Situated at the ventral-most part of the vertebrate neural tube, the floor plate (FP) is an important signalling centre that controls the regional differentiation of neurons in the nervous system. It secretes guidance molecules that direct ventrally navigating axons crucial for the correct wiring of neuronal circuits. Although the function of the FP is well-conserved from fish to humans, discrepancies exists with respect to both the signalling system involved in FP induction, and the origin of the FP in various vertebrate species. Recent findings from the embryos of zebrafish, chicken and mouse provide insights that reconcile previous results and suggest common themes in vertebrate FP specification.  相似文献

The molecular basis governing functional behavior of human hematopoietic stem cells (HSCs) is largely unknown. Here, using in vitro and in vivo assays, we isolate and define progenitors versus repopulating HSCs from multiple stages of human development for global gene expression profiling. Accounting for both the hierarchical relationship between repopulating cells and their progenitors, and the enhanced HSC function unique to early stages of ontogeny, the human homologs of Hairy Enhancer of Split-1 (HES-1) and Hepatocyte Leukemia Factor (HLF) were identified as candidate regulators of HSCs. Transgenic human hematopoietic cells expressing HES-1 or HLF demonstrated enhanced in vivo reconstitution ability that correlated to increased cycling frequency and inhibition of apoptosis, respectively. Our report identifies regulatory factors involved in HSC function that elicit their effect through independent systems, suggesting that a unique orchestration of pathways fundamental to all human cells is capable of controlling stem cell behavior.  相似文献

Kisspeptin (Kp) expression in testis has caused most of the recent research surveying its functional role in this organ. This peptide influences spermatogenesis and sperm capacitation, so it is considered as a regulator of reproduction. Kp roles exert through hypothalamic/pituitary/gonadal axis. We aimed to evaluate direct roles for Kp on proliferation and differentiation of spermatogonial cells (SCs) when the cells are cocultured with somatic cells. Somatic cells and SCs were isolated from adult azoospermic and newborn mice and then enriched using a differential attachment technique. After the evaluation of identity and colonization for SCs, the cells were cocultured with somatic cells, and three doses of Kp (10−8-10−6 M) was assessed on proliferation (through evaluation of MVH and ID4 markers) and differentiation (via evaluation of c-Kit and SCP3, TP1, TP2, and, Prm1 markers) of the coculture system. Investigations were continued for four succeeding weeks. At the end of each level of testosterone in the culture media was also evaluated. We found positive influence from Kp on proliferative and differentiative markers in SCs cocultured with somatic cells. These effects were dose-dependent. There was no effect for Kp on testosterone level. From our findings, we simply conclude that Kp as a neuropeptide for influencing central part of reproductive axis could also positively affect peripheral processes related to spermatogenesis without having an effect on steroidogenesis.  相似文献

Plasma membrane of most cells is not smooth. The surfaces of both small and large micropermeable cells are folded and corrugated which makes mammalian cells to have a larger membrane surface than the supposed ideal mode, that is, the smooth sphere of the same volume. Since cancer is an anthropic disease, cancer cells tend to have a larger membrane area than normal cells. Therefore, cancer cells have higher folding factor and larger radius than normal and healthy cells. On the other hand, the prevalence of breast cancer has prompted researchers to improve the treatment options raised for the disease in the past. In this paper, the impact of folding factor of the cell surface has been investigated. Considering that AFM is one of the most effective tools in performing the tests at micro- and nanoscales, it was used to determine the topography of MCF10 cells and then the resulting images and results were used to experimentally extract the folding factor of cells. By applying this factor in the Hertz, DMT and JKR contact models in the elastic and viscoelastic states, these models have been modified and the simulation of the three models shows that the simulation results are closer to the experimental results by considering the folding in the calculations. Additionally, the simulation of 3D manipulation has been done in both elastic and viscoelastic states with and without consideration of folding. Finally, the results were compared to investigate the effects of folding of the cell surface to the critical force and critical time of sliding and rolling in contact with the substrate and AFM tip in the 3D manipulation model.  相似文献

Abstract: Nuclear poly(ADP-ribose) polymerase levels as well as the DNA strand break levels of whole-brain neuronal and astroglial cells were investigated. Three- and 30-month-old rats were used. Low-molecular-weight neurofilaments and glutamine synthetase served as neuronal and astroglial markers, respectively. A large increase in the poly(ADP-ribose) polymerase activity was observed in the neurons (threefold) and astrocytes (3.7-fold) derived from 30-month-old rats. Similarly, the amount of poly(ADP-ribose) polymerase, evaluated per milligram of DNA, increased ∼3.5-fold in neurons and 3.9-fold in astrocytes prepared from 30-month-old rats. Whether the increase in the poly(ADP-ribose) polymerase activity was due to an enhanced rate of DNA strand break was investigated by determining the rate of DNA unwinding. A significant increase in DNA unwinding rate was detected in the neurons (2.7-fold), although a lower increase was observed in the astroglia (1.3-fold) of aged animals.  相似文献

Human knee specimens were subjected to anterior-posterior, medial-lateral, varus-valgus, and torsional displacement tests. Loads were recorded for the intact joint and for the joint with all soft tissues cut except for the cruciate ligaments. The effect of condylar interference was determined for anterior-posterior, medial-lateral, and torsional displacements. The variation in load with flexion angle was considerable for medial-lateral (0-90-deg flexion) displacements, and less for varus-valgus (0-45-deg flexion) displacements. The cruciates were found to carry almost the entire anterior-posterior load; they carried a significant percentage of the medial-lateral load which varied considerably with flexion angle. A small, but not insignificant percentage of the varus-valgus load was carried by the cruciates and the variations with flexion angle were small. In torsion, the cruciates resisted only internal rotation. In the tested displacement ranges, condylar interference had a small effect on the medial-lateral load but did not affect anterior-posterior or torsional loads.  相似文献

This study investigates the spectral shifting of UV-A radiation, using fluorescent material, as a tool for enhancing Chlorella sp. growth rate in simulated photobioreactors made of UV-stabilized polycarbonate (PC) and poly (methyl methacrylate) (PMMA). The feasibility of using a fluorescent coating as a wavelength shifter layer, to shift UV-A radiation to the PAR range, was explored. For this purpose, a variety of concentrations of fluorescent dye dissolved in thermoplastic acrylic resin were prepared and used to coat PMMA and PC sheets which then were placed between the radiation source and the culture flask. Compared with the uncoated sheets, the panels coated with the wavelength shifter layer exhibited 74% and 45% (for PC and PMMA substrates, respectively) increase in biomass productivity during the same culture period. It was also found that the elimination of UV-A radiation increased chlorophyll a content of the cells.  相似文献

Redox meditators (RMs) are soluble catalysts located in an electrolyte that can improve the energy efficiency (reduced overpotential) and cyclability of Li–oxygen (Li–O2) batteries. In this work, 20 RMs within a Li–O2 system with dimethyl sulfoxide and tetraethylene glycol dimethyl ether electrolytes are studied and their electrochemical features such as redox potential, the separation of cathodic and anodic peaks, and their current intensities are measured using cyclic voltammetry (CV) experiments. Six RMs are selected as “primary” choices based on their electrochemical performance, and stability tests are then performed to examine their electrochemical responses after consecutive cycles. Moreover, galvanostatic cycling tests are performed within a Li–O2 battery system assembled with selected six RMs for real case consistency investigations. It is found that results from CV to galvanostatic cycling tests are consistent for halides and organometallic RMs, where the former exhibit much higher stability. However, the organic RMs show high reversibility in CV but low in battery cycling results. Density functional theory calculations are carried out to gain more understanding of the stability and redox potentials of the RMs. This study provides comparative information to select the most reliable RMs for Li–O2 batteries along with new fundamental understanding of their electrochemical activity and stability.  相似文献

The two main phases of the mammalian cardiac cycle are contraction and relaxation; however, whether there is a connection between them in humans is not well understood. Routine exercise has been shown to improve cardiac function, morphology, and molecular signatures. Likewise, the acute and chronic changes that occur in the heart in response to injury, disease, and stress are well characterized, albeit not fully understood. In this study, we investigated how exercise and myocardial injury affect contraction–relaxation coupling. We retrospectively analyzed the correlation between the maximal speed of contraction and the maximal speed of relaxation of canine myocardium after receiving surgically induced myocardial infarction, followed by either sedentary recovery or exercise training for 10–12 wk. We used isolated right ventricular trabeculae, which were electrically paced at different lengths, frequencies, and with increasing β-adrenoceptor stimulation. In all conditions, contraction and relaxation were linearly correlated, irrespective of injury or training history. Based on these results and the available literature, we posit that contraction–relaxation coupling is a fundamental myocardial property that resides in the structural arrangement of proteins at the level of the sarcomere and that this may be regulated by the actions of cardiac myosin binding protein C (cMyBP-C) on actin and myosin.  相似文献
