Single sequence repeat (SSR) and amplified fragment length polymorphic (AFLP) molecular marker genotypes in cheatgrass (Bromus tectorum) were compared to published data on phenotypic variation in seed dormancy, vernalization requirement, and resistance to the pathogen Ustilago bullata. Several features of cheatgrass facilitated this study: it is a recent invader in the western United States, has considerable phenotypic polymorphism, and is an obligate self-pollinator. Forty self-pollinating lines from four populations common to the three phenotypic data sets were analyzed for molecular genetic variation using seven SSR loci and 31 AFLP loci. We examined correlations between distance matrices using the Mantel test for each pair of studies. The two molecular data sets were significantly correlated (r = 0.636). The AFLP markers often distinguished among several lines with identical SSR genotypes. The AFLP data were also significantly correlated with the phenotypic data (r values from 0.4640 to 0.5658), but the SSR data were much more highly correlated (r values from 0.677 to 0.844). The difference between molecular marker systems was especially notable when an outlier population from Potosi Pass, Nevada, was excluded from the analysis. These results suggest that SSR markers may be good surrogates for phenotypic traits in population genetic studies of strongly inbreeding species such as cheatgrass.  相似文献

Nonhuman primates from domestic sources constitute an important resource for the research community. The life history of the Old World monkey species that comprise the bulk of this resource is described, and issues that colony managers and researchers alike should consider regarding animal selection (e.g., species, age, sex, rearing history, temperament, genotype, viral status, geographic origin) are discussed. Preparation of domestically bred animals for research usually involves some combination of social separation, relocation, resocialization, alterations in physical space, photoperiod, and diet, as well as exposure to novel environments. The research literature that has focused on these issues is reviewed, and authors suggest that once animals have been assigned to their project housing situation, a period ranging up to 3 mo (depending on the magnitude of the change in housing) might be warranted before an experimental protocol should begin. Attention to issues of animal selection and conditioning by both researchers and colony managers can lead to the shared goal of high-quality research that utilizes the minimal number of animals.  相似文献

A series of UDP-D-glucose derivatives and precursors that have been modified at C-3 were synthesised from D-glucose as potential chain terminators of beta-glucan biosynthesis. Bromus tectorum (cheatgrass) is a flourishing invasive weed in the western United States. The objective of our study is to characterize its genetic diversity. We made a B. tectorum genomic library in lambda phage and screened approximately 4000 clones for poly CA and poly CT dinucleotide repeats. Of 38 sequences with dinucleotide repeats isolated from the library, we designed primer sets for 18 loci. A preliminary screen of 40 individuals from four populations indicated that seven loci were polymorphic. These loci will be valuable for elucidation of cheatgrass genetic diversity and population structure.  相似文献

Phosphite is a cost-effective fungicide used to control the pathogen Phytophthora cinnamomi which is damaging the diverse flora of the southwest of Western Australia. Three annual species of the southwest jarrah (Eucalyptus marginata) forest of Western Australia (Pterocheata paniculata, Podotheca gnaphalioides and Hyalosperma cotula), were studied to determine the effect of the fungicide phosphite on the species’ reproduction. Phosphite at concentrations of 2.5, 5 and 10 g L–1 reduced pollen fertility of Pt. paniculata when plants were sprayed at the vegetative stage. Pollen fertility of all three species was reduced when plants were sprayed at anthesis with 10 g L–1 phosphite. Seed germination was reduced by phosphite in Pt. paniculata and H. cotula when plants were sprayed in the vegetative stage. Phosphite when sprayed at anthesis at a concentration of 5 g L–1 reduced seed germination of H. cotula. Phosphite at concentrations of 5 and 10 g L–1 killed a proportion of plants from all three species and up to 90% of Po. gnaphalioides plants. The frequent application of phosphite, therefore, may reduce the abundance of annual plants in this ecosystem. Received: 14 December 2000 / Revision accepted: 10 March 2001  相似文献

Microoxygen polarographic electrodes were constructed and used to measure oxygen tension (POO2) in the eyes of rainbow trout (Salmo gairdneri). The values obtained are compared with arterial blood and environmental water POO2 and indicate that there is an oxygen-concentrating mechanism in the eye supplying oxygen to the avascular retina. Anatomically similar retes suggest that the mechanism is similar to the one which exists in the swim bladder. Elimination of the arterial blood supply to the choroidal gland rete mirabile of the eye (through pseudobranchectomy) and the consequent lowering of ocular oxygen tensions implicate the choroidal gland as one of the major components of the oxygen-concentrating mechanism. After pseudobranchectomy the presence of ocular POO2 above that of arterial blood is indicative of a secondary structure in the eye capable of concentrating oxygen. Inhibition of carbonic anhydrase, using acetazolamide, is shown to result in complete suppression of the oxygen-concentrating mechanism. A hypothesis is advanced for the participation of retinal-choroidal and erythrocyte carbonic anhydrase in the oxygen-concentrating mechanism.  相似文献

Vervet monkeys (Cercopithecus aethiops sabaeus) show individual differences in approach-avoidance behavior when faced with an unfamiliar and potentially threatening situation. Prior research from our colony demonstrated that juveniles who had experienced high levels of early maternal protectiveness were more cautious in response to novelty, compared to juveniles who had had less protective mothers. The research reported here was designed to verify this result in a paradigm that experimentally varied maternal protectiveness through the introduction of new breeding adult males. Mothers responded to the presence of new males by increased maternal protectiveness toward infants born in the year following the introductions. Individual differences in response to the unfamiliar were later evaluated by measuring the latency to approach within 1 m of novel food containers placed into the home enclosure of four naturally composed social groups. Infants approached with the same latency and in the same order as their mothers. Juveniles approached sooner than, and independent of, their mother's current behavior, but their latency to approach could be predicted by the experimentally induced variation in maternal protectiveness they had experienced as infants. Immatures who had been born in New Male years, when maternal protectiveness was high, were more cautious and had significantly longer latencies to approach the novel stimulus compared to immatures who had been born in Resident Male years. © 1993 Wiley-Liss, Inc.  相似文献
