表达HSV－2 gD基因的重组痘苗病毒保护小鼠对抗致死量HSV病霉攻击

利用ＰＣＲ方法对单纯疱疹病毒Ⅱ型糖蛋白Ｄ（ＨＳＶ－２ｇＤ）基因进行了修饰,在其５＇端删去约５００ｂｐ的非编码区,仅保留ＡＴＧ上游７个ｂｐ。将修饰后的ＨＳＶ－２ｇＤ基因插入到带有痘苗病毒天坛株ＴＫ基因区段的痘苗表达质粒ｐＪＳＡ１１７５,置于痘苗病毒Ｐ７．５ｋ早／晚期启动子控制下。将此重组质粒用脂质体Ｌｉｐｏｆｅｃｔｉｎ方法转染已受野型ＴＫ￣＋痘苗病毒天坛株感染的ＴＫ￣－１４３细胞,通过同源重组机制和标志基因ＬａｃＺ产物的蓝斑显色作用,以及ＢｕｄＲ试剂对ＴＫ表型的选择压力,筛选出整合有ＨＳＶ－２ｇＤ基因的重组痘苗病毒。Ｓｏｕｔｈｅｍ杂交表明,ＨＳＶ－２ｇＤ基因已正确地插入痘苗病毒ＴＫ基因区内;间接免疫荧光检测显示,ＨＳＶ－２ｇＤ蛋白已得到有效表达,且主要分布于细胞膜。重组病毒免疫家兔可产生明显的抗ＨＳＶ－２ｇＤ中和抗体。用重组病毒免疫小鼠,３周后可使９４％（１７／１８）的小鼠对抗ＨＳＶ－２的致死量攻击,表明重组病毒具有明显的免疫保护作用。     

中国典型草原放牧草场蝗虫为害损失及经济阈值的估算(英语)

传统上估算蝗虫在放牧草场为害损失的方法几乎都是用来测定对牧草秋季产量的影响,而实际上,在估算放牧草场蝗虫为害损失及经济阈值时,牧草的现存量而非秋季产量是更应考虑的因素.本文提出了一种适合测定蝗虫对牧草现存生物量影响的新方法,即野外挂笼饲养与蝗虫种群动态相结合的估算方法,并在此基础上组建了放牧草场蝗虫种群经济阈值模型;α_0 α_1M_1_α_2M_2_ α_3S_1 α_4S_2=C其中,M_1:狭翅雏蝗生物量;M_2:宽须蚁蝗生物量;S_1:狭翅雏蝗平均个体重量,S_2:宽须蚁蝗平均个体重量;α_0-α_4:常数.同时引入蝗虫种群数量和生物量两项参数来表达蝗虫种群的发生程度.     

多异瓢虫生物学的研究

多异瓢虫在山东省德州地区一年发生４代,部分个体５代,以成虫在杂草丛内、残枝落叶及土块下越冬。在自然变温下,卵、幼虫、预蛹、蛹、成虫产卵前期和全世代的发育始点和有效积温分别为１８．８±０．５２℃和１５．０日度、１６．９７±０．０３℃和７３．３日度、１５．９７±０．０１℃和１０．２３日度、１５．４７±０．０５℃和２３．０５日度、１７．６±０．０４℃和３４．１日度、１８．２±０．１０℃和１７０．３日度。据Ｈｏｌｉｎｇ圆盘方程测定,１─４龄幼虫和产卵前期成虫日最大捕食棉蚜量依次力６．７头、３６．４头、７７．５头、８１．３头和６８．０头。     

LC－CW的抗肿瘤作用及其机理的研究

提取干酪乳酸杆菌细胞壁成分（ＬＣ－ＣＷ）,研究其抗肿瘤作用及其机理。结果表明：１００μｇＬＣ－ＣＷ,ｉｐ,连续４天,可明显抑制小鼠Ｓ１８腹水瘤移植物的生长,抑瘤率为５４％。增强ＩＬ－２诱导的ＬＡＫ杀伤活性,可明显促进小鼠ＮＫ杀伤活性,明显促进小鼠Ｔ细胞转化,促进ＣｏｎＡ和ＰＨＡ－Ｐ诱导的ＩＬ－２产生,促进ＳＩＬ－２Ｒ的减少。研究结果表明ＬＣ－ＣＷ是一种重要的抗肿瘤免疫调节因子。     

Functional response of Neoseiulus barkeri Hughes on two-spotted spider mite (Acari: Tetranychidae)

Generalist predatory mites are the common phytoseiid fauna in many agroecosystems, but little attention has been paid to their potential as biological control agents. In this study, we determined the functional responses of adult females of the generalist predator Neoseiulus barkeri Hughes on eggs, larvae, and adults of the two-spotted spider mite, Tetranychus urticae Koch, in the laboratory. Predation experiments were conducted on pepper leaf discs over a 24 h period at 25±1°C, 70–80% RH and 16L:8D photoperiod. Prey densities ranged 5 to 80 eggs, or 5 to 40 larvae, or 1 to 8 female adults of T. urticae per disc. The predation rate of N. barkeri adult females on T. urticae eggs was the same as on its larvae, but the predation rate on adult females was much lower. The role of generalist predatory mites in integrated and biological control of greenhouse pests was discussed.     

Microbial oxidation of cumene by octane-grown cells

Previously, we reported that eight glucose-grown microbial cultures out of 1229 screened oxidize the alkyl side-chain of 2-phenylpropane (cumene) stereospecifically. Now, we have adapted these cultures to grow on n-octane and found that their cumene oxidation activities increased more than 30 times. We also found an additional 11 cultures (ten bacteria, one actinomycete) that oxidized cumene when grown on octane but not on glucose. In general, octane-grown cells were more active in cumene oxidation than glucose-grown cells. Rhodococcus rhodochrous NRRL B-2153 showed the best conversion yield (2-phenyl-1-propanol plus 2-phenyl-1-propionic acid was 5.5%) at 25°C, pH 8.0, 250 rpm, and 12 h of reaction. Structures of the reaction products were confirmed by gas chromatography (GC)/mass spectrometry and GC/infrared analyses. Products contained 84% ee (enantiomeric excess) of the R(–) isomer, as analyzed with a GC cyclodextrin chiral column. Strain B-2153 oxidized alkylbenzenes in the following order of reaction rate: ethylbenzene >amylbenzene > butylbenzene > cumene > propylbenzene > sec-butylbenzene. tert-Butylbenzene was not oxidized.     

A proper transmembrane Ca2+ gradient is essential for the stimulation of adenylate cyclase activity by stimulatory GTP-binding protein

Stimulatory GTP-binding Protein (Gs) and adenylate cyclase prepared from bovine brain cortices were co-reconstituted into asolectin vesicles with or without 1000-fold transmembrane Ca²⁺ gradient. The results showed that both basal activity and Gs-stimulated activity of adenylate cyclase were highest in proteoliposomes with a transmembrane Ca²⁺ gradient similar to physiological condition (1 M Ca²⁺ outside and 1 mM Ca²⁺ inside) and lowest when the transmembrane Ca²⁺ gradient was in the inverse direction. Such a difference could be diminished following dissipation of the transmembrane Ca²⁺ gradient by A23187. Comparable conformational changes of Gs in proteoliposomes were also observed when Gs was labeled with the fluorescence probe, acrylodan. These results may indicate that a proper transmembrane Ca²⁺ gradient is essential not only for higher adenylate cyclase activity but also for its stimulation by Gs.
