MSC display potent suppressive properties initially described a decade ago. More recently, MSC suppressive activities on T-cell effector pathways have been investigated. MSC modulate CD4 differentiation through different mechanisms depending on culture conditions and display disparate activities on T cells according to their differentiation status. A significant amount of evidence for MSC effects on Th17 cells revealed that MSC could be suppressive under diverse circumstances but also enhance Th17 cell activity under other conditions. In the present study, we investigated the suppressive effects of MSC on Th1 and Th17 subsets of T cells using T cells undergoing Th1 and Th17 polarization or mature Th1 and Th17 cells. MSC inhibited the proliferation of T cells during their differentiation toward Th1 cells and mature Th1 cells. This suppressive effect was maintained in a transwell cell culture insert demonstrating the major role played by soluble factors. Using the transwell cell separation barrier, we observed that MSC decrease the number of T cells undergoing Th17 differentiation whereas they did not affect IL-17 production by mature Th17, demonstrating the need for cell contact for suppressing Th17 cell function. Moreover, we reported that PD-L1 is highly expressed on MSC co-cultured with differentiating or polarized Th1 and Th17 cells. Using neutralizing antibodies specific for PD-L1 and PD-1 we showed that the mechanisms by which MSC mediate Th17 cell repolarization depend on PD-L1 expression on MSC. Taken together our results demonstrated a cell-to-cell contact depend mechanism in the selective immunosuppression of MSC on mature Th17 cells through up-regulation of PD-L1.  相似文献

Studying regeneration in the central nervous system (CNS) is hampered by current histological and imaging techniques because they provide only partial information about axonal and glial reactions. Here we developed a tetrahydrofuran-based clearing procedure that renders fixed and unsectioned adult CNS tissue transparent and fully penetrable for optical imaging. In large spinal cord segments, we imaged fluorescently labeled cells by 'ultramicroscopy' and two-photon microscopy without the need for histological sectioning. We found that more than a year after injury growth-competent axons regenerated abundantly through the injury site. A few growth-incompetent axons could also regenerate when they bypassed the lesion. Moreover, we accurately determined quantitative changes of glial cells after spinal cord injury. Thus, clearing CNS tissue enables an unambiguous evaluation of axon regeneration and glial reactions. Our clearing procedure also renders other organs transparent, which makes this approach useful for a large number of preclinical paradigms.  相似文献

A major unanswered question regarding the dispersal of modern humans around the world concerns the geographical site of the first human steps outside of Africa. The "southern coastal route" model predicts that the early stages of the dispersal took place when people crossed the Red Sea to southern Arabia, but genetic evidence has hitherto been tenuous. We have addressed this question by analyzing the three minor west-Eurasian haplogroups, N1, N2, and X. These lineages branch directly from the first non-African founder node, the root of haplogroup N, and coalesce to the time of the first successful movement of modern humans out of Africa, ~60 thousand years (ka) ago. We sequenced complete mtDNA genomes from 85 Southwest Asian samples carrying these haplogroups and compared them with a database of 300 European examples. The results show that these minor haplogroups have a relict distribution that suggests an ancient ancestry within the Arabian Peninsula, and they most likely spread from the Gulf Oasis region toward the Near East and Europe during the pluvial period 55-24 ka ago. This pattern suggests that Arabia was indeed the first staging post in the spread of modern humans around the world.  相似文献

Desmoglein 3 (Dsg3), a member of the desmoglein sub-family, serves as an adhesion molecule in desmosomes. Our previous study showed that overexpression of human Dsg3 in several epithelial lines induces formation of membrane protrusions, a phenotype suggestive of Rho GTPase activation. Here we examined the interaction between Dsg3 and actin in detail and showed that endogenous Dsg3 colocalises and interacts with actin, particularly the junctional actin in a Rac1-dependent manner. Ablation of Rac1 activity by dominant negative Rac1 mutant (N17Rac1) or the Rac1 specific inhibitor (NSC23766) directly disrupts the interaction between Dsg3 and actin. Assembly of the junctional actin at the cell borders is accompanied with enhanced levels of Dsg3, while inhibition of Dsg3 by RNAi results in profound changes in the organisation of actin cytoskeleton. In accordance, overexpression of Dsg3 results in a remarkable increase of Rac1 and Cdc42 activities and to a lesser extent, RhoA. The enhancements in Rho GTPases are accompanied by the pronounced actin-based membrane structures such as lamellipodia and filopodia, enhanced rate of actin turnover and cell polarisation. Together, our results reveal an important novel function for Dsg3 in promoting actin dynamics through regulating Rac1 and Cdc42 activation in epithelial cells.  相似文献

The genus Origanum L. (Lamiaceae) enjoys a socioeconomic interest and constitutes one of the most popular spices. In the literature, the study of this taxon is limited mainly to the chemical composition, antimicrobial, antifungal, and antioxidant activities of the essential oils. In Algeria, few works have been done in this area, including the endemic and rare species: Origanum floribundum Munby . In this study, our contribution highlights the influence of the ecological parameters and of the variation of morphological traits on the chemical composition and the antioxidant activity of the essential oils of six populations. The major oils constituents are carvacrol, γ‐terpinene, and p‐cymene with carvacrol predominance (31.8 – 60.8%) in over five populations (MS1 – MS4 and MS6). However, the population of Ain Terraeur (MS5) of which individuals present a foliar polymorphism, distinguishes itself by p‐cymene predominance (42.6%). This would denote a new chemotype and/or variety which seem well differentiated on the dendrogram of the Ascending Hierarchical Classification (A.H.C) and its foliar morphology. The antioxidant activity of the six samples evaluated by the scavenging activity of free radicals DPPH? and ABTS?+ showed a potent efficiency for the sample with foliar polymorphism population (IC50 = 51.6 ± 0.31 and 12.71 ± 0.37 μg/ml, respectively).  相似文献

AIMS: To determine the efficiency of an electric discharge of the gliding arc type for the destruction of Staphylococcus epidermidis planktonic, adherent and biofilm cells. METHODS AND RESULTS: Bacterial cells were treated in humid air and at atmospheric pressure by a nonthermal quenched plasma of the glidarc type. The kinetics of destruction (followed by plating) were modelled by an Add-inn for Microsoft Excel, GInaFiT. For planktonic cells, log-linear destruction was obtained, whereas biphasic kinetics were observed for sessile cells. An increased resistance of biofilm cells was observed: the reduction of 6 logarithm units of the population was obtained in 15, 30 and 70 min for planktonic, adherent and biofilm cells, respectively. The experiments also show that the cells destruction did not depend on the adhesion surface but was governed by the gap between the target and the plasma source. CONCLUSION: The complete destruction of planktonic, adherent and more resistant biofilm cells of Staph. epidermidis is achieved by a glidarc air plasma at atmospheric pressure. SIGNIFICANCE AND IMPACT OF THE STUDY: The glidarc plasma technology is a promising candidate among the emerging nonthermal techniques for decontamination, as it can destroy even biofilms that are known as particularly resistant to various antimicrobials.  相似文献

Chondrogenesis is a process involving stem-cell differentiation through the coordinated effects of growth/differentiation factors and extracellular matrix (ECM) components. Recently, mesenchymal stem cells (MSCs) were found within the cartilage, which constitutes a specific niche composed of ECM proteins with unique features. Therefore, we hypothesized that the induction of MSC differentiation towards chondrocytes might be induced and/or influenced by molecules from the microenvironment. Using microarray analysis, we previously identified genes that are regulated during MSC differentiation towards chondrocytes. In this study, we wanted to precisely assess the differential expression of genes associated with the microenvironment using a large-scale real-time PCR assay, according to the simultaneous detection of up to 384 mRNAs in one sample. Chondrogenesis of bone-marrow-derived human MSCs was induced by culture in micropellet for various periods of time. Total RNA was extracted and submitted to quantitative RT-PCR. We identified molecules already known to be involved in attachment and cell migration, including syndecans, glypicans, gelsolin, decorin, fibronectin, and type II, IX and XI collagens. Importantly, we detected the expression of molecules that were not previously associated with MSCs or chondrocytes, namely metalloproteases (MMP-7 and MMP-28), molecules of the connective tissue growth factor (CTGF); cef10/cyr61 and nov (CCN) family (CCN3 and CCN4), chemokines and their receptors chemokine CXC motif ligand (CXCL1), Fms-related tyrosine kinase 3 ligand (FlT3L), chemokine CC motif receptor (CCR3 and CCR4), molecules with A Disintegrin And Metalloproteinase domain (ADAM8, ADAM9, ADAM19, ADAM23, A Disintegrin And Metalloproteinase with thrombospondin type 1 motif ADAMTS-4 and ADAMTS-5), cadherins (4 and 13) and integrins (α4, α7 and β5). Our data suggest that crosstalk between ECM components of the microenvironment and MSCs within the cartilage is responsible for the differentiation of MSCs into chondrocytes.  相似文献

This study aimed to characterize the bacterium-destroying properties of a gliding arc plasma device during electric discharges and also under temporal postdischarge conditions (i.e., when the discharge was switched off). This phenomenon was reported for the first time in the literature in the case of the plasma destruction of microorganisms. When cells of a model bacterium, Hafnia alvei, were exposed to electric discharges, followed or not followed by temporal postdischarges, the survival curves exhibited a shoulder and then log-linear decay. These destruction kinetics were modeled using GinaFiT, a freeware tool to assess microbial survival curves, and adjustment parameters were determined. The efficiency of postdischarge treatments was clearly affected by the discharge time (t*); both the shoulder length and the inactivation rate kmax were linearly modified as a function of t*. Nevertheless, all conditions tested (t* ranging from 2 to 5 min) made it possible to achieve an abatement of at least 7 decimal logarithm units. Postdischarge treatment was also efficient against bacteria not subjected to direct discharge, and the disinfecting properties of “plasma-activated water” were dependent on the treatment time for the solution. Water treated with plasma for 2 min achieved a 3.7-decimal-logarithm-unit reduction in 20 min after application to cells, and abatement greater than 7 decimal logarithm units resulted from the same contact time with water activated with plasma for 10 min. These disinfecting properties were maintained during storage of activated water for 30 min. After that, they declined as the storage time increased.  相似文献

The heterogeneity of savanna ecosystems is guaranteed by disturbance events like fires, droughts, floods and browsing and grazing by herbivores. For conservation areas with limited space to preserve biodiversity, fire monitoring is crucial. Long periods of satellite remotely sensed data provide an alternative solution to estimate the distribution of different vegetation types and fire-affected patches over time. This study focusses on the application of MODIS data to detect, identify and delineate fire-affected areas in Kruger National Park (KNP), South Africa, for the period 2001–2003. Fire scars on KNP’s savanna were identified using threshold and supervised classification methods on moderate-resolution imaging spectroradiometer (MODIS) with 500-m resolution and 32-day global composites using a combination of band 1 (red), 2 (NIR, near infrared), 4 (green) and 6 (SWIR, short wave infrared). On identified fire scars, the spectral indexes of albedo, normalised difference infrared index (NDII) and normalised difference vegetation index (NDVI) were extracted. The following four broad habitat types were used for this analysis: riparian woodland, dense woodland, mixed woodland and open-tree savanna. The values of albedo, NDII and NDVI during the dry season (June to October) for different years are lower on fire-affected patches. Mixed woodland is the largest habitat burned with 21%, 43% and 2% of the KNP area affected by fire in 2001, 2002 and 2003, respectively. Riparian woodland is the least affected by fire. The supervised classification method has a greater accuracy for fire scars detection in KNP savannas during the dry season. We conclude that MODIS data can be used successfully for fire monitoring in savanna ecosystems.  相似文献
