Cationic amino acids, arginine and lysine partition differentially from water into aqueous micellar sodium dodecanoate. Conversely, partitioning of serine, glycine, aspartic acid, glutamic acid, threonine, alanine, proline, valine, leucine, phenylalanine and isoleucine do not vary appreciably. Partitioning from neat hexane into dodecylammonium propionate trapped water in hexane is, however, dependent upon both electrostatic and hydrophobic interactions. These results imply that the interior of dedecylammonium propionate aggregates is negatively charged and is capable of hydrogen bonding in addition to providing a hydrophobic enviroment. The solubilities of amino acids in neat hexane substantiate the previously derived amino acid hydrophobicity scale. Relevance of partitioning in these systems to the postulated selective amino acid compartmentalization is discussed.  相似文献

Spectral similarity is used as a proxy for structural similarity in many tandem mass spectrometry (MS/MS) based metabolomics analyses such as library matching and molecular networking. Although weaknesses in the relationship between spectral similarity scores and the true structural similarities have been described, little development of alternative scores has been undertaken. Here, we introduce Spec2Vec, a novel spectral similarity score inspired by a natural language processing algorithm—Word2Vec. Spec2Vec learns fragmental relationships within a large set of spectral data to derive abstract spectral embeddings that can be used to assess spectral similarities. Using data derived from GNPS MS/MS libraries including spectra for nearly 13,000 unique molecules, we show how Spec2Vec scores correlate better with structural similarity than cosine-based scores. We demonstrate the advantages of Spec2Vec in library matching and molecular networking. Spec2Vec is computationally more scalable allowing structural analogue searches in large databases within seconds.  相似文献

Nannospalax is a genus of blind rodents adapted to living in underground. The species have numerous chromosomal forms in Turkey, and their taxonomic position is still unknown. In this study, 15 mole rats of four different 2n = 50 forms were used; C- and G- banding processes were applied; and a comparison was made accordingly. Karyological results showed that the 2n = 50S form is a new form for Turkish blind mole rats. 2n = 50S form is determined from Andirin (Kahramanmara?) and has NF = 70. The 2n = 50W form, on the other hand, differs from the others with NF = 74 form. C-banding results showed that heterochromatin blocks of all 2n = 50 are different, while only the 2n = 50W form has telomeric heterochromatin blocks. G-banding results, however, displayed homologies and differences among the chromosomal forms. After comparison, we determined that Robertsonian fusion is an efficient force on chromosomal evolution in blind mole rats in Turkey, and that telomeric heterochromatin is a distinctive character for the 2n = 50W form. We suggest that the chromosomal changing mechanism should be independent from climatic peculiarities. These results support the theory that ancestral karyotype should have the largest distribution in a chromosomally variable species.  相似文献

A three-layer artificial neural network (ANN) was constructed to predict the removal efficiency of Lanaset Red (LR) G on Chara contraria based on 2304 experimental sets. The effects of operating variables (particle size, adsorbent dosage, pH regimes, dye concentration, and contact time) were studied to optimize the sorption conditions of this dye. The operating variables were used as the input to the constructed neural network to predict the dye uptake at any time as the output. This adsorbent was characterized by FTIR. Pseudo second-order model was also fitted to the experimental data. According to values of error analyses and determinations coefficient, the ANN was more appropriate to describe this adsorption process. Result of this model indicated that pH regimes had the highest importance effect (49%) on the dye uptake.  相似文献

Antibiotic resistant hospital acquired infections are on the rise, creating an urgent need for novel bactericidal drugs. Enzymes involved in lipopolysaccharide (LPS) biosynthesis are attractive antibacterial targets since LPS is the major structural component of the outer membrane of Gram-negative bacteria. Lipid A is an essential hydrophobic anchor of LPS and the first committed step in lipid A biosynthesis is catalyzed by a unique zinc dependent metalloamidase, UDP-3-O-(R-3-hydroxymyristoyl)-N-acetylglucosamine deacetylase (LpxC). LpxC is an attractive Gram-negative only target that has been chemically validated by potent bactericidal hydroxamate inhibitors that work by coordination of the enzyme’s catalytic zinc ion. An exploratory chemistry effort focused on expanding the SAR around hydroxamic acid zinc-binding ‘warheads’ lead to the identification of novel compounds with enzyme potency and antibacterial activity similar to CHIR-090.  相似文献

Cellular adhesion molecules might be good markers in some types of malignant tumors, thus providing useful information in diagnosis and prognosis. The objective of this study was to determine the clinical significance of the serum levels of epithelial cell adhesion molecule (EPCAM) in lung cancer patients. One hundred and thirty lung cancer patients were enrolled in this study. Serum EPCAM levels were determined by the solid-phase sandwich ELISA method. Age- and sex-matched 34 healthy controls were included in the analysis. The median age was 58 years, ranging 35–80 years. The majority of the patients had NSCLC (83.8 %) and stage IV disease (60.8 %). There was no significant difference in the serum EPCAM levels between lung cancer patients and healthy controls (p = 0.16). Moreover, known clinical variables including age of patient, gender, histology, stage of disease, and response to chemotherapy were not found to be correlated with serum EPCAM concentrations (p > 0.05). Similarly, no prognostic role was found for outcome (1-year survival rate 62 vs. 65.1 %, p = 0.89). In conclusion, serum EPCAM concentrations have no diagnostic, predictive, and prognostic roles in lung cancer patients.  相似文献

Traditional microbiological methods are dependent on the growth of microorganisms, and hence require prolonged periods. The methods used to detect resistance in Staphylococcus aureus should have high sensitivity and specificity, yet provide results in a timely manner. The aim of this study was to evaluate the use of Quicolor (QC) ES® agar for the rapid detection of resistance in S. aureus. We evaluated 100 clinical S. aureus isolates. Resistance detection was performed using traditional microbiological methods. Methicillin resistance detection was evaluated using traditional and molecular microbiological methods. Traditional antibiotic susceptibility testing methods, such as disc diffusion, were conducted using QC ES and Mueller–Hinton (MH) media. The plates were incubated at 36 °C for 5, 6 and 24 h. Rapid results obtained using QC ES agar after 5 h of incubation were consistent with those using the overnight procedure with MH agar for 83 of the 100 S. aureus (including methicillin-susceptible S. aureus) strains. However, the correlation for oxacillin between MH (24 h) and QC ES (5 h) was not satisfactory (r = 0.770). The total agreement between QC ES and MH agar was 83 % after 5 h, 89 % after 6 h, and 94 % after 24 h. The accurate and rapid detection of resistance in S. aureus is critical due to the associated therapeutic problems and infection control measures. We believe that the use of QC ES for S. aureus will reduce the delay in resistance detection, thus providing physicians and infection control practitioners with early information for better management.  相似文献

In epithelial and stem cells, lethal giant larvae (Lgl) is a potent tumour suppressor, a regulator of Notch signalling, and a mediator of cell fate via asymmetric cell division. Recent evidence suggests that the function of Lgl is conserved in mammalian haematopoietic stem cells and implies a contribution to haematological malignancies. To date, direct measurement of the effect of Lgl expression on malignancies of the haematopoietic lineage has not been tested. In Lgl1−/− mice, we analysed the development of haematopoietic malignancies either alone, or in the presence of common oncogenic lesions. We show that in the absence of Lgl1, production of mature white blood cell lineages and long-term survival of mice are not affected. Additionally, loss of Lgl1 does not alter leukaemia driven by constitutive Notch, c-Myc or Jak2 signalling. These results suggest that the role of Lgl1 in the haematopoietic lineage might be restricted to specific co-operating mutations and a limited number of cellular contexts.  相似文献

Poly(lactic acid) (PLA) was foamed with an endothermic chemical foaming agent (CFA) through an extrusion process. The effects of polymer melt flow index, CFA content, and processing speed on the cellular structures, void fraction, and cell-population density of foamed PLA were investigated. The apparent melt viscosity of PLA was measured to understand the effect of melt index on the cell morphology of foamed PLA samples. The void fraction was strongly dependent on the PLA melt index. It increased with increasing melt index, reaching a maximum value, after which it decreased. Melt index showed no significant effect on the cell-population density of foamed samples within the narrow range studied. A gas containment limit was observed in PLA foamed with CFA. Both the void fraction and cell-population density increased with an initial increase in CFA content, reached a maximum value, and then decreased as CFA content continued to increase. The processing speed also affected the morphology of PLA foams. The void fraction reached a maximum value as the extruder’s screw speed increased to 40 rpm and a further increase in the processing speed tended to reduce the void fraction of foamed samples. By contrast, cell-population density increased one order of magnitude by increasing the screw speed from 20 to 120 rpm. The experimental results indicate that a homogeneous and finer cellular morphology could be successfully achieved in PLA foamed in an extrusion process with a proper combination of polymer melt flow index, CFA content, and processing speed.  相似文献
