AbstractThis study examined how different nitrogen (N) forms and application levels promote plant growth and assist in manganese (Mn) remediation of Polygonum pubescens Blume (P. pubescens) cultured in soil with a high Mn level. The effects of ammonium chloride (a) and urea (u), at three application levels (10, 20, and 30?mg L?1 N) and control (no N addition, CK) on the growth, Mn accumulation, and enzymatic anti-oxidative defenses of P. pubescens were examined. In general, both ammonium-N and urea-N promoted the plant mass and height of P. pubescens. The total Mn amount of roots, stems, and leaves in N treatments were higher (p?<?0.05) than that of CK. The ammonium-N treatments showed greater plant biomass and Mn accumulation compared to the urea-N ones. In general, the accumulations of Mn, Cr, Zn, and Cu were significantly lower (p?<?0.05) in the N fertilizer treatment than those in the control; while the accumulations of Pb were higher (p?<?0.05) in P. pubescens across all N fertilizer treatments than those in the control. The N addition decreased the contents of O2? and H2O2 in the leaves of P. pubescens, while increasing the activities of enzymatic anti-oxidative defenses. 相似文献

采用基因工程技术从发菜总DNA中克隆了一段的基因序列,该序列与基因库中已公布的编码地木耳(Nostoc commnue)超氧化物歧化酶的氨基酸序列同源性为97%。将该基因插入含T7启动子质粒pET-32中构建表达质粒pET-sod,然后将该表达质粒转入大肠杆菌BL21中进行蛋白表达,表达菌株用1 mmol·L-1 IPTG诱导表达数小时后,产生较多的重组的蛋白,且该蛋白以可溶性蛋白形式存在。SDS-PAGE分析表明,在相对分子量约为22 kd的位置有一条明显蛋白质带。将诱导表达后的蛋白通过亲和层析的方法进行蛋白纯化;NBT光还原法测定表达产物的比活力,每毫克纯化蛋白约为2 550 U。对纯化后的蛋白进行高温胁迫研究,将该纯化蛋白在60℃高温下胁迫90 min后,其活性为原(未经胁迫)蛋白活性的85%。 相似文献

Osteoporosis, arthritis, Peget's disease, bone tumor, periprosthetic joint infection, and periprosthetic loosening have a common characteristic of osteolysis, which is characterized by the enhanced osteoclastic bone resorptive function. At present, the treatment target of these diseases is to interfere with osteoclastic formation and function. Scutellarein (Scu), a flavonoids compound, can inhibit the progress of tumor and inflammation. However, the role of Scu in inflammatory osteolysis isn’t elucidated clearly. Our study showed that Scu inhibited bone destruction induced by LPS in vivo and OC morphology and function induced by RANKL in vitro. Mechanistic studies revealed that Scu suppressed osteoclastic marker gene expression by RANKL-induced, such as Ctsk9, Mmp9, Acp5, and Atp6v0d2. In addition, we found that the inhibition effects of osteoclastogenesis and bone resorption function of Scu were mediated via attenuating NF-κB and NFAT signaling pathways. In conclusion, the results showed that Scu may become a potential new drug for the treatment of inflammatory osteolysis. 相似文献

The increased activation of osteoclasts is the major manifestation of several lytic bone diseases, including osteoporosis, rheumatoid arthritis, aseptic loosening of orthopedic implants, Paget disease and malignant bone diseases. One important bone-protective therapy in these diseases focuses on the inhibition of osteoclast differentiation and resorptive function. Given that the deleterious side-effects of currently available drugs, it is beneficial to search for effective and safe medications from natural compounds. Cepharanthine (CEP) is a compound extracted from Stephania japonica and has been found to have antioxidant and anti-inflammatory effects. In this study, we found that CEP inhibited receptor activator of nuclear factor-κB (NF-κB) ligand (RANKL)-induced osteoclast formation and bone-resorbing activities using osteoclastogenesis and bone resorption assay. By polymerase chain reaction, we also found that CEP inhibited the expression of osteoclast-differentiation marker genes including Ctsk, Calcr, Atp6v0d2, Mmp9 and Nfatc1. Mechanistic analyses including Western blot and luciferase reporter assay revealed that CEP inhibited RANKL-induced activation of NF-κB and nuclear factor of activated T-cell, which are essential for the formation of osteoclast. Collectively, these data suggested that CEP can potentially be used as an alternative therapy for preventing or treating osteolytic diseases. 相似文献

Apoptin is a small molecular weight protein encoded by the VP3 gene of chicken anemia virus (CAV). It can induce apoptosis of tumor cells and play anti-tumorigenic functions. In this study, we identified a time-dependent inhibitory role of apoptin on the viability of HCT116 cells. We also demonstrated that apoptin induces pyroptosis through cleaved caspase 3, and with a concomitant cleavage of gasdermin E (GSDME) rather than GSDMD. GSDME knockdown switched the apoptin-induced cell death from pyroptosis to apoptosis in vitro. Furthermore, we demonstrated that the effect of apoptin on GSDME-dependent pyroptosis could be mitigated by caspase-3 and caspase-9 siRNA knockdown. Additionally, apoptin enhanced the intracellular reactive oxygen species (ROS), causing aggregation of the mitochondrial membrane protein Tom20. Moreover, bax and cytochrome c were released to the activating caspase-9, eventually triggering pyroptosis. Therefore, GSDME mediates the apoptin-induced pyroptosis through the mitochondrial apoptotic pathway. Finally, using nude mice xenografted with HCT116 cells, we found that apoptin induces pyroptosis and significantly inhibits tumor growth. A systematic study of the characterization for racemic species of 4-hydroxy-2-pyrrolidone was undertaken. The melting point phase diagram of (R)- and (S)-4-hydroxy-2-pyrrolidone was determined by differential scanning calorimetry. The ternary phase diagram of (R)- and (S)-4-hydroxy-2-pyrrolidone with isopropanol was constructed at 15, 20, 25, and 35 degrees C. The crystalline nature of 4-hydroxy-2-pyrrolidone racemate was also characterized by means of comparison of solid-state FTIR spectra and powder X-ray diffraction patterns of the racemic mixture with those of one of the enantiomers. It is shown that (+/-)-4-hydroxy-2-pyrrolidone is a racemic conglomerate. The enthalpies of fusion of (R)-4-hydroxy-2-pyrrolidone and (+/-)-4-hydroxy-2-pyrrolidone and entropy of mixing of (R)- and (S)-4-hydroxy-2-pyrrolidone were calculated using the thermodynamic data. The solubility and supersolubility diagrams of (R)- and (S)-4-hydroxy-2-pyrrolidone in isopropanol were determined over a temperature range of 4-35 degrees C. The optical resolution of (+/-)-4-hydroxy-2-pyrrolidone was successfully achieved by preferential crystallization. 相似文献

通过田间和网室测定结果,抗小象虫较好的甘薯品种有抗虫1号、台农26、Tis2534、Ricin和鸡蛋黄;抗小象虫较弱的品种有新种花、惠红早、“329”、广薯15等。甘薯品种的营养成份与抗小象虫相关性分析结果表明,抗、感品种与粗纤维、粗脂肪含量无明显相关,与粗淀粉有显负相关,R1=-0.9935,而与粗蛋白和18种氨基酸总量有显正相关,R2=0.9741,R3=0.9621。表现粗淀粉含量高的品种,其虫害指敦较低,抗虫性强;而粗蛋白含量和18种氨基酸总量高的品种,其虫害指数较高,抗虫性则表现较弱。测定分析说明了甘薯品种营养成份与抗虫性存在一定的相关性。 相似文献

We have constructed a 27-kDa hTERT C-terminal polypeptide (hTERTC27) devoid of domains required for telomerase activity and demonstrated that it is capable of nuclear translocation/telomere-end targeting. Here we showed that expression of a low level of hTERTC27 renders hTERT positive HeLa cells sensitive to H(2)O(2)-induced oxidative stress and subsequent cell senescence. The senescence-associated gene, the cyclin/cdk inhibitor p21(Waf1), was up-regulated. This occurs without changing the expression of endogenous hTERT, causing significant telomere shortening or inhibiting telomerase activity. Results from this study suggest for the first time that in addition to telomerase activity, the C-terminus of hTERT also plays a role in hTERT-mediated cellular resistance to oxidative stress. 相似文献
