There is a growing body of data reporting the association of genetic alterations in chromosome 9P21 with the risk of developing cancer. In the current study, we studied the association of a genetic variant in CDKN2A/B, rs1333049, with the risk of developing breast cancer. A total of 339 participants with and without breast cancer entered to the study. Genotyping was done by the TaqMan real-time polymerase chain reaction (RT-PCR) method and gene expression analysis was ran by RT-PCR. Our data showed that the minor allele homozygote in the total population was 10%, whereas for heterozygote was 38%. The dominant genetic model demonstrated that individuals with breast cancer had advanced TNM classification. Moreover, the logistic regression revealed that individuals who had CC/CG genotypes might have an enhanced risk of developing breast cancer when compared to the holders of GG genotype (e.g., OR = 2.8; 95% CI,1.4–5.4; p = .001), after regulated for confounders; age and body mass index. Furthermore, our analysis showed that the CDKN2A/B gene was downregulated in patients (p < .001). We showed a meaningful relationship of CDKN2A/B with the risk of breast cancer, cancer, showing the importance of studies in great sample size and several centers for studying the value of the marker as a risk classification in the management of patients with breast cancer.  相似文献

Nitrogenase, the enzyme system responsible for biological nitrogen fixation, is believed to utilize two unique metalloclusters in catalysis. There is considerable interest in understanding how these metalloclusters are assembled in vivo. It has been presumed that immature iron-molybdenum cofactor-deficient nitrogenase MoFe proteins contain the P-cluster, although no biosynthetic pathway for the assembly of this complex cluster has been identified as yet. Through the comparison by iron K-edge x-ray absorption edge and extended fine structure analyses of cofactor-deficient MoFe proteins resulting from nifH and nifB deletion strains of Azotobacter vinelandii, a novel [Fe-S] cluster is identified in the DeltanifH MoFe protein. The iron-iron scattering displayed by the DeltanifH MoFe protein is more similar to that of a standard [Fe(4)S(4)]-containing protein than that of the DeltanifB MoFe protein, which is shown to contain a "normal" P-cluster. The iron-sulfur scattering of the DeltanifH MoFe protein, however, indicates differences in its cluster from an [Fe(4)S(4)](Cys)(4) site that may be consistent with the presence of either oxygenic or nitrogenic ligation. Based on these results, models for the [Fe-S] center in the DeltanifH MoFe protein are constructed, the most likely of which consist of two separate [Fe(4)S(4)] sites, each with some non-cysteinyl coordination. This type of model suggests that the P-cluster is formed by the condensation of two [Fe(4)S(4)] fragments, possibly concomitant with Fe protein (NifH)-induced conformational change.  相似文献

Providing “real blank sample” is a problem in determination of endogenous steroids in complex matrices. A new quantification strategy is proposed in the present study, which is based on using isotope-labeled steroids instead of natural steroids for constructing calibration line. This approach is called surrogate analyte and it is shown that its accuracy is better than some of the previously described methods at low concentrations and comparable to standard addition method at medium and high concentration levels. The method was fully validated to satisfy the ICH criteria and it was applied for determination of endogenous steroids in several urine samples.  相似文献

Adherence of Helicobacter pylori to inflamed gastric mucosa is dependent on the sialic acid-binding adhesin (SabA) and cognate sialylated/fucosylated glycans on the host cell surface. By in situ hybridization, H. pylori bacteria were observed in close association with erythrocytes in capillaries and post-capillary venules of the lamina propria of gastric mucosa in both infected humans and Rhesus monkeys. In vivo adherence of H. pylori to erythrocytes may require molecular mechanisms similar to the sialic acid-dependent in vitro agglutination of erythrocytes (i.e., sialic acid-dependent hemagglutination). In this context, the SabA adhesin was identified as the sialic acid-dependent hemagglutinin based on sialidase-sensitive hemagglutination, binding assays with sialylated glycoconjugates, and analysis of a series of isogenic sabA deletion mutants. The topographic presentation of binding sites for SabA on the erythrocyte membrane was mapped to gangliosides with extended core chains. However, receptor mapping revealed that the NeuAcalpha2-3Gal-disaccharide constitutes the minimal sialylated binding epitope required for SabA binding. Furthermore, clinical isolates demonstrated polymorphism in sialyl binding and complementation analysis of sabA mutants demonstrated that polymorphism in sialyl binding is an inherent property of the SabA protein itself. Gastric inflammation is associated with periodic changes in the composition of mucosal sialylation patterns. We suggest that dynamic adaptation in sialyl-binding properties during persistent infection specializes H. pylori both for individual variation in mucosal glycosylation and tropism for local areas of inflamed and/or dysplastic tissue.  相似文献

Yellow rust caused by Puccinia striiformis f. sp. hordei is an important disease of barley (Hordeum vulgare L.) in some parts of the world. We compared the effectiveness of different types of resistance in field plots at Ardabil Agricultural Research Station (Iran) during 2010–2011. Yield components along with slow rusting parameters including final rust severity (FRS), apparent infection rate (r), relative area under disease progress curve (rAUDPC) and coefficient of infection (CI) were evaluated for 25 barley cultivars. In all, two barley cultivars with race-specific resistance, 19 cultivars with different levels of slow rusting resistance and four susceptible cultivars were included in two experiments with and without fungicide protection under high disease pressure. Barley cultivars with slow rusting resistance displayed a range of severity responses indicating phenotypic diversity. Mean thousand kernels weight (TKW) losses for susceptible, race-specific and slow rusting genotypes were 31, 3 and 12%, respectively, and mean kernels per spike (KPS) losses for susceptible, race-specific and slow rusting genotypes were 19, 0.2 and 8%, respectively. Correlation coefficient of mean TKW and KPS losses with epidemiological parameters; rAUDPC, r, CI and FRS were highly significant. Slow rusting cultivars with low values of different parameters as well as genotypes with low yield component losses despite moderate disease levels were identified. Such genotypes can be used for breeding barely genotypes with high levels of resistance and negligible yield losses.  相似文献

This article draws on Islamic perspectives that conceive of visions (ru'yā) and nightmares (kābūs) as instances whereby Jamilā, a 13-year-old Syrian refugee girl in Brooklyn, New York, imagines and makes sense of the dead. She uses the Arabic words ma‘rwf (familiar) and gharīb (strange) to describe her dead elderly neighbour, Safiyya, in her visions and nightmares. Jamilā’s familiar-strange experiences imagine the dead both as good (the divine nature of death fundamental to Islamic values) and as evil (the pain and suffering of the departed). These familiar-strange feelings constitute an irresolvable uncertainty about death, shaping an imaginative space for Jamilā to be with Safiyya, who was like a mother to her. The manifestation of the dead consists of dynamic interpretations for the living. This results in a familiar-strange experience granted through the in-between space of what the philosopher Ibn ‘Arabī calls barzakh (‘obstacle’ or ‘separation’), which allows for an understanding of the departed as both blissful and suffering. The vernacular Arabic used by Jamilā in describing her familiar-strange experiences and the Islamic values upheld by her family underscore the importance of knowledge accumulated in the Islamic tradition for understanding the perception of loss and death for an adolescent Syrian refugee.  相似文献
