The study of functional structure in species assemblages emphasizes the detection of significant guild aggregation patterns. Thus, protocols based on intensive resampling of empirical data have been proposed to assess guild structure. Such protocols obtain the frequency distribution of a given functional similarity metric, and identify a threshold value (often the 95th percentile) beyond which clusters in a functional dendrogram are considered as significant guilds (using one-tailed tests). An alternative approach sequentially searches for significant differences between clusters at decreasing levels of similarity in a dendrogram until one is detected, then assumes that all subsequent nodes should also be significant. Nevertheless, these protocols do not test both the significance and sign of deviations from random at all levels of functional similarity within a dendrogram. Here, we propose a new bootstrapping approach that: (1) overcomes such pitfalls by performing two-tailed tests for each node in a dendrogram of functional similarity after separately determining their respective sample distributions, and (2) enables the quantification of the relative contribution of guild aggregation and functional divergence to the overall functional structure of the entire assemblage. We exemplify this approach by using long-term data on guild dynamics in a vertebrate predator assemblage of central Chile. Prenatal maternal psychological distress increases risk for adverse infant outcomes. However, the biological mechanisms underlying this association remain unclear. Prenatal stress can impact fetal epigenetic regulation that could underlie changes in infant stress responses. It has been suggested that maternal glucocorticoids may mediate this epigenetic effect. We examined this hypothesis by determining the impact of maternal cortisol and depressive symptoms during pregnancy on infant NR3C1 and BDNF DNA methylation. Fifty-seven pregnant women were recruited during the second or third trimester. Participants self-reported depressive symptoms and salivary cortisol samples were collected diurnally and in response to a stressor. Buccal swabs for DNA extraction and DNA methylation analysis were collected from each infant at 2 months of age, and mothers were assessed for postnatal depressive symptoms. Prenatal depressive symptoms significantly predicted increased NR3C1 1F DNA methylation in male infants (β = 2.147, P = 0.044). Prenatal depressive symptoms also significantly predicted decreased BDNF IV DNA methylation in both male and female infants (β = −3.244, P = 0.013). No measure of maternal cortisol during pregnancy predicted infant NR3C1 1F or BDNF promoter IV DNA methylation. Our findings highlight the susceptibility of males to changes in NR3C1 DNA methylation and present novel evidence for altered BDNF IV DNA methylation in response to maternal depression during pregnancy. The lack of association between maternal cortisol and infant DNA methylation suggests that effects of maternal depression may not be mediated directly by glucocorticoids. Future studies should consider other potential mediating mechanisms in the link between maternal mood and infant outcomes. 相似文献

In this paper, the acidic pretreatment of microalgal biomass is investigated, and the solubilized biomass and hydrolyzed sugars were evaluated. The process is analyzed through the severity factor approach (acidic combined severity factor (ACSF)). A suitable kinetic model is developed and applied, and it is shown that the severity factor theory works. A discussion and comparison are presented with respect to the literature methods, which are mainly related to lignocellulosic biomass. In the case of microalgae, reaction orders for biomass and acid are shown to be the main parameters, and no other assumptions are needed. Two regions of the acidic treatment process have to be evaluated: low and high reactivity regions. Furthermore, a suitable experimental design is required in order to provide an appropriate reaction spectrum to obtain a good estimation of the kinetic parameters. A logarithmic severity factor range (ln ACSF) between 5 and 6 is able to solubilize around 80% of biomass and to hydrolyze more than 90% of sugars present in the biomass. 相似文献

Serologic diagnosis has been presented as a safe alternative for coccidioidomycosis. However, commercial kits based on coccidioidal antibodies available in the USA are considered too expensive for laboratories outside that country. In this study, we describe the preparation of antigens for detection of human coccidioidal antibodies by the immunodiffusion test (ID) and enzyme immunoassay (EIA). Antigens were tested against serum samples from patients with coccidioidomycosis, histoplasmosis and paracoccidioidomycosis, as well as healthy individuals. The highest reactivity in the ID tests was seen in the F0-90 antigen. In the EIAs, the best results were obtained with the F60-90 antigen. None of the serum samples from healthy individuals were recognized by any of the antigen extracts tested by ID or EIA. In conclusion, the F0-90 and F60-90 antigens have the potential to be commercially employed in presumptive diagnosis of coccidioidomycosis by ID or EIA, respectively. The criteria to classify the diversity of Neotropical Primates have recently taken the discussion boards due to the reappraisal on the phylogenetic relationship of some groups. Such controversial and arbitrary decisions, however, can hamper conservation actions in as much as it becomes difficult to prioritise and set meaningful targets. It is the case for dwarf marmosets from central Amazonia. Today, the classification of dwarf marmosets in Mico or Callibella genus has not been satisfactorily settled. Aiming to contribute to the taxonomic and conservation assessment of dwarf marmosets, we conducted new data collection during 3‐year fieldwork in the Aripuanã River, where the species was discovered. We present the first phylogenomic analysis of the evolutionary relationships between marmosets, new data from mitochondrial DNA and morphology, as well new records to clarify geographic distribution. With this new evidence, we support dwarf marmosets as the genus Callibella. We further discuss the implications for the conservation of this marmoset. 相似文献

We performed a combined DFT and Monte Carlo 13C NMR chemical-shift study of azadirachtin A, a triterpenoid that acts as a natural insect antifeedant. A conformational search using a Monte Carlo technique based on the RM1 semiempirical method was carried out in order to establish its preferred structure. The B3LYP/6-311++G(d,p), wB97XD/6-311++G(d,p), M06/6-311++G(d,p), M06-2X/6-311++G(d,p), and CAM-B3LYP/6-311++G(d,p) levels of theory were used to predict NMR chemical shifts. A Monte Carlo population-weighted average spectrum was produced based on the predicted Boltzmann contributions. In general, good agreement between experimental and theoretical data was obtained using both methods, and the 13C NMR chemical shifts were predicted highly accurately. The geometry was optimized at the semiempirical level and used to calculate the NMR chemical shifts at the DFT level, and these shifts showed only minor deviations from those obtained following structural optimization at the DFT level, and incurred a much lower computational cost. The theoretical ultraviolet spectrum showed a maximum absorption peak that was mainly contributed by the tiglate group. 相似文献
