The criteria to classify the diversity of Neotropical Primates have recently taken the discussion boards due to the reappraisal on the phylogenetic relationship of some groups. Such controversial and arbitrary decisions, however, can hamper conservation actions in as much as it becomes difficult to prioritise and set meaningful targets. It is the case for dwarf marmosets from central Amazonia. Today, the classification of dwarf marmosets in Mico or Callibella genus has not been satisfactorily settled. Aiming to contribute to the taxonomic and conservation assessment of dwarf marmosets, we conducted new data collection during 3‐year fieldwork in the Aripuanã River, where the species was discovered. We present the first phylogenomic analysis of the evolutionary relationships between marmosets, new data from mitochondrial DNA and morphology, as well new records to clarify geographic distribution. With this new evidence, we support dwarf marmosets as the genus Callibella. We further discuss the implications for the conservation of this marmoset. 相似文献
Visceral leishmaniasis (VL) is a severe disease caused by infection with protozoa of the genus Leishmania. Classic VL is characterized by a systemic infection of phagocytic cells and an intense activation of the inflammatory response. It is unclear why 90% of infected individuals do not develop the disease while a minority develop the classical form. Furthermore, among those that develop disease, a small group progresses to more severe form that is unresponsive to treatment. The presence of inflammatory mediators in serum could theoretically help to control the infection. However, there is also a release of anti-inflammatory mediators that could interfere with the control of parasite multiplication. In this study, we took advantage of the spectrum of outcomes to test the hypothesis that the immune profile of individuals infected with Leishmania (L.) infantum is associated with the development and severity of disease.
Methodology/Principal Findings
Sera from patients with confirmed diagnosis of VL were evaluated for the presence of numerous molecules, and levels compared with healthy control and asymptomatic infected individuals.
Conclusions/Principal Findings
Although differences were not observed in LPS levels, higher levels of sCD14 were detected in VL patients. Our data suggest that L. infantum may activate the inflammatory response via CD14, stimulating a generalized inflammatory response with production of several cytokines and soluble molecules, including IFN-γ, IL-27, IL-10, IL-6 and sCD14. These molecules were strongly associated with hepatosplenomegaly, neutropenia and thrombocytopenia. We also observed that IL-6 levels greater than 200 pg/ml were strongly associated with death. Together our data reinforce the close relationship of IFN-γ, IL-10, IL-6, TNF-α and IL-27 in the immune dynamics of VL and suggest the direct participation of sCD14 in the activation of the immune response against L. infantum. 相似文献
1. Patients affected by isovaleric acidemia (IVAcidemia) suffer from acute episodes of encephalopathy. However, the mechanisms
underlying the neuropathology of this disease are poorly known. The objective of the present study was to investigate the
in vitro effects of the metabolites that predominantly accumulate in IVAcidemia, namely isovaleric acid (IVA), 3-hydroxyisovaleric
acid (3-OHIVA) and isovalerylglycine (IVG), on important parameters of energy metabolism, such as 14CO2 production from acetate and the activities of the respiratory chain complexes I–IV, creatine kinase and Na+, K+-ATPase in synaptic plasma membranes from cerebral cortex homogenates of 30-day-old rats.
2. We observed that 3-OHIVA acid and IVG did not affect all the parameters analyzed. Similarly, 14CO2 production from acetate (Krebs cycle activity), the activities of creatine kinase, and of the respiratory chain complexes
was not modified by IVA. In contrast, IVA exposition to cortical homogenates provoked a marked inhibition of Na+, K+-ATPase activity. However, this activity was not changed when IVA was directly exposed to purified synaptic plasma membranes,
suggesting an indirect effect of this organic acid on the enzyme. Furthermore, pretreatment of cortical homogenates with α-tocopherol
and creatine totally prevented IVA-induced inhibition on Na+, K+-ATPase activity from synaptic plasma membranes, whereas glutathione (GSH) and the NO synthase inhibitor Nω-nitro-l-arginine methyl ester (L-NAME) did not alter this inhibition.
3. These data indicate that peroxide radicals were probably involved in this inhibitory effect. Since Na+, K+-ATPase is a critical enzyme for normal brain development and functioning and necessary to maintain neuronal excitability,
it is presumed that the inhibitory effect of IVA on this activity may be involved in the pathophysiology of the neurological
dysfunction of isovaleric acidemic patients. 相似文献
Flooding regulates the amount and quality of habitat available for fish populations in river-floodplains. Although previous studies assessed fish population responses to river hydrology, the processes by which flooding affects fish abundance and catch remain unclear. Here, we investigated whether degree of flooding affects abundance and catch of Colossoma macropomum, a long-lived, overexploited fish population of the central Amazon Basin. We computed the degree of flooding corresponding to the feeding area of young-of-the-year C. macropomum as the annual magnitude of the moving littoral zone (ML). We estimated abundance of age classes one, two, and three of C. macropomum using a modeling program based on catch, fishing effort, and fish length frequency data from the principal commercial fishery. We found that flooding positively and non-linearly affected abundance of the age-one cohort but not of older age classes. ML data corresponding to a late rising water phase in which zooplankton, seeds, and fruits dominated the diet provided the strongest effect on age-one abundance. However, flooding effects on total catch were not found, likely due to catches comprising several age classes. These results provide support to existing evidence that the magnitude of the moving littoral zone regulates abundance of juvenile fish. Because the ML quantifies food and habitat availability for various other fish species, it may constitute an important control of fish abundance in these systems. Management of these fisheries may be improved by adjusting fishing effort based on hydrology. More generally, the information also serves to assess the impacts of hydrological alterations (e.g., dams) on fish recruitment.
Hydrological processes in riparian forests affect their geomorphology and drainage, and define their structure. Interactions among many environmental variables and the tree community in continuous riverine forests affect their formation and add to their complexity, making our understanding of these habitats particularly challenging. We evaluated the tree species diversity and richness in relation to relief and flooding regimes in the watershed of a riparian forest in southern Brazil. The environments of the topographic gradients were classified according to relief and soil. Canonical correspondence analysis and indicator species analysis were used for the environmental classification. A detailed soil survey showed that the organic matter content and pH are quite distinct among the environmental categories. Frequent flooding of low intensity on the floodplain was associated with incipient and shallow channels, greater frequency of livestock encroachment, and lower frequency of shade-tolerant species dominance in the deep relief of the valleys and the headwaters. We conclude that there is a dynamic flow of species migration between the flooded and non-flooded environments in small channels and that the narrow deep channels in the floodplain with less frequent and intense flooding favor an expansion of the upland tree species into riparian areas, with channel shape and depth important variables in the impact of cattle encroachment on vegetation. The greatest tree diversity was found in the ravine habitats in the intermediate landscapes.
Here we report the presence and expression levels of the vanC
1 and vanC 2/3 genes in vancomycin-susceptible strains of Enterococcus
faecalis. The vanC 1 and vanC 2/3 genes were located in the plasmid DNA and on the chromosome,
respectively. Specific mRNA of the vanC 1 gene was detected in one of these strains. Additionally, analysis of
the vanC gene sequences showed that these genes are related to
the vanC genes of Enterococcus gallinarum and
Enterococcus casseliflavus. The presence of
vanC genes is useful for the identification of E.
gallinarum and E. casseliflavus. Moreover, this is
the first report of vanC mRNA in E.
faecalis. 相似文献
Material brought to midden piles of leaf-cutting ants is considered to be hazardous. It is therefore expected that midden workers should not re-enter the colony, to reduce pathogen transmission. Here, we examined whether the midden workers of Atta sexdens rubropilosa remain confined to the waste compartment and, if not, whether they could perform many different behaviors in the absence of specialized nestmates. Eleven subcolonies received either midden workers or non-midden workers in addition to pupae and 50 mL of fungus garden. Survival, fungal and brood care, foraging and waste manipulation were observed daily until subcolonies death. Subcolonies maintained by midden workers died earlier: average survival times were 13.92 and 22.66 days for midden and non-midden workers, respectively. Midden workers cared for the brood and foraged as non-midden workers did but they were not as efficient in caring for the garden. Activities related to waste manipulation were more frequently performed by midden workers. These results show that midden workers are not a behaviorally rigid caste and retain many labor capabilities being able to leave the waste compartment and perform internal activities in the absence of specialized internal workers. 相似文献
Chagas disease affects approximately 10 million people mainly in Latin America. The immune regulation by the host seems to be an essential factor for disease evolution, and immune system inhibitory molecules such as CTLA-4 and PD-1 favor the maintenance of peripheral tolerance. Considering that polymorphisms at the immunoregulatory CTLA-4 and PDCD1 genes may alter their inhibitory function, we investigated the association of alleles, genotypes and haplotypes of polymorphic sites observed at the CTLA-4 and PDCD1 genes with different clinical manifestations of chronic Chagas disease (indeterminate, cardiac, digestive and mixed).
Methods
The polymorphisms at the CTLA-4 (-1722T/C, -318C/T and +49A/G) and PDCD1 (PD-1.3G/A) genes were typed using TaqMan methodology in 277 chronic Chagas disease patients classified into four groups, according to clinical characteristics, and 326 non-infected controls.
Results
Our results showed that CTLA-4 -1722CC genotype (22%), -1722C allele (27%) and CTLA-4 TCG (8.6%), TCA (26%) and CCA (15%) haplotypes were strongly associated with the indeterminate form, while the CTLA-4-318CT genotype (82%) and CTLA-4-318T allele (47%) were found mainly in patients with the mixed form of the disease. The CTLA-4 TCG haplotype (10.2%) was associated with the digestive form. On the other hand, the PD-1.3G/A polymorphism was not associated with chronic Chagas disease and its clinical manifestations.
Conclusions
Here, we showed that alleles, genotypes and haplotypes reported to increase the expression of the regulatory molecule CTLA-4 were associated with the indeterminate form of the disease. Taken together, our data support the idea that polymorphic sites at immunoregulatory genes may influence the development of Chagas disease variants. 相似文献
