TGF-β is a ubiquitous protein that exhibits a broad spectrum of biological activity. The prokaryotic expression and purification of the extracellular domain of the type II TGF-β receptor (TβR-II-ED), without the need for fusion protein cleavage and refolding, is described. The recombinant TβR-II-ED fusion protein bound commercially available TGF-β1 and displayed an affinity of 11.1 nM. In a modified ELISA, receptor binding to TGF-β1 was inhibited by TGF-β3. The technique lends itself to high-throughput screening of combinatorial libraries for the identification of TGF-β agonists and antagonists and this, in turn, may have important therapeutic implications. 相似文献

The 70-kilodalton heat shock protein (hsp70) family of molecular
chaperones, which contains both stress-inducible and normally abundant
constitutive members, is highly conserved across distantly related taxa.
Analysis of this protein family in individuals from an outbred population
of tropical topminnows, Poeciliopsis gracilis, showed that while
constitutive hsp70 family members showed no variation in protein isoforms,
inducibly synthesized hsp70 was polymorphic. Several species of
Poeciliopsis adapted to desert environments exhibited lower levels of
inducible hsp70 polymorphism than the tropical species, but constitutive
forms were identical to those in P. gracilis, as they were in the
confamilial species Gambusia affinis. These differences suggest that
inducible and constitutive members of this family are under different
evolutionary constraints and may indicate differences in their function
within the cell. Also, northern desert species of Poeciliopsis synthesize a
subset of the inducible hsp70 isoforms seen in tropical species. This
distribution supports the theory that ancestral tropical fish migrated
northward and colonized desert streams; the subsequent decrease in
variation of inducible hsp70 may have been due to genetic drift or a
consequence of adaptation to the desert environment. Higher levels of
variability were found when the 30- kilodalton heat shock protein (hsp30)
family was analyzed within different strains of two desert species of
Poeciliopsis and also in wild-caught individuals of Gambusia affinis. In
both cases the distribution of hsp30 isoform diversity was similar to that
seen previously with allozyme polymorphisms.

Mononuclear cells (MNC) derived from peripheral blood (PBMNC) of 23 normal donors and 4 AIDS patients, and from bone marrow (BMMNC) of 15 normal donors were incubated at 37 degrees C in culture medium alone or in the presence of either natural or recombinant human interleukin-2 (IL-2) or recombinant human interferon-gamma (IFN-gamma; 1-1,000 U/ml). The cultured cells were washed on days 1, 4 or 7 and tested for various immune functions in vitro and for cell surface phenotype. IL-2, but not IFN-gamma, was found mitogenic for both PBMNC and BMMNC. The natural killer (NK) activity of both PBMNC and BMMNC was the only function tested that was markedly augmented (over 100-fold compared to medium control) by both lymphokines. Pretreatment of PBMNC with IL-2 at greater than or equal to 10 U/ml profoundly suppressed (up to 90%) various functions, such as mitogenic responses (phytohemmagglutinin, concanavalin A, pokeweed mitogen), allogeneic mixed leukocyte reaction, antibody production and T cell colony formation in agar. In contrast, some BMMNC functions were elevated at low doses of IL-2 and IFN-gamma, and significant suppression of BMMNC was seen only with high doses of IL-2 (greater than or equal to 100 U/ml) and IFN-gamma (1,000 U/ml). IL-2 was by far more effective than IFN-gamma in both the amplification of NK activity and the suppression of most of the other functions. IL-2, but not IFN-gamma, was found to activate/induce suppressor cells and increased the proportion of Leu-2+ (CD8) cells in PBMNC; the suppressive effect was time- and dose-dependent. The IL-2-induced suppression could be diminished by inclusion of anti-IL-2 antibody during the pretreatment phase. Similar suppressive effects were noted in PBMNC from AIDS patients. These findings suggest that: (a) high-dose IL-2 may elicit immunosuppression which can be mediated by nondiscriminative highly cytotoxic cells (i.e. lymphokine-activated killer cells) and/or by noncytotoxic, nonspecific suppressor cells, and (b) that PBMNC respond differently to the lymphokines than do BMMNC. 相似文献

OBJECTIVES--To investigate and quantify the extent to which variations in guidelines influence assessment of control of hypertension. DESIGN--Cross sectional study. Selected patients had hypertension assessed as controlled or uncontrolled with guidelines from New Zealand, Canada, the United States, Britain, and the World Health Organisation. SETTING--18 general practices in Oxfordshire. SUBJECTS--876 patients with diagnosed hypertension and taking antihypertensive drugs. MAIN OUTCOME MEASURES--Proportion of patients with controlled hypertension according to each set of guidelines. RESULTS--The proportion of patients with controlled hypertension varied from 17.5% to 84.6% with the different guidelines after adjustment for the sampling method. All five sets of guidelines agreed on the classification for 31% (277) of the patients. The New Zealand guidelines calculate an absolute risk of a cardiovascular event. When this was taken as the standard half of the patients with uncontrolled hypertension by the United States criteria would be treated unnecessarily and 31% of those classified as having controlled hypertension by the Canadian guidelines would be denied beneficial treatment. CONCLUSIONS--Hypertension guidelines are inconsistent in their recommendations and need to make clear the absolute benefits and risks of treatment. 相似文献

Summary Mycorrhizae play an important role in regulating patterns of energy and nutrient flux in terrestrial ecosystems. To conceptualize this role I develop the theory behind a simple index of the efficiency of soil resource acquisition by plant root systems (E). The morphological, physiological and demographic characteristics of mycorrhizae that define E appear to vary with environment and with plant community composition. This theory is elaborated with examples drawn from forest ecology literature. Some inconsistencies among observations of fine root dynamics are particularly revealing: (1) belowground carbon allocation vs soil fertility; (2) causes of root mortality; (3) root longevity vs decomposition rates. A comprehensive theory of mycorrhizal and ecosystem dynamics must await resolution of these inconsistencies and better quantitative information on mycorrhizal features affecting E. 相似文献

Reductions in snow cover undera warmer climate may cause soil freezing eventsto become more common in northern temperateecosystems. – leaching. Elevated leaching of Pi from the forestfloor was coupled with enhanced retention ofPi in the mineral soil Bs horizon. Thequantities of Pi mobilized from the forestfloor were significant relative to theavailable P pool (22 mol ha–1) as well asnet P mineralization rates in the forest floor(180 mol ha–1 yr–1). Increased fineroot mortality was likely an important sourceof mobile N and Pi from the forest floor,but other factors (decreased N and P uptake byroots and increased physical disruption of soilaggregates) may also have contributed to theenhanced leaching of nutrients. Microbialmortality did not contribute to the acceleratedN and P leaching after soil freezing. Despite its importance in the terrestrial C cycle rhizosphere carbon flux (RCF) has rarely been measured for intact root–soil systems. We measured RCF for 8‐year‐old saplings of sugar maple (Acer saccharum) and yellow birch (Betula allegheniensis) collected from the Hubbard Brook Experimental Forest (HBEF), NH and transplanted into pots with native soil horizons intact. Five saplings of each species were pulse labeled with 13CO2 at ambient CO2 concentrations for 4–6 h, and the 13C label was chased through rhizosphere and bulk soil pools in organic and mineral horizons for 7 days. We hypothesized yellow birch roots would supply more labile C to the rhizosphere than sugar maple roots based on the presumed greater C requirements of ectomycorrhizal roots. We observed appearance of the label in rhizosphere soil of both species within the first 24 h, and a striking difference between species in the timing of 13C release to soil. In sugar maple, peak concentration of the label appeared 1 day after labeling and declined over time whereas in birch the label increased in concentration over the 7‐day chase period. The sum of root and rhizomicrobial respiration in the pots was 19% and 26% of total soil respiration in sugar maple and yellow birch, respectively. Our estimate of the total amount of RCF released by roots was 6.9–7.1% of assimilated C in sugar maple and 11.2–13.0% of assimilated C in yellow birch. These fluxes extrapolate to 55–57 and 90–104 g C m?2 yr?1 from sugar maple and yellow birch roots, respectively. These results suggest RCF from both arbuscular mycorrhizal and ectomycorrhizal roots represents a substantial flux of C to soil in northern hardwood forests with important implications for soil microbial activity, nutrient availability and C storage. 相似文献
