Numerous iron-sulfur (Fe-S) proteins with diverse functions are present in the matrix and respiratory chain complexes of mitochondria. Although [4Fe-4S] clusters are the most common type of Fe-S cluster in mitochondria, the molecular mechanism of [4Fe-4S] cluster assembly and insertion into target proteins by the mitochondrial iron-sulfur cluster (ISC) maturation system is not well-understood. Here we report a detailed characterization of two late-acting Fe-S cluster-carrier proteins from Arabidopsis thaliana, NFU4 and NFU5. Yeast two-hybrid and bimolecular fluorescence complementation studies demonstrated interaction of both the NFU4 and NFU5 proteins with the ISCA class of Fe-S carrier proteins. Recombinant NFU4 and NFU5 were purified as apo-proteins after expression in Escherichia coli. In vitro Fe-S cluster reconstitution led to the insertion of one [4Fe-4S]2+ cluster per homodimer as determined by UV-visible absorption/CD, resonance Raman and EPR spectroscopy, and analytical studies. Cluster transfer reactions, monitored by UV-visible absorption and CD spectroscopy, showed that a [4Fe-4S]2+ cluster-bound ISCA1a/2 heterodimer is effective in transferring [4Fe-4S]2+ clusters to both NFU4 and NFU5 with negligible back reaction. In addition, [4Fe-4S]2+ cluster-bound ISCA1a/2, NFU4, and NFU5 were all found to be effective [4Fe-4S]2+ cluster donors for maturation of the mitochondrial apo-aconitase 2 as assessed by enzyme activity measurements. The results demonstrate rapid, unidirectional, and quantitative [4Fe-4S]2+ cluster transfer from ISCA1a/2 to NFU4 or NFU5 that further delineates their respective positions in the plant ISC machinery and their contributions to the maturation of client [4Fe-4S] cluster-containing proteins.  相似文献

The aim of this study was to investigate the diurnal variation in core temperature in aluminium shift-workers exposed to hot ambient conditions. Core temperature was continuously recorded via an ingestible radio-telemetry thermistor in 29 shift-workers. Data from the morning, afternoon and night shifts were aggregated for each participant to obtain 24-h recordings during work duties. Complete data were obtained from 10 participants. Results showed that body core temperatures recorded in the afternoon (from 12:00 h to 20:00 h) were significantly higher (P<0.05) than in the late evening, night and early morning (from 21:00 h to 08:00 h). In addition, core temperature displayed a circadian variation with a mesor of 37.45 (±0.19) °C, an amplitude of 0.23 (±0.12) °C and an acrophase at 16:36 h (±3:37 h). The peak values of core temperature recorded at each hour of the day on the work site followed the same pattern with an acrophase in the early afternoon. In summary, our data showed that shift-workers present higher core temperatures in the afternoon than in the morning or during the night. In addition, it was not the work duration but the hour-of-day that triggered the variation in core temperature. This result partly explains previous observations that workers under heat stress have a higher probability of heat illness during daytime shifts than during the night shift, and suggests that special care should be given to the afternoon shift and to the end of the morning shift.  相似文献

Temperature stress (cold, heat) during reproductive development is one of the serious constraints to the productivity of grain legumes as their cultivation is expanding to warmer environments and temperature variability is increasing due to climate change. Grain legumes exposed to temperature below 10-15°C or above 30°C show flower abortion, pollen and ovule infertility, impaired fertilization, and reduced seed filling, leading to substantial reduction in grain yield. For managing these effects of temperature extremes, it is important to improve the resistance of grain legumes by using improved breeding and genetic engineering tools. In this review article, the impact of both high and low temperature stress on different phases of the reproductive stage, from meiosis to grain filling, and the sensitivity of different reproductive organs to temperature extremes are discussed. The review also covers the management options to improve resistance to temperature stress in grain legumes. Furthermore, innovative breeding, genetic and molecular strategies in grain legumes against temperature stress are also discussed.  相似文献

Crude oil is a complex mixture ofseveral different structural classes of compoundsincluding alkanes, aromatics, heterocyclic polarcompounds, and asphaltenes. The rate and extent ofmicrobial degradation of crude oil depends on theinteraction between the physical and biochemicalproperties of the biodegradable compounds and theirinteractions with the non-biodegradable fraction. Inthis study we have systematically altered theconcentration of non-biodegradable material in thecrude oil and analyzed its impact on transport of thebiodegradable components of crude oil to themicroorganisms. We have also developed a mathematicalmodel that explains and accounts for the dependence ofbiodegradation of crude oil through a putativebioavailability parameter. Experimental resultsindicate that as the asphaltene concentration in oilincreases, the maximum oxygen uptake in respirometersdecreases. The mathematically fitted bioavailabilityparameter of degradable components of oil alsodecreases as the asphaltene concentration increases.  相似文献

Malnutrition is considered as major public health concern and is emerging challenge to food and nutrition security particularly in developing countries. Rice is the staple food and consumed by the half of the world's population which is the source of daily requirement of the nutrients. Attempts are being made to fortify rice with micronutrients, but the loss or retention of these micronutrients in different cooking methods is not well studied and documented especially in fortified rice. In the present study, paddy seeds of six Indian varieties were fortified with iron and zinc by parboiling process. Consequently, fortified polished rice had higher micronutrient contents (Fe, 106.31 ± 12.56; Zn, 97.72 ± 9.75) than non-fortified polished rice (Fe, 7.44 ± 1.05; Zn, 14.74 ± 2.94) expressed in ppm. Polished rice of both fortified and non-fortified were cooked under five different cooking conditions and analyzed for remaining iron and zinc content. Cooking rice in rice cooker without prior washing (NRC) retained highest concentration of Fe and Zinc in both fortified and non-fortified rice varieties. It also showed that fortified rice suffered higher percentage loss of micronutrient, than the non-fortified rice. But the average retained micronutrient amount measured in ppm, was higher in fortified rice (Fe, 43.54 ± 6.88; Zn, 36.7 ± 3.12) than in non-fortified rice (Fe, 4.24 ± 0.87; Zn, 9.3 ± 2.11). Hence, adopting appropriate cooking method, higher amount of micronutrients will be retained in the cooked food which will in turn help in combating the malnutrition and improve health.  相似文献

Insulin receptor activity and its relationship with catecholamines in rat young, middle aged and old red blood cells were investigated in short term (4-6) weeks and long term (6-8 months) hyperglycemia and hyperinsulinemia. Loss of insulin receptor activity is linear with cellular ageing and norepinephrine and epinephrine levels increase with age together with levels of glycosylated hemoglobin in control animals and this correlation is altered in hyperglycemia and hyperinsulinemia. These results suggest that loss of insulin receptor in cellular ageing is probably part of a more generalised alteration which is possibly brought about by glycosylation.  相似文献

Maximum decomposition was caused bySporotrichum pruinosum, and maximum humification byStachybotrys atra.
