The protein composition of Mycobacterium tuberculosis and the molecular weight of proteins contained in these organisms have been determined by the method of electrophoresis in the porosity gradient of polyacrylamide gel. Close similarity between the electrophoregrams of M. tuberculosis clinical and laboratory strains has been revealed. The study of 18 strains of different groups of mycobacteria has shown that M. tuberculosis are essentially different from opportunistic mycobacteria and acid-resistant saprophytes. These data may be important for the identification and taxonomy of mycobacteria.  相似文献

Each of two hydrophobic subunits of Na+-translocating NADH:quinone oxidoreductase (NQR), NqrD and NqrE, contain a pair of strictly conserved cysteine residues within their transmembrane alpha-helices. Site-directed mutagenesis showed that substitutions of these residues in NQR of Vibrio harveyi blocked the Na+-dependent and 2-n-heptyl-4-hydroxyquinoline N-oxide-sensitive quinone reductase activity of the enzyme. However, these mutations did not affect the interaction of NQR with NADH and menadione. It was demonstrated that these conserved cysteine residues are necessary for the correct folding and/or the stability of the NQR complex. Mass and EPR spectroscopy showed that NQR from V. harveyi bears only a 2Fe-2S cluster as a metal-containing prosthetic group.  相似文献

Degradation of juvenile hormone and reproductive function during starvation and experimental increase of the juvenile hormone titer were studied in wild type and mutant D. virilis females incapable to respond to heat stress by changes in juvenile hormone metabolism and fertility. After 24-hour starvation, the females of both lines were characterized by a decreased level of juvenile hormone degradation, 24-hour delay of egg laying, increased egg laying within 3 h after the termination of starvation, and decreased fertility within three days. Application of exogenous juvenile hormone also led to a decreased level of its degradation and 24-hour arrest of egg laying. Experimental increase of the juvenile hormone titer before the beginning of starvation led to a sharply increased fertility (number of laid eggs and number of progenies) within the first 24 h after the termination of starvation. The dynamics of juvenile hormone degradation and of fertility were similar after starvation and upon application of the exogenous hormone. The role of juvenile hormone in the control of egg maturation and laying under stress conditions has been discussed.  相似文献

Computer-assisted superimposition of the equivalent dipole sources (EDSs) of the main pathological EEG patterns with the results of computer tomography and magnetic resonance imaging of the brain was performed for patients with organic CNS lesions. The greatest degree of structural determination was revealed for local and lateralized (in a traumatic injury) slow waves, whose EDSs mapped to the perifocal zone of the injury focus. The EDSs of epileptiform patterns could be located in the zone of damaged brain substance or in the intact hemisphere (when the limbic structures were damaged), reflecting the formation of secondary functional foci in the latter case. Bilateral paroxysmal patterns were more frequently determined by the functional state of the medial brain structures. The methods of the retrospective epidemiological analysis of morbidity rate among students attending a specialized polyclinic for students have made it possible to establish the fact that acute respiratory diseases of non-influenza etiology are of the greatest socio-economic importance for students living under the conditions of a hostel. Such importance of these infections is linked with the autonomous character of the epidemic process due to the "mixing" factor, the intensive realization of the drop mechanism of infection transfer, and high morbidity risk in the absence of immunoprophylactic remedies. The results of this study indicate that the student body may be considered a group of primary necessity for prophylactic medical examination aimed at the early detection of the consequences of the past disease and at the organization of timely and effective treatment of young people.  相似文献

Abstract

Structurally isomeric complexes formed between homopyrimidine bis-PNAs (T2JT2JT4-linker-T4CT2CT2) and single- and double-stranded DNA targets were investigated. These complexes are triplexes designated S1, S2 and S3 in order of increased mobility by polyacrylamide gel electrophoresis. It is shown that the S3 isomer is formed only on double-stranded DNA and possesses highest stability. Isomers S2 and S1 are formed upon binding of bis-PNA to double-stranded as well as to single-stranded DNA. It was found that the stability of the isomer S1 increases dramatically in the presence of excess single-stranded oligonucleotide complementary to the bis-PNA. The structure of the stabilized S1 isomer is proposed to consist of two bis-PNA/DNA triplexes. The relationship between the yield of the isomer S1 formed on single-stranded DNA and the bis-PNA concentration was investigated and a kinetic model of the formation of S1 is presented.  相似文献

Cultured mesenchymal stromal cells (MSCs) from different sources represent a heterogeneous population of proliferating non-differentiated cells that contains multipotent stem cells capable of originating a variety of mesenchymal cell lineages. Despite tremendous progress in MSC biology spurred by their therapeutic potential, current knowledge on receptor and signaling systems of MSCs is mediocre. Here we isolated MSCs from the human adipose tissue and assayed their responsivity to GPCR agonists with Ca2 + imaging. As a whole, a MSC population exhibited functional heterogeneity. Although a variety of first messengers was capable of stimulating Ca2+ signaling in MSCs, only a relatively small group of cells was specifically responsive to the particular GPCR agonist, including noradrenaline. RT-PCR and immunocytochemistry revealed expression of α1B-, α2A-, and β2-adrenoreceptors in MSCs. Their sensitivity to subtype-specific adrenergic agonists/antagonists and certain inhibitors of Ca2+ signaling indicated that largely the α2A-isoform coupled to PLC endowed MSCs with sensitivity to noradrenaline. The all-or-nothing dose-dependence was characteristic of responsivity of robust adrenergic MSCs. Noradrenaline never elicited small or intermediate responses but initiated large and quite similar Ca2+ transients at all concentrations above the threshold. The inhibitory analysis and Ca2+ uncaging implicated Ca2+-induced Ca2+ release (CICR) in shaping Ca2+ signals elicited by noradrenaline. Evidence favored IP3 receptors as predominantly responsible for CICR. Based on the overall findings, we inferred that adrenergic transduction in MSCs includes two fundamentally different stages: noradrenaline initially triggers a local and relatively small Ca2+ signal, which next stimulates CICR, thereby being converted into a global Ca2+ signal.  相似文献
