Ensuring adequate food production is a major issue in the context of an increasing human population, limit to the areas of new land that can be cultivated, and loss of existing cultivated lands to abiotic stresses. Of these stresses, salinity consistently has the greatest impact in reducing the area of cultivated land, often due to inappropriate irrigation techniques. To increase food supply, there is a need to produce salt-tolerant crops, which can grow successfully on salt-affected lands. Among crops, vegetables possess a central position in the human diet because of their nutritional value providing vitamins, carbohydrates, proteins, and mineral nutrients. There are many vegetable crops of local importance around the world but others that are very widely cultivated. All of these vegetable crops are affected by salinity more or less severely. Salinity affects every aspect of vegetable crop development including their morphology, physiological function and yield. Although efforts have been made to understand the mechanisms of salt tolerance in vegetable crops, less attention has been paid to these than to the staple crops. Where attempts have been made to improve salt tolerance of vegetables, the strategies have ranged from exogenous application of fertilizers, compatible solutes or plant growth regulators, to use of advanced molecular techniques for genetic modifications. This review focuses on the responses of pea, okra, tomato, eggplant, pepper, carrot, broccoli, cauliflower, and potato to salt stress and the strategies being used to enhance their salt tolerance.  相似文献

The Harboi rangeland, Kalat, Pakistan, covers an area of 22 351 hectares and it lies between 29°N and 66°45' to 67°E. It was declared as Protected Forest since January 1967 (Fig.1). CD8+ T cells play a crucial role against chronic viral infections, however, their effector functions are influenced by the expression of co-stimulatory/inhibitory receptors. For example, CD73 works with CD39 to convert highly inflammatory ATP to adenosine. However, its expression on T cells in the context of viral infections has not been well defined. Here, we analyzed the expression of CD73 on human T cells in a cohort of 102 HIV-infected individuals including those on antiretroviral therapy (ART), ART-naïve, and long-term non-progressors who were not on ART. We found that the frequency of CD73+ T cells was markedly lower among T cell subsets (e.g. naïve, effector or memory) in the peripheral blood of all HIV-infected individuals. Notably, CD73 was decreased at the cell surface, intracellular and gene levels. Functionally, CD8+CD73+ T cells exhibited decreased cytokine expression (TNF-α, IFN-γ and IL-2) upon global or antigen-specific stimulation and impaired expression of cytolytic molecules at the gene and protein levels. In contrast, CD8+CD73+ T cells expressed elevated levels of homing receptors such as CCR7, α4β7 integrin, which suggests a migratory advantage for these cells as observed in vitro. We also observed significant migration of CD73+CD8+ T cells into the cerebrospinal fluids of multiple sclerosis (MS) patients at the time of disease relapse. Moreover, we found that elevated levels of ATP in the plasma of HIV-infected individuals upregulates the expression of miRNA30b-e in T cells in vitro. In turn, inhibition of miRNAs (30b, 30c and 30e) resulted in significant upregulation of CD73 mRNA in CD8+ T cells. Therefore, we provide a novel mechanism for the downregulation of CD73 via ATP-induced upregulation of miRNA30b, 30c and 30e in HIV infection. Finally, these observations imply that ATP-mediated downregulation of CD73 mainly occurs via its receptor, P2X1/P2RX1. Our results may in part explain why HIV-infected individuals have reduced risk of developing MS considering the role of CD73 for efficient T cell entry into the central nervous system.  相似文献

Twenty-five genotypes of early CIMMYT hexaploid wheat were screened for salt tolerance in a glasshouse experiment using photosynthetic capacity and water relation parameters as selection criteria. Under salt stress (150 mM NaCl) the genotypes Frontana, Norin-10, Mayo-54, Noreste-66, and Yaktana-54 excelled all other lines in shoot dry mass, and Na(20)TPP, Penjamo-62, Inia-66, Frontana, Siete Cerros, and Jaral-66 in grain yield per plant in both absolute and relative (percent of control) terms. Although net photosynthetic rate (P N) declined in all genotypes due to salt stress, it was not helpful in discriminating among genotypes according to salt tolerance. Similarly, no positive relationships of salt tolerance of the genotypes with stomatal conductance, leaf water potential, or turgor pressure were found. Every genotype used its own specific mechanism to tolerate salt stress. However, a large amount of variation in salt tolerance observed in 25 early CIMMYT wheat genotypes can be of considerable practical value for improving salt tolerance in the existing commercial hexaploid wheats. Abstract

Plant extracts and streptomycin sulfate were evaluated against Xanthomonas axonopodis pv. citri. In first experiment, Azadirachta indica, Allium cepa, Catharanthus roseus, Allium sativum, Zingiber officinale (Roscoe) were investigated in vitro through inhibition zone technique against the growth of X. a. pv. citri. Results indicated that A. indica exhibited statistically significant inhibition (4?cm) zone over control. In second experiment, A. indica and streptomycin sulfate disjointedly and in amalgamation were evaluated in vitro. Streptomycin alone and in permutation with A. indica articulated significant inhibition of the bacterium. In third study, streptomycin sulfate and A. indica (S) and in combination were evaluated against citrus canker disease in green house. Results showed that streptomycin sulfate reduced disease significantly than control. In fourth experiment, streptomycin sulfate, A. indica, in combination and their interaction with days were evaluated under field condition. Streptomycin sulfate proved to be most effective and reduced the disease severity as compared to control.  相似文献

The objective of the present work was to express a truncated form of Pseudomonas putida PutA that shows proline dehydrogenase (ProDH) activity. The putA gene encoding ProDH enzyme was cloned into pET23a vector and expressed in Escherichia coli strain BL-21 (DE3) plysS. The recombinant P. putida enzyme was biochemically characterized and its three dimensional structure was also predicted. ProDH encoding sequence showed an open reading frame of 1,035-bp encoding a 345 amino acid residues polypeptide chain. Purified His-tagged enzyme gave a single band with a molecular mass of 40 kDa on SDS-PAGE. The molecular mass of the isolated enzyme was found to be about 40 kDa by gel filtration. This suggested that the enzyme of interest consists of one subunit. The K m and V max values of recombinant P. putida ProDH were estimated to be 31 mM and 132 μmol/min, respectively. The optimum pH and temperature for the catalytic activity of the enzyme was about pH 8.5 and 30 °C. The modeling analysis of the three dimensional structure elucidated that Ser-165, Lys-195 and Ala-252 were key residues for the ProDH activity. This study provides data on the cloning, sequencing and recombinant expression of PutA ProDH domain from P. putida POS-F84.  相似文献

This study evaluates the role of exogenous foliar application of 5-aminolevulinic acid (ALA) on water relations, gas exchange, chlorophyll fluorescence, and the activities and gene expression patterns of antioxidant enzymes in leaves of oilseed rape under drought stress and recovery conditions. Seedlings at four-leaf stage were imposed to well-watered condition (80 % of water-holding capacity) or drought stress (40 % of water-holding capacity) and subsequently foliar sprayed with water or ALA (30 mg l?1). Drought suppressed the accumulation of plant biomass and decreased chlorophyll content and leaf water status (relative water content and water potential). The actual quantum yield of photosystem II and electron transport rates were hampered in parallel to net photosynthetic rate. However, drought stress induced the accumulation of malondialdehyde (MDA) and hydrogen peroxide, enhanced the activities of catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), glutathione reductase (GR) and superoxide dismutase and up-regulated the expression of APX and GR. After rehydration for 4 days, the growth of drought-treated seedlings was restored to normal level for most of the physiological parameters. Foliar application of ALA maintained relatively higher leaf water status and enhanced chlorophyll content, net photosynthetic rate, actual quantum yield of photosystem II, photochemical quenching, non-photochemical quenching and electron transport rates in stressed leaves. Exogenous ALA also alleviated the accumulation of MDA and hydrogen peroxide, increased the activities of antioxidant enzymes and enhanced the expression of CAT and POD in drought-treated plants. These results indicate that ALA may effectively protect rapeseed seedlings from damage induced by drought stress.  相似文献

This study presents the partitioning and purification of recombinant Bacillus badius phenylalanine dehydrogenase (PheDH) in aqueous two-phase systems (ATPS) composed of polyethylene glycol 6000 (PEG-6000) and ammonium sulfate. A single-step operation of ATPS was developed for extraction and purification of recombinant PheDH from E. coli BL21 (DE3). The influence of system parameters including; PEG molecular weight and concentration, pH, (NH(4))(2)SO(4) concentration and NaCl salt addition on enzyme partitioning were investigated. The best optimal system for the partitioning and purification of PheDH was 8.5% (w/w) PEG-6000, 17.5% (w/w) (NH(4))(2)SO(4) and 13% (w/w) NaCl at pH 8.0. The partition coefficient, recovery, yield, purification factor and specific activity values were of 92.57, 141%, 95.85%, 474.3 and 10424.97 U/mg, respectively. Also the K(m) values for L-phenylalanine and NAD(+) in oxidative deamination were 0.020 and 0.13 mM, respectively. Our data suggested that this ATPS could be an economical and attractive technology for large-scale purification of recombinant PheDH.  相似文献

BackgroundEvidence suggests that recurrent nocturnal hypoxemia may affect pain response and/or the sensitivity to opioid analgesia. We tested the hypothesis that nocturnal hypoxemia, quantified by sleep time spent at an arterial saturation (SaO2) < 90% and minimum nocturnal SaO2 on polysomnography, are associated with decreased pain and reduced opioid consumption during the initial 72 postoperative hours in patients having laparoscopic bariatric surgery.MethodsWith Institutional Review Board approval, we examined the records of all patients who underwent laparoscopic bariatric surgery between 2004 and 2010 and had an available nocturnal polysomnography study. We assessed the relationships between the time-weighted average of pain score and total opioid consumption during the initial 72 postoperative hours, and: (a) the percentage of total sleep time spent at SaO2 < 90%, (b) the minimum nocturnal SaO2, and (c) the number of apnea/hypopnea episodes per hour of sleep. We used multivariable regression models to adjust for both clinical and sleep-related confounders.ResultsTwo hundred eighteen patients were included in the analysis. Percentage of total sleep time spent at SaO2 < 90% was inversely associated with total postoperative opioid consumption; a 5-%- absolute increase in the former would relatively decrease median opioid consumption by 16% (98.75% CI: 2% to 28%, P = 0.006). However, the percentage of total sleep time spent at SaO2 < 90% was not associated with pain. The minimum nocturnal SaO2 was associated neither with total postoperative opioid consumption nor with pain. In addition, neither pain nor total opioid consumption was significantly associated with the number of apnea/hypopnea episodes per hour of sleep.ConclusionsPreoperative nocturnal intermittent hypoxia may enhance sensitivity to opioids.  相似文献
