Orchids of the genus Dendrobium hold a high economical value in the international markets both as pot plants or cut flowers, and for the production of some metabolites with antioxidant and anti-tumoral activities. Manipulating ploidy levels of Dendrobium species is one of the possible methods to develop new varieties with increased ornamental value and a higher production of secondary metabolites. In this work, we present a new and fast flow cytometry approach to obtain and select Dendrobium phalaenopsis?×?Dendrobium loddigesii polyploids, through an early in vitro screening on protocorm like bodies (PLBs) after antimitotic treatment. Our approach allows the identification of the best time of treatment on control PLBs and the assessment of best conditions for polyploid recovery just one month after treatments, by using Cycle Value. We were able to discard about the two-third of the unchanged material by drastically reducing the explants to work with and the corresponding costs. Different conditions, regarding concentrations and exposition time, were tested using colchicine or amiprophos-methyl (APM). A high polyploids recovery, up to 80%, were obtained with both antimitotic agents, and those materials were further characterized by liquid chromatography coupled to high resolution mass spectrometry (LC-HRMS), to identify independent polyploids explants with increased levels in high-value molecules as shihunidine and hircinol, together with stochastic events and genotype-specific metabolite fluctuations. 相似文献

It was assumed that factors different from maximizing energy intake, such as water balance and viscosity of concentrated nectars,

Various conventional methods to estimate the mean and median power spectral frequencies, and amplitude of the surface electromyogram during 30-90 min, cyclic, force-varying, constant-posture contractions were cross-compared in an experimental trial. The aim was to determine the most appropriate algorithm implementations and reduce the total number of algorithms that need to be considered when monitoring time trends. Subjects produced hand-grip contractions in a repeated intermittent pattern until exhaustion. For all estimated parameters: analysis of contraction levels below 25% maximum voluntary contraction produced poor estimates due to high relative measurement noise; parameter reproducibility was best when comparisons were aligned to the actual force produced rather than the target force and when the biomechanics of the contraction were more consistent; and estimates were not greatly influenced by the rate of change of the force trajectory. For frequency parameters: estimates based on the short-time Fourier transform were similar to those based on time-varying autoregressive methods; longer duration analysis windows exhibited better repeatability; and simple frequency-domain noise filters were not effective in reducing the impact of measurement noise. For amplitude estimates: whitening reduced the variance of the amplitude estimate; and the best analysis window duration was a trade-off between bias (decreased with a short duration window) and variance (decreased with a long duration window). 相似文献

The aim of the study was to investigate the correlation between myosin heavy chain (MHC) composition, lactate threshold (LT), maximal oxygen uptake VO2max, and average muscle fiber conduction velocity (MFCV) measured from surface electromyographic (EMG) signals during cycling exercise. Ten healthy male subjects participated in the study. MHC isoforms were identified from a sample of the vastus lateralis muscle and characterized as type I, IIA, and IIX. At least three days after a measure of LT and VO2max, the subjects performed a 2-min cycling exercise at 90 revolutions per minute and power output corresponding to LT, during which surface EMG signals were recorded from the vastus lateralis muscle with an adhesive electrode array. MFCV and instantaneous mean power spectral frequency of the surface EMG were estimated at the maximal instantaneous knee angular speed. Output power corresponding to LT and VO2max were correlated with percentage of MHC I (R2=0.77; and 0.42, respectively; P<0.05). MFCV was positively correlated with percentage of MHC I, power corresponding to LT and to VO2max (R2=0.84; 0.74; 0.53, respectively; P<0.05). Instantaneous mean power spectral frequency was not correlated with any of these variables or with MFCV, thus questioning the use of surface EMG spectral analysis for indirect estimation of MFCV in dynamic contractions. 相似文献

Beta diversity can provide insights into the processes that regulate communities subjected to frequent disturbances, such as flood pulses, which control biodiversity in floodplains. However, little is known about which processes structure beta diversity of amphibians in floodplains. Here, we tested the influence of flood pulses on the richness, composition, and beta diversity of amphibians in Amazonian floodplain environments. We also evaluated indicator species for each environment. We established linear transects in three environments: low várzea, high várzea, and macrophyte rafts. Species richness decreased and beta diversity increased according to the susceptibility of habitats to flood pulses. Indicator species differed among environments according to forest succession promoted by the flood pulse. The decrease in species richness between high and low várzea is due to non‐random extinctions. The higher rates of species turnover between várzeas and macrophyte rafts are driven by the colonization of species adapted to open areas. Our results highlight that the maintenance of complex environments is needed to protect biodiversity in floodplains. 相似文献

Large bowel carcinogenesis involves accumulation of genetic alterations leading to transformation of normal mucosa into dysplasia and, lastly, adenocarcinoma. It is pertinent to elucidate the molecular changes occurring in the pre-neoplastic lesions to facilitate early diagnosis and treatment. Heat shock proteins (Hsps), many of which are molecular chaperones, are implicated in carcinogenesis, and their variations with tumor progression encourage their study as biomarkers. There are many reports on Hsps and cancer but none to our knowledge on their systematic quantification in pre-neoplastic lesions of the large bowel. We performed immunohistochemical determinations of Hsp10, Hsp60, Hsp70, and Hsp90 in biopsies of large bowel tubular adenomas with moderate grade of dysplasia and compared to normal mucosa and adenocarcinoma with a moderate grade of differentiation (G2). A significant elevation of Hsp10 and Hsp60 only, i.e., in the absence of elevation of Hsp70 or Hsp90, in both epithelium and lamina propria was found in tubular adenoma by comparison with normal mucosa. In contrast, adenocarcinoma was characterized by the highest levels of Hsp10 and Hsp60 in epithelium and lamina propria, accompanied by the highest levels of Hsp70 only in epithelium and of Hsp90 only in lamina propria, by comparison with normal and tubular adenoma counterparts. Hsp10 and Hsp60 are promising biomarkers for early diagnosis of tubular adenoma and for its differentiation from more advanced malignant lesions. Hsp10 and Hsp60 may be implicated in carcinogenesis from its very early steps and, thus, are potentially convenient targets for therapy. 相似文献
