The present study was designed to elucidate the outcome of subchronic co-administration of black tea and nicotine on cardiovascular performance and whether these substances could modulate the isoproterenol-induced cardiac injury. Animal groups were control, black tea, nicotine and black tea plus nicotine. Test groups received nicotine (2 mg/kg s.c.) and black tea brewed (p.o.) each alone and in combination for 4 weeks. On the 28th day, myocardial damage was induced by isoproterenol (50 mg/kg i.p.), and blood samples were taken. On day 29, after hemodynamic parameters recording, hearts were removed for histopathological evaluation. Tea or nicotine consumption had no significant effects on hemodynamic indices of animals without heart damage. When the cardiac injury was induced, tea consumption maintained the maximum dp/dt, and nicotine significantly decreased the pressure–rate product. Moreover, severity of heart lesions was lower in the presence of nicotine or black tea. Concomitant use of these materials did not show extra effects on mentioned parameters more than the effect of each of them alone. The results suggest that subchronic administration of black tea or nicotine for a period of 4 weeks may have a mild cardioprotective effect, while concomitant use of these materials cannot intensify this beneficial effect.  相似文献

Objective

To gain insights on the degree of heterogeneity and kinetic differences of streptokinase (SK) from group G (SKG) Streptococci compared with standard SK from group C (SKC) and identification of potentially contributing critical residues (hotspots).

Results

DNA and sequencing analyses confirmed the proper construction of all SK encoding vectors (two SKGs and one standard SKC). SDS-PAGE and western blot analyses confirmed the expression and proper purification of the recombinant SKs from E.coli with the expected size of 47 kDa. Kinetic analyses of two SKGs, compared with SKC, showed higher levels of specific [(×103 IU/mg) of 725 and 715 vs. 536] and fibrin-dependent proteolytic activities [Kcat/KM (min?1/µM) of 37 and 30 vs. 23], accompanied by declined fibrin-independent amidolytic activities [Kcat/KM (min?1/mM) of 109 and 84 vs. 113], respectively. Sequence alignments identified 10 novel residual substitutions scattered in SKα (I33F, R45Q, SKG132, A47D, and G55 N), SKβ (N228 K, F287I), and SKγ domains (L335 V, V396A, T403S) of SKGs, as potential hotspots.

Conclusion

The residue substitutions identified might critically contribute as hot spots to different kinetic parameters of SKGs and might assist in further elucidation of structure/function relations and rational design of SKs with improved (fibrin-dependent) therapeutic properties.

Chromosomal Studies in Infertile Men   总被引:1,自引:0,他引:1  
Prometaphase and metaphase chromosome analyses performed on 70 consecutive men with primary infertility (for a period of at least 2 years) revealed 8 (11.42%) men with some kind of chromosomal abnormality. The highest frequency of abnormal karyotypes (10%) was found among patients with azoospermia and the most frequent anomaly was 47, XXY chromosomal constitution, found in 6 (8.57%) patients. All the chromosomal aberrations found in this study was sex chromosomal type and we did not find any autosomal aberration. All patients with numerical chromosomal anomalies had azoospermia. The incidence of structural aberration in our study was 1.42%. Fifteen patients had different chromosomal variants (21.38%). We suggest that men with azoospermia should be considered for cytogenetic investigation and we report that variants of the Y chromosome have no influence on the sperm count (million/ml) and fertility of men.  相似文献

Dendritic cells (DC) have been successfully used in clinical pilot studies to induce tumor-specific immunity as well as clinical response in selected patients. However, DC-based immunotherapy remains a challenge and several parameters need to be examined in order to optimize the induction of anti-tumor immune responses. This study focuses on DC vaccination for leukemia and evaluates the in vitro efficacy of three different strategies for generating antigen-loaded DC-based vaccines for the induction of major histocompatibility complex (MHC) class I-restricted anti-leukemia cytotoxic T lymphocyte (CTL) responses. These included direct fusion of DC with leukemia cells to generate DC-leukemia cell hybrids, and DC pulsed with either apoptotic leukemia cell fragments or whole tumor cell lysates. Using either the U937 cell line or primary human acute myeloid leukemia blasts (AML), DC-leukemia cell hybrids were found to be the most potent in vitro inducers of CTL activity. DC pulsed with apoptotic tumor cell fragments were less efficient, but induced a more potent CTL response compared to tumor lysate-pulsed DC. The CTL responses were both MHC class I-restricted and antigen-specific, as shown by the inability of the CTL to lyse other control targets. The data presented here suggest that the method of antigen loading onto DC may be critical in the design of tumor vaccines.  相似文献

BACKGROUND: The therapeutic potential of retroviruses can be significantly enhanced by display of specific molecules on the retroviral surface. This has been conventionally achieved by the manipulation of retroviral envelope proteins. In this report we have tested whether the natural budding mechanism of the retrovirus could be exploited to incorporate a specific molecule into the retroviral surface. METHODS: Retroviral packaging cells were engineered to express the membrane-bound form of human stem cell factor (mbSCF). Surface expression of mbSCF on retroviral packaging cells was confirmed by immunofluorescence and flow cytometry. Incorporation of mbSCF into retroviral particles was demonstrated by virus-binding assay and immunomagnetic capture of virus using antibody to SCF. Retroviral supernatants were tested for activity of the incorporated cytokine by proliferation assays on factor-dependent cells. Amphotropic retrovirus displaying surface mbSCF was used to transduce SCF receptor-positive haematopoietic cells. RESULTS: Retroviruses incorporating surface SCF showed increased levels of binding to cells (MO7e) expressing the SCF receptor, c-kit. mbSCF displayed on the viral surface retained levels of biological activity comparable with those of soluble recombinant growth factor. Transduction of c-kit-positive target cells with viruses displaying mbSCF showed enhanced levels of transduction in comparison with unmodified viruses. CONCLUSIONS: Expression of the membrane-bound form of human stem cell factor (mbSCF) on the surface of retroviral packaging cells allows its efficient incorporation into retrovirus particles in a biologically active form, opening up the possibility for the use of retroviral display in many therapeutic areas, such as in gene therapy, drug delivery and in the development of novel vaccines.  相似文献

This study set out to build a model identifying areas where a positive Net Present Value (NPV) could be obtained from date palm (Phoenix dactylifera) using CLIMEX and six parameters including (a) suitable soil taxonomy and physicochemical soil properties, (b) slopes of less than 10°, (c) land uses suitable for date palm cultivation, (d) availability of roads, (e) availability of water, and (f) low risk of the lethal disease caused by Fusarium oxysporum f. spp. in the years 2030, 2050, 2070, and 2100 in Iran. Here, we utilized the A2 scenario and two global climate models (GCMs): CSIRO-Mk3.0 (CS) and MIROC-H (MR). Economic feasibility was estimated based on the assumption that the decision to plant date palms by landholders is motivated by a desire to maximize their return to land. Our results indicate that only 5450 km2 of southern Iran will be highly profitable for cultivation of date palm, with NPV > 10,000, while profitable (with NPV between 4200 and 10,000) and moderately profitable (with NPV between 0 and 4200) areas would cover only 500 and 50 km2, respectively, in future. A comparison of mean outputs from the two chosen GCMs and those of the economic and CLIMEX output combination indicates that only about 0.01% of areas from both GCMs will be highly economically viable for cultivation of date palm. In this study we ensure that the predictions become robust, rather than producing hypothetical findings, limited purely to publication.  相似文献

We have studied the synthesis of poly(ADP-ribose) by nuclei isolated from Xenopuslaevis embryos at different stages of development. Determination of the total chain length of poly(ADP-ribose) molecules by hydroxylapatite column chromatography generally gave higher values than when the radioactive portions of these molecules, synthesized invitro, were measured by poly(ethyleneimine)-cellulose thin layer chromatography, after snake venom phosphodiesterase digestion. The results show that most of the poly(ADP-ribose) synthesized invitro is a covalent elongation of molecules previously initiated invivo.  相似文献

Using CLIMEX and the Taguchi Method, a process-based niche model was developed to estimate potential distributions of Phoenix dactylifera L. (date palm), an economically important crop in many counties. Development of the model was based on both its native and invasive distribution and validation was carried out in terms of its extensive distribution in Iran. To identify model parameters having greatest influence on distribution of date palm, a sensitivity analysis was carried out. Changes in suitability were established by mapping of regions where the estimated distribution changed with parameter alterations. This facilitated the assessment of certain areas in Iran where parameter modifications impacted the most, particularly in relation to suitable and highly suitable locations. Parameter sensitivities were also evaluated by the calculation of area changes within the suitable and highly suitable categories. The low temperature limit (DV2), high temperature limit (DV3), upper optimal temperature (SM2) and high soil moisture limit (SM3) had the greatest impact on sensitivity, while other parameters showed relatively less sensitivity or were insensitive to change. For an accurate fit in species distribution models, highly sensitive parameters require more extensive research and data collection methods. Results of this study demonstrate a more cost effective method for developing date palm distribution models, an integral element in species management, and may prove useful for streamlining requirements for data collection in potential distribution modeling for other species as well.  相似文献

Light-induced activation of the LOV2-Jα domain of the photoreceptor phototropin from oat is believed to involve the detachment of the Jα helix from the central β-sheet and its subsequent unfolding. The dynamics of these conformational changes were monitored by time-resolved emission spectroscopy with 100 ns time resolution. Three transitions were detected during the LOV2-Jα photocycle with time constants of 3.4 μs, 500 μs, and 4.3 ms. The fastest transition is due to the decay of the flavin phosphorescence in the transition of the triplet LOV(L)(660) state to the singlet LOV(S)(390) signaling state. The 500 μs and 4.3 ms transitions are due to changes in tryptophan fluorescence and may be associated with the dissociation and unfolding of the Jα helix, respectively. They are absent in the transient absorption signal of the flavin chromophore. The tryptophan fluorescence signal monitors structural changes outside the chromophore binding pocket and indicates that there are at least three LOV(S)(390) intermediates. Since the 500 μs and 4.3 ms components are absent in a construct without the Jα helix and in the mutant W557S, the fluorescence signal is mainly due to tryptophan 557. The kinetics of the main 500 μs component is strongly temperature dependent with activation energy of 18.2 kcal/mol suggesting its association with a major structural change. In the structurally related PAS domain protein PYP the N-terminal cap dissociates from the central β-sheet and unfolds upon signaling state formation with a similar time constant of ~1 ms. Using transient fluorescence we obtained a nearly identical activation energy of 18.5 kcal/mol for this transition.  相似文献
