Summary The rheological properties of yeast (Baker's yeast) suspensions in water and in water-methanol-electrolyte solutions have been investigated using a Fann V.G. concentric cylinder viscometer. The resulted rheograms could be described by the power law model for non-Newtonian fluids with the flow behaviour index n<1 indicating pseudo-plastic behaviour. This flow behaviour index was constant under all operating conditions while the consistancy index k increased exponentially with increasing wet yeast volume fraction. The addition of methanol and electrolytes had a negligible effect on the rheological properties. Changing the pH of the suspension acidic to lowered the apparent viscosities.  相似文献

The aim of this study is to estimate the total above‐ground biomass (TAGB), stem height (H), diameter at breast height (dbh) and basal area of five tree species (ages 7‐8 years) irrigated by municipal sewage water in the Egyptian‐Chinese friendship forest, Sadat City, Egypt. From the biomass data that obtained through destructive sampling, models for predicting aboveground biomass were developed. The highest values for stem density and height were estimated for Eucalyptus citriodora, while the lowest value for density was obtained for Dalbergia sissoo and stem height for Khaya senegalensis. The highest values for basal area and dbh were obtained for Casuarina spp., while the lowest values were recorded for Dalbergia sissoo. Eucalyptus camaldulensis had the highest stand stem biomass and TAGB (55.5, 83.9 t DW ha‐1, respectively). In addition, Casuarina spp. had the highest leafy branches biomass (32.5 t DW ha‐1) while Dalbergia sissoo had the lowest values for all tree components. All the generated allometric equations had high correlation coefficients at high probability levels. Moreover, the results revealed that not only the dbh data can be used as independent variable for biomass determination, but also stem height and size index are recommended for biomass estimation (© 2012 WILEY‐VCH Verlag GmbH & Co. A novel series of extended DAPI analogues were prepared by insertion of either a carbon–carbon triple bond (16ad) or a phenyl group (21a,b and 24) at position-2. The new amidines were evaluated in vitro against both Trypanosoma brucei rhodesiense (T. b. r.) and Plasmodium falciparum (P. f.). Five compounds (16a, 16b, 16d, 21a, 21b) exhibited IC50 values against T. b. r. of 9 nM or less which is two to nine folds more effective than DAPI. The same five compounds exhibited IC50 values against P. f. of 5.9 nM or less which is comparable to that of DAPI. The fluorescence properties of these new molecules were recorded, however; they do not offer any advantage over those of DAPI.  相似文献

Heterocyclic diamidines are compounds with antiparasitic properties that target the minor groove of kinetoplast DNA. The mechanism of action of these compounds is unknown, but topological changes to DNA structures are likely to be involved. In this study, we have developed a polyacrylamide gel electrophoresis-based screening method to determine topological effects of heterocyclic diamidines on four minor groove target sequences: AAAAA, TTTAA, AAATT and ATATA. The AAAAA and AAATT sequences have the largest intrinsic bend, whereas the TTTAA and ATATA sequences are relatively straight. The changes caused by binding of the compounds are sequence dependent, but generally the topological effects on AAAAA and AAATT are similar as are the effects on TTTAA and ATATA. A total of 13 compounds with a variety of structural differences were evaluated for topological changes to DNA. All compounds decrease the mobility of the ATATA sequence that is consistent with decreased minor groove width and bending of the relatively straight DNA into the minor groove. Similar, but generally smaller, effects are seen with TTTAA. The intrinsically bent AAAAA and AAATT sequences, which have more narrow minor grooves, have smaller mobility changes on binding that are consistent with increased or decreased bending depending on compound structure.  相似文献

We demonstrate ensemble three-dimensional cell cultures and quantitative analysis of angiogenic growth from uniform endothelial monolayers. Our approach combines two key elements: a micro-fluidic assay that enables parallelized angiogenic growth instances subject to common extracellular conditions, and an automated image acquisition and processing scheme enabling high-throughput, unbiased quantification of angiogenic growth. Because of the increased throughput of the assay in comparison to existing three-dimensional morphogenic assays, statistical properties of angiogenic growth can be reliably estimated. We used the assay to evaluate the combined effects of vascular endothelial growth factor (VEGF) and the signaling lipid sphingoshine-1-phosphate (S1P). Our results show the importance of S1P in amplifying the angiogenic response in the presence of VEGF gradients. Furthermore, the application of S1P with VEGF gradients resulted in angiogenic sprouts with higher aspect ratio than S1P with background levels of VEGF, despite reduced total migratory activity. This implies a synergistic effect between the growth factors in promoting angiogenic activity. Finally, the variance in the computed angiogenic metrics (as measured by ensemble standard deviation) was found to increase linearly with the ensemble mean. This finding is consistent with stochastic agent-based mathematical models of angiogenesis that represent angiogenic growth as a series of independent stochastic cell-level decisions.  相似文献

The broad mite Polyphagotarsonemus latus (Banks) was studied for one season (2009/2010) on three pepper cultivars (Travita, Top star and Habeba) in the nethouses in Egypt. The tarsonemid mite was fed upon by the predatory phytoseiid mite Amblyseius swirskii (Athias–Henriot). Population abundance of the tarsonemid pepper prey was affected by climatic conditions and predation. A control measure of one preventive treatment in nursery and two successive curative treatments, applied mid-September, in nethouses seemed to be the most successful management of the harmful mite. The effect of constant different temperatures on P. latus development, reproduction and population growth was investigated. At 11?°C, all activity ceased and by 36?°C the adult began to slow down. Life table parameters varied greatly, especially at 21?°C and 75%?R.H.  相似文献

Integrative approaches to studying the coupled dynamics of skeletal muscles with their loads while under neural control have focused largely on questions pertaining to the postural and dynamical stability of animals and humans. Prior studies have focused on how the central nervous system actively modulates muscle mechanical impedance to generate and stabilize motion and posture. However, the question of whether muscle impedance properties can be neurally modulated to create favorable mechanical energetics, particularly in the context of periodic tasks, remains open. Through muscle stiffness tuning, we hypothesize that a pair of antagonist muscles acting against a common load may produce significantly more power synergistically than individually when impedance matching conditions are met between muscle and load. Since neurally modulated muscle stiffness contributes to the coupled muscle-load stiffness, we further anticipate that power-optimal oscillation frequencies will occur at frequencies greater than the natural frequency of the load. These hypotheses were evaluated computationally by applying optimal control methods to a bilinear muscle model, and also evaluated through in vitro measurements on frog Plantaris longus muscles acting individually and in pairs upon a mass-spring-damper load. We find a 7-fold increase in mechanical power when antagonist muscles act synergistically compared to individually at a frequency higher than the load natural frequency. These observed behaviors are interpreted in the context of resonance tuning and the engineering notion of impedance matching. These findings suggest that the central nervous system can adopt strategies to harness inherent muscle impedance in relation to external loads to attain favorable mechanical energetics.  相似文献

In the present study the optimized parameters for highest ethanol productivity by Kluyveromyces lactis immobilized cells bioreactor were obtained using the method of Lagrange multipliers. Immobilized growing yeast cells in PVA: HEMA (7%: 10%, w/w) hydrogel copolymer carrier produced by radiation polymerization were used in a packed-bed column reactor for the continuous production of ethanol from lactose at different levels of concentrations (50, 100 and 150) gL(-1). The results indicate that volumetric ethanol productivity is influenced by substrate concentration and dilution rate. The highest value 7.17 gL(-1) h(-1) is obtained at higher lactose concentration (150 gL(-1)) in feed medium and 0.3 h(-1) dilution rate. The same results have been obtained through the application of "LINGO" software for mathematical optimization.  相似文献

Chagas disease is caused by infection with the intracellular protozoan parasite Trypanosoma cruzi. At present, nifurtimox and benznidazole, both compounds developed empirically over four decades ago, represent the chemotherapeutic arsenal for treating this highly neglected disease. However, both drugs present variable efficacy depending on the geographical area and the occurrence of natural resistance, and are poorly effective against the later chronic stage. As a part of a search for new therapeutic opportunities to treat chagasic patients, pre-clinical studies were performed to characterize the activity of a novel arylimidamide (AIA--DB1831 (hydrochloride salt) and DB1965 (mesylate salt)) against T. cruzi. These AIAs displayed a high trypanocidal effect in vitro against both relevant forms in mammalian hosts, exhibiting a high selectivity index and a very high efficacy (IC(50) value/48 h of 5-40 nM) against intracellular parasites. DB1965 shows high activity in vivo in acute experimental models (mouse) of T. cruzi, showing a similar effect to benznidazole (Bz) when compared under a scheme of 10 daily consecutive doses with 12.5 mg/kg. Although no parasitological cure was observed after treating with 20 daily consecutive doses, a combined dosage of DB1965 (5 mg/kg) with Bz (50 mg/kg) resulted in parasitaemia clearance and 100% animal survival. In summary, our present data confirmed that aryimidamides represent promising new chemical entities against T. cruzi in therapeutic schemes using the AIA alone or in combination with other drugs, like benznidazole.  相似文献
