gamma-Aminobutyric acid (GABA) was applied to the superior cervical ganglion (SCG) of CFY rats in vitro and in vivo, with or without implantation of a hypoglossal nerve, to evaluate the effects of these experimental interventions on the acetylcholine (ACh) system, which mainly serves the synaptic transmission of the preganglionic input. Long-lasting GABA microinfusion into the SCG in vivo apparently resulted in a "functional denervation." This treatment reduced the acetylcholinesterase (AChE; EC 3.1.1.7) activity by 30% (p less than 0.01) and transiently increased the number of nicotinic acetylcholine receptors, but had no significant effect on the choline acetyltransferase (acetyl-coenzyme A:choline-O-acetyltransferase; EC 2.3.1.6) activity, the ACh level, or the number of muscarinic acetylcholine receptors. The relative amounts of the different molecular forms of AChE did not change under these conditions. In vivo GABA application to the SCG with a hypoglossal nerve implanted in the presence of intact preganglionic afferent synapses exerted a significant modulatory effect on the AChE activity and its molecular forms. The "hyperinnervation" of the ganglia led to increases in the AChE activity (to 142.5%, p less than 0.01) and the 16S molecular form (to 200%, p less than 0.01). It is concluded that in vivo GABA microinfusion and GABA treatment in the presence of additional cholinergic synapses has a modulatory effect on the elements of the ACh system in the SCG of CFY rats. 相似文献

The effects of in vivo treatment with recombinant human IFN-alpha 2a (rIFN-alpha 2a) on the distribution of T cell subpopulations were examined in 21 patients with renal cell adenocarcinoma, using two-color flow cytometry with anti-Leu8 in combination with anti-Leu2 and anti-Leu3. Other parameters indicative of immune status, such as the number and percentage of total (CD3) T cells, in vitro proliferation to mitogen, and spontaneous immunoglobulin secretion, were also measured, prior to the initiation of treatment with rIFN-alpha 2a, and during treatment. Total T cell number decreased after treatment with rIFN-alpha 2a, to a low of 54% of mean pretreatment values after 4 weeks. The CD4/CD8 ratio did not change appreciably following treatment with rIFN-alpha 2a. However, the number of Leu8-negative T cells, within both the CD4 and the CD8 T cell populations, decreased more than the number of Leu8-positive T cells. D1 and D2 dopamine receptors were characterized in the caudate-putamen region of nonhuman primate brains (Macaca fascicularis). D1 dopamine receptors were identified with [3H]SCH 23390 and D2 receptors with [3H]-spiperone. Scatchard analysis of [3H]SCH 23390 saturation data using washed membranes revealed a single high-affinity binding site (KD, 0.352 +/- 0.027 nM) with a density (Bmax) of 35.7 +/- 2.68 pmol/g original wet tissue weight (n = 10). The affinity of [3H]spiperone for the D2 site was 0.039 +/- 0.007 nM and the density was 25.7 +/- 1.97 pmol/g original wet tissue weight (n = 10). D1 and D2 receptors in nonhuman primates may be differentiated on the basis of drug affinities and stereoselectivity. In competition experiments, RS-SKF 38393 was the most selective D1 agonist, whereas (+)-4-propyl-9-hydroxynaphthoxazine [(+)-PHNO] was the most selective D2 agonist. Apomorphine was essentially nonselective for D1 or D2 binding sites. Of the antagonists, R-SKF 83566 and SCH 23390 were the most selective for the D1 site, whereas YM-09151-2 was the most selective for the D2 site. cis-Flupentixol and (S)-butaclamol were the least selective dopamine antagonists. D1 receptors bound benzazepine antagonists (SCH 23390/SCH 23388, R-SKF 83692/RS-SKF 83692) stereoselectively whereas D2 receptors did not. Conversely D2 receptors bound (S)-sulpiride and (+)-PHNO more potently than their enantiomers whereas D1 receptors showed little stereoselectively for each of these isomeric pairs. Aerobic radioprotection of pBR322 by thiols: effect of thiol net charge upon scavenging of hydroxyl radicals and repair of DNA radicals. 总被引:1,自引:0,他引:1
The extent of conversion of supercoiled pBR322 plasmid DNA to the open circular and linear forms can be measured by HPLC on a Waters Gen Pak FAX column following in vitro gamma irradiation of the DNA. This radiation effect has proven to be useful for the study of the radioprotection of DNA by thiols and other drugs. This system was used with gamma irradiation in air at pH 7.0 and physiological ionic strength to compare radioprotection by a series of thiols, disulfides, and thioethers, all having approximately 10(8) s-1 effective hydroxyl radical scavenging rate (10 mm dm-3 drug) and having net charge (Z) ranging from -2 to +3. All sulfur compounds exhibited substantial protection due to scavenging of hydroxyl radicals in bulk solution but thiols exhibited a 24-fold variation in relative ability to protect the plasmid DNA from strand breaks, as assessed from the dose-response curves: mercaptosuccinate (Z = -2), 0.53; GSH (Z = -1), 0.67; 3-mercaptopropionate (Z = -1) 0.80; mercaptoethanol (Z = 0), 1.00; dithiothreitol (Z = 0), 1.5; cysteamine (Z = +1), 3.7; N-(2-mercaptoethyl)-1,3-diaminopropane (WR-1065, Z = +2), 6.7; N1-(2-mercaptoethyl)spermidine (WR-35980, Z = +3), 12. Comparison of these results with those obtained using disulfide and thioether radioprotectors indicated that local scavenging of hydroxyl radicals near DNA increases slightly with Z, apparently as a result of variations in thiol concentration near DNA, but this accounts for only a small fraction of the change with Z found for cationic thiols. The marked increase in protection found for cationic thiols was attributed to chemical repair of DNA radicals and was in accord with predictions based upon recently measured rates for chemical repair of DNA radicals and was in accord with predictions based upon recently measured rates for chemical repair of pBR322 radicals. It is concluded that chemical repair of DNA radicals by anionic thiols does not compete with the oxygen fixation reaction in air and that protection by these thiols occurs primarily via the scavenging of hydroxyl radicals. However, chemical repair of DNA radicals is significantly enhanced by counterion condensation for cationic thiols and becomes a significant factor in their ability to protect DNA against radiation damage under aerobic conditions. 相似文献

The Experimental Watershed Liming Study (EWLS) was initiated to evaluate the application of CaCO3 to a forested watershed in an effort to mitigate the acidification of surface water. The objective of the EWLS was to assess the response of the Woods Lake watershed to an experimental addition of CaCO3. During October 1989, 6.89 Mg CaCO3/ha was applied by helicopter to two subcatchments comprising about 50% (102.5 ha) of the watershed area. The EWLS involved individual investigations of the response of soil and soil water chemistry, forest and wetland vegetation, soil microbial processes, wetland, stream and lake chemistry, and phytoplankton and fish to the CaCO3 treatment. In addition, the Integrated Lake/Watershed Acidification (ILWAS) model was applied to the site to evaluate model performance and duration of the treatment. The results of these studies are detailed in this volume. The purposes of this introduction and synthesis paper are to: 1) present the overall design of the EWLS, 2) discuss the linkages between the individual studies that comprise the EWLS, and 3) summarize the response of the lakewater chemistry to watershed addition of CaCO3 and compare these results to previous studies of direct lake addition. An analysis of lake chemistry revealed the watershed treatment resulted in a gradual change in pH, acid neutralizing capacity (ANC) and Ca2+ in the water column. This pattern was in contrast to direct lake additions of CaCO3 which were characterized by abrupt changes following base addition and subsequent rapid reacidification. Over the three-year study period, the supply of ANC to drainage waters was largely derived from dissolution of CaCO3 in wetlands. Relatively little dissolution of CaCO3 occurred in freely draining upland soils. The watershed treatment had only minor effects on forest vegetation. The watershed treatment eliminated the episodic acidification of streamwater and the near-shore region of the lake during snowmelt, a phenomenon that occurred during direct lake treatments. Positive ANC water in the near-shore area may improve chemical conditions for fish reproduction, and allow for the development of a viable fish population. The watershed CaCO3 treatment also decreased the transport of Al from the watershed to the lake, and increased the concentrations of dissolved organic carbon (DOC) and dissolved silica (H4SiO4) in stream and lakewater. The watershed treatment appeared to enhance soil nitrification, increasing concentrations of NO3
– in soilwater and surface waters. However, the acidity associated with this NO3
– release was small compared to the increase in ANC due to CaCO3 addition and did not alter the acid-base status of Woods Lake. Acid neutralizing capacity (ANC) budgets for 12-month periods before and after the watershed treatment showed that the lake shifted from a large source of ANC to a minor source due to retention of SO4
2–, NO3
–, Al and the elevated inputs of Ca2+ associated with the watershed CaCO3 application. In contrast to the direct lake treatments, Ca2+ inputs from the watershed application were largely transported from the lake. 相似文献

Genetic constitution in the intertidal gastropod Nucella lapillus (L.) influences shell shape, growth rate and physiology. Clinal variation in these traits along a 5 km stretch of coastline in south Devon can be related to environmental variation in temperature and desiccation stress. We have examined mtDNA variation along this shore to investigate whether the cline represents primary or secondary contact. Two distinct mtDNA haplotypes were found which exhibit coincident step clines with karyotypic, allozymic and phenotypic variation and covary with the environmental pressures of temperature and desiccation. These results are interpreted in the context of the wider scale distribution of genetic and phenotypic variation in N. lapillus. It is suggested that the shore studied may represent one of a number of regions of secondary contact within a mosaic hybrid zone in N. lapillus , where coadapted phenotypic variation correlates with habitat and the position of the clines represents an environmental transition. 相似文献

Utility of the white gene in estimating phylogenetic relationships among mosquitoes (Diptera: Culicidae) 总被引:2,自引:0,他引:2
The utility of a nuclear protein-coding gene for reconstructing
phylogenetic relationships within the family Culicidae was explored.
Relationships among 13 species representing three subfamilies and nine
genera of Culicidae were analyzed using a 762-bp fragment of coding
sequence from the eye color gene, white. Outgroups for the study were two
species from the sister group Chaoboridae. Sequences were determined from
clone PCR products amplified from genomic DNA, and aligned following
conceptual intron splicing and amino acid translation. Third codon
positions were characterized by high levels of divergence and biased
nucleotide composition, the intensity and direction of which varied among
taxa. Equal weighting of all characters resulted in parsimony and
neighboring-joining trees at odds with the generally accepted phylogenetic
hypothesis based on morphology and rDNA sequences. The application of
differential weighting schemes recovered the traditional hypothesis, in
which the subfamily Anophelinae formed the basal clade. The subfamily
Toxorhynchitinae occupied an intermediate position, and was a sister group
to the subfamily Culicinae. Within Culicinae, the genera Sabethes and
Tripteroides formed an ancestral clade, while the Culex-Deinocerites and
Aedes- Haemagogus clades occupied increasingly derived positions in the
molecular phylogeny. An intron present in the Culicinae- Toxorhynchitinae
lineage and one outgroup taxon was absent in the basal Anophelinae lineage
and the second outgroup taxon, suggesting that intron insertions or
deletions may not always be reliable systematic characters.
