在全球气候变化的背景下,合理协调低碳减排与经济效益之间的关系,进而指导土地利用优化配置,对于推动经济社会低碳转型与生态系统服务价值的提升具有重要意义。通过耦合多目标规划(MOP)与斑块生成土地利用模拟模型(PLUS),针对北京市构建了自然演变、经济优先与低碳发展这3种未来发展情景,并对不同情景下2030年土地利用与生态系统服务价值进行模拟评估。在此基础上,进一步对北京市未来生态系统服务提升区域进行识别,将传统的多情景土地利用模拟研究进行了延伸与扩展。结果表明:(1)3种情景下的土地利用结构与空间布局存在差异,低碳发展情景下东南部平原区建设用地的扩张与耕地面积的减少趋势得到了有效控制,西北部山区的林地面积有较大幅度的增长。(2)低碳发展情景下的生态系统服务价值总量约为590亿元,高于另外两种情景,且从空间分布上来看,该情景下服务价值提升区域的面积也显著高于其他情景。(3)低碳发展情景下,北京市生态系统服务提升潜力较大的区域面积占比约7.98%、潜力中等区域的面积占比约26.57%,潜力较低区域的面积占比约65.45%。  相似文献

The count and diffusion of Lactobacilli species in the differ ent gastrointestinal tract (GI) regions of broilers were investigated by quantitative realtime polymerase chain reaction, and the probiotic characteristics of six L. reuteri species isolated from broilers' GI tract were also investi gated to obtain the potential target for genetic engineering. Lactobacilli had the highest diversity in the crop and the lowest one in the cecum. Compared with the lower GI tract, more LactobaciUi were found in the upper GI tract. Lactobacillus reuteri, johnsonii, L. acidophilus, L. crispatus, L. salivarius, and L. aviarius were the predominant Lactobacillus species and present throughout the GI tract of chickens. Lactobacillus reuteri was the most abundant Lactobacillus species. Lactobacillus reuteri XC1 had good probiotic characteristics that would be a potential and desirable target for genetic engineering.  相似文献

Bcl-2, a prominent member of the Bcl-2 family proteins, is responsible for the dysregulation of apoptosis and resistance to chemotherapy. In this study, we inves-tigated whether small hairpin RNA (shRNA) targeting at Bcl-2 mRNA could enhance cytarabine (Ara-C)- induced apoptosis in Raji cells. Bcl-2 shRNA was trans-fected into Raji cells and the expression levels of Bcl-2 mRNA and protein were assayed by RT-PCR and immunofluorescence. Cell proliferation was determined by MTT assay. Apoptosis was determined by morphological observation and flow cytometric analysis. Our results show that expression levels of Bcl-2 mRNA and protein from Raji cells transfected with Bcl-2 shRNA decreased, compared with either negative control shRNA group or untransfected cells group (P 〈 0.05). Viability of cells transfected with Bcl-2 shRNA was less than the cells transfected with control shRNA and untransfected Raji cells, respectively (P〈 0.05). Bcl-2 shRNA combined with Ara-C significantly inhibited the growth of cells (P 〈 0.05). There was no difference in cell survival between control shRNA/Ara-C combination and cells treated with Ara-C alone. Using Giemsa staining, cells treated with Bcl-2 shRNA plus Ara-C at 48h displayed changes of apoptosis. Apoptotic rates of Raji cells treated with Bcl-2 shRNA combined with Ara-C significantly increased (P 〈 0.05), compared with either control shRNA/Ara-C combi- nation or Ara-C-treated cells alone. Our results suggest that the shRNA against Bcl-2 mRNA could increase Ara-C-induced apoptosis in Raji cells.  相似文献

We previously identified a matrix protein, MSI7, from pearl oyster Pinctada fucata. According to the structural analysis, the DGD site in the N-terminal of MSI7 is crucial for its role in the shell formation. In this study, we expressed a series of recombinant MSI7 proteins, including the wild-type and several mutants directed at the DGD site, using an Escherichia coli expression system to reveal the structure-function relationship of MSI7. Furthermore, in vitro crystallization, crystallization speed assay, and circular dichroism spectrometry were carried out. Results indicated that wild-type MSI7 could induce the nucleation of aragonite and inhibit the crystallization of calcite. However, none of the mutants could induce the nucleation of aragonite, but all of them could inhibit the crystallization of calcite to some extent. And all the proteins accelerated the crystallization process. Taken together, the results indicated that MSI7 could contribute to aragonite crystallization by inducing the nucleation of aragonite and inhibiting the crystallization of calcite, which agrees with our prediction about its role in the nacreous layer formation of the shell. The DGD site was critical for the induction of the nucleation of aragonite.  相似文献

Androgen receptor (AR) plays a critical role in prostate cancer and male sexual differentiation. We have identified AR from a primitive vertebrate with a sex reversal characteristic, the rice field eel. AR of this species (eAR) is distinct from human AR, especially in the ligand binding domain (LBD), and its expression in gonads shows an increasing tendency during gonadal transformation from ovary via ovotestis to testis. eAR has a restricted androgen-dependent transactivation function after a nuclear translocation upon dihydrotestosterone exposure. A functional nuclear localization signal was further identified in the DNA binding domain and hinge region. Although nuclear export is CRM1-independent, eAR has a novel nuclear export signal, which is negatively charged, indicating that a nuclear export pathway may be mediated by electrostatic interaction. Further, our studies have identified critical sequences for ligand binding in the C terminus. A structure of three α-helices in the LBD has been conserved from eels to humans during vertebrate evolution, despite a distinct amino acid sequence. Mutation analysis confirmed that the LBD is essential for dihydrotestosterone-induced nuclear import of eAR and following transactivation function in the nucleus. In addition, eAR interacts with both Sox9a1 and Sox9a2, and their interaction regulates transactivation of eAR. Our data suggest that the primitive species conserves and especially acquires key novel domains, the nuclear export signal and LBD, for the eAR function in spite of a rapid sequence evolution.  相似文献

In this paper, multi-wall carbon nanotubes (MWNTs)/Pt microparticles nanocomposite was prepared by electrodepositing Pt microparticles onto the MWNTs matrix. The surface of glassy carbon electrode was modified with this kind of nanocomposite for measurement of thiols, such as L-cysteine (L-Cys) and glutathione (GSH). Compared with the MWNTs or Pt microparticles modified electrode, the nanocomposite modified electrode exhibited high sensitivity and good stability for detection of thiols. According to the results of experiments, the peak currents of L-Cys and GSH are linear with their concentrations and the detection limits (S/N=3) are 2.9 x 10(-8) mol/L and 4.5 x 10(-8) mol/L, respectively. Coupled with microdialysis, the method has been successfully applied to the determination of these two thiols in rat striatal microdialysates.  相似文献

An effective procedure for obtaining healthy shoots from nodal segments of Scrophularia yoshimurae is described. Nodal segments cultured on Murashige and Skoog's (MS) basal medium supplemented with 1.0 mg L(-1) benzyladenine (BA) and 0.2 mg L(-1) alpha-naphthaleneacetic acid (NAA) induced multiple shoots in conical flasks that differed in the way they were closed and sealed. Hermitically sealed culture vessels resulted in high hyperhydricity/vitrification. High concentrations of ethylene and CO2 were found to accumulate in these vessels. The hyperhydricity of the shoot cultures could be decreased by progressively ventilating the vessels. Exchange of gases was achieved by removing the Parafilm sealing without affecting sterility. This reduced the hyperhydricity rate and gave a good recovery of vitrified shoots, but resulted in decreased proliferation and a dehydration of proliferating nodal segments and the culture medium. The best number of normal shoots was observed when the parafilm was removed for gaseous exchange after four weeks of culture incubation. The results show that hyperhydricity in shoot cultures of S. yoshimurae could be prevented by sufficient gas exchange during culture.  相似文献
