH provided long-term sterilizing immunity against rinderpest. In addition to being highly safe and efficacious, v2RVFH is a heat-stable, inexpensive, and easily administered vaccine that allows the serological differentiation between vaccinated and naturally infected animals. Consequently, mass vaccination of cattle with v2RVFH could eradicate rinderpest.  相似文献

Intestinal development in amniotes is driven by interactions between progenitor cells derived from the three primary germ layers. Genetic analyses and gene targeting experiments in zebrafish offer a novel approach to dissect such interactions at a molecular level. Here we show that intestinal anatomy and architecture in zebrafish closely resembles the anatomy and architecture of the mammalian small intestine. The zebrafish intestine is regionalized and the various segments can be identified by epithelial markers whose expression is already segregated at the onset of intestinal differentiation. Differentiation of cells derived from the three primary germ layers begins more or less contemporaneously, and is preceded by a stage in which there is rapid cell proliferation and maturation of epithelial cell polarization. Analysis of zebrafish mutants with altered epithelial survival reveals that seemingly related single gene defects have different effects on epithelial differentiation and smooth muscle and enteric nervous system development.  相似文献

Water-use efficiency (WUE) of Leptochloa fusca (L.) Kunth (Kallar grass) and Sporobolus arabicus Boiss. was determined under different soil moisture regimes. Plants grown in lysimeters were subjected to three soil moisture regimes, viz. well-watered (100%), medium-watered (75%), and low-watered (50%) of total available water (TAW). The soil moisture was restored on alternate days by adding the required volume of water on the basis of neutron moisture meter readings taken from neutron access tubes installed in each lysimeter. The grasses were harvested after suitable intervals (4 months) to obtain maximum biomass. Leaf samples collected at each harvest were analyzed for carbon-isotope discrimination (13C) with an isotope ratio (13C/12C) mass spectrometer. Results indicated significant differences in WUE of both grasses subjected to different water regimes. Sporobolus arabicus showed higher WUE than Kallar grass. However, Kallar grass showed better value of yield response factor (k y = 0.649) compared with Sporobolus (k y = 1.06) over the entire season. The data confirm that these grasses can be grown successfully in water-limited environments by selecting an optimum soil moisture level for maximum biomass production. The mean carbon-isotope discrimination (13C) of Kallar grass (–14.4) and Sporobolus (–12.8) confirm that both are C4 plants. The carbon-isotope discrimination () was significantly and negatively correlated with WUE of the two species studied. The results of the present study confirm that 13C or of leaves can be used as good predictor of WUE in some C4 plants.  相似文献

A field experiment was conducted to evaluate the effectiveness of growing salt tolerant plants to improve the physical characteristics of a saline-sodic soil. Kallar grass [Leptochloa fusca (L.) Kunth], a species tolerant to salinity, sodicity and alkalinity, was irrigated for five years with poor quality ground water (EC = 0.14 S m–1, SARadj=19.3, RSC = 9.7 meq L–1). The soil physical properties of plant available water, saturated hydraulic conductivity, structural stability, bulk density and porosity were determined at the end of each year. The growth of kallar grass for three years significantly improved the physical properties of the soil and these were maintained with further growth of grass up to five years. Kallar grass significantly increased plant available water with time (r=0.97**). The available water was highly correlated (r=0.92**) with increases in soil organic matter content, porosity (r=0.99**) and other physical properties. Soil hydraulic conductivity increased substantially with time from 0.035 to 55.6 mm d–1 in the topsoil (0–20 cm) in five years and was significantly correlated with porosity, water retention, structural stability and organic matter content of soil. The soil structural stability index improved significantly from 32 to 151 with kallar grass and showed greater increases in the surface soil than at depth. The cropping of kallar grass resulted in a linear increase of soil organic matter content (r=0.92**) which improved porosity and other soil physical properties (r0.82*). This study confirmed that kallar grass is effective for rehabilitation and restoration of soil fertility in saline-sodic areas on a sustainable basis.  相似文献
