For many years, the search for efficient surface plasmon polariton (SPP) excitation mechanisms has been a recurring matter in the development of compact plasmonic devices. In this work, we excited SPPs illuminating a subwavelength metallic ridge with a focused spot to characterize the coupling efficiency by varying the incidence angle of the excitation beam from ??50 to 50°. The intensity distribution of the excited SPPs was measured using leakage radiation microscopy to determine the relative coupling efficiency in the wavelength interval from 740 to 840 nm. We modeled the excitation efficiency as a function of the incidence angle using a simple analytical diffraction model. Two ridges of different width (200 and 500 nm) were used to compare results and validate the model. The experimental results show a higher coupling efficiency at oblique incidence, where the coupling was enhanced by factors of 2× for the 500-nm-wide ridge, and 3× for the 200-nm-wide ridge, as well as unidirectional SPP excitation. The experimental results are in good agreement with the proposed model.  相似文献

HLA-B is the most polymorphic of the major histocompatibility complex classical class I loci. This polymorphism is mainly in exons 2 and 3, which code for the molecule’s α1 and α2 domains and include the antigenic peptide binding site. Recent studies have indicated that not only exons but also the intron 2 region may be involved in the generation of certain HLA-B alleles such as B * 3906 and B * 1522. To study the degree of intron 2 participation and the mechanisms that generate polymorphism at the HLA-B locus, intron 1 and 2 sequences from the HLA-B35, -B5, -B16 and -B15 groups of alleles were obtained. A group-specific intronic polymorphism was found: namely, B * 5301 shows intron 1 and 2 sequences identical to those found in all B35 alleles studied. On the other hand, B * 5101 and B * 52012 show the same intron 1 and 2 sequences and their intron 1 is the same as that found in the B35 group. This suggests that B5 and B35 groups of alleles may have arisen from a common ancestor. All known B16 alleles show the same introns 1 and 2, with the exception of B * 39061 and B * 39062, and all B15 alleles also bear the same introns 1 and 2, with the exception of B * 1522. Variability at intron 1 is more restricted than at intron 2, and the use of intron 1 for HLA-B allele phylogenetic analysis is better for grouping alleles of a postulated common origin. In conclusion, there is a remarkable conservation of intronic sequences within related HLA-B alleles, which probably reflects a common origin and perhaps a selective force avoiding DNA changes. J. Appl.Physiol. 81(4): 1605-1609, 1996.Thisstudy was designed to determine whether subjects born at high altitude(HA; 2,000 m or above) who subsequently move to near sea level (SL)develop end-tidal PCO2(PETCO2) andPO2(PETO2) valuesthat equal those of SL natives living near SL. A total of 108 male HAnatives living near SL were identified by survey of a district in Lima,Peru, and a further 108 male SL natives from the same district wereidentified as control subjects. Of these subjects, satisfactory datafor inclusion in the study were obtained from 93 HA and 82 SL subjects.Mean PETCO2 and PETO2 values were 37.7 ± 2.5 (SD) and 104.7 ± 3.2 Torr, respectively, in HA subjects and37.7 ± 2.2 and 104.8 ± 3.0 Torr, respectively, in SL subjects.The average difference between SL natives and HA natives forPETCO2 was 0.07 Torr(0.64 to 0.78; 95% confidence interval) and forPETO2 was 0.05 Torr(0.89 to 0.99, 95% confidence interval). The average age andweight of the SL and HA subjects did not differ, but the HA subjectswere shorter and tended to have larger vital capacities, consistentwith their origin at HA. We conclude that thePETCO2 andPETO2 near SL of SL nativesand HA natives do not differ.

Free iron concentration, as determined by electron paramagnetic resonance (EPR) spectroscopy, and lipid peroxidation (LPO), as determined by thiobarbituric acid test, were assessed in the lung, heart, liver, spleen, brain and kidney of rats subjected to experimental iron overload. Two tests, Desferal- and NO-available iron, were used to measure free iron and gave comparable results. The most pronounced accumulation of free iron was observed in liver, kidney and spleen. Differences between control and iron loaded animals increased during the initial 90 days of treatment. Between 90 and 180 days free iron concentration reached a steady state level, or even decreased, as in the case of liver. Lipid peroxidation level, measured in the organs of both treated and matched controls, did not give any significant difference during the initial 90 days of treatment. A significant augmentation was observed in liver, kidney, spleen and heart at 180 days. The results of the present research show that, under conditions of moderate siderosis, the occurrence of LPO is partially related to the level of free iron.  相似文献

The mitochondrial permeability transition pore is a recognized drug target for neurodegenerative conditions such as multiple sclerosis and for ischemia-reperfusion injury in the brain and heart. The peptidylprolyl isomerase, cyclophilin D (CypD, PPIF), is a positive regulator of the pore, and genetic down-regulation or knock-out improves outcomes in disease models. Current inhibitors of peptidylprolyl isomerases show no selectivity between the tightly conserved cyclophilin paralogs and exhibit significant off-target effects, immunosuppression, and toxicity. We therefore designed and synthesized a new mitochondrially targeted CypD inhibitor, JW47, using a quinolinium cation tethered to cyclosporine. X-ray analysis was used to validate the design concept, and biological evaluation revealed selective cellular inhibition of CypD and the permeability transition pore with reduced cellular toxicity compared with cyclosporine. In an experimental autoimmune encephalomyelitis disease model of neurodegeneration in multiple sclerosis, JW47 demonstrated significant protection of axons and improved motor assessments with minimal immunosuppression. These findings suggest that selective CypD inhibition may represent a viable therapeutic strategy for MS and identify quinolinium as a mitochondrial targeting group for in vivo use.  相似文献

Pentalinon andrieuxii (Müll .Arg .) B.F.Hansen & Wunderlin (Apocynaceae) is a vine native to the Yucatan peninsula, where it is widely used in Mayan traditional medicine to treat, among other ailments, the wounds caused by cutaneous leishmaniasis. Among the secondary metabolites isolated from Pandrieuxii are the triterpene betulinic acid and the chemically unusual tri‐norsesquiterpene urechitol A; however, to date, there is no existing knowledge about the accumulation dynamics of the ubiquitous betulinic acid or the novel urechitol A in the plant. In this article, we report on the accumulation of both secondary metabolites in wild individuals of Pandrieuxii; our results show that while the content of betulinic acid in plant leaves bears no apparent relation to plant ontogeny, the content of urechitol A in root tissue is clearly related to plant development.  相似文献

We hypothesized that analysis of single nucleotide polymorphism arrays (SNP-A) and new molecular defects may provide new insight in the pathogenesis of systemic mastocytosis (SM). SNP-A karyotyping was applied to identify recurrent areas of loss of heterozygosity and bidirectional sequencing was performed to evaluate the mutational status of TET2, DNMT3A, ASXL1, EZH2, IDH1/IDH2 and the CBL gene family. Overall survival (OS) was analyzed using the Kaplan-Meier method. We studied a total of 26 patients with SM. In 67% of SM patients, SNP-A karyotyping showed new chromosomal abnormalities including uniparental disomy of 4q and 2p spanning TET2/KIT and DNMT3A. Mutations in TET2, DNMT3A, ASXL1 and CBL were found in 23%, 12%, 12%, and 4% of SM patients, respectively. No mutations were observed in EZH2 and IDH1/IDH2. Significant differences in OS were observed for SM mutated patients grouped based on the presence of combined TET2/DNMT3A/ASXL1 mutations independent of KIT (P = 0.04) and sole TET2 mutations (P<0.001). In conclusion, TET2, DNMT3A and ASXL1 mutations are also present in mastocytosis and these mutations may affect prognosis, as demonstrated by worse OS in mutated patients.  相似文献

More freshwater ecosystems are drying in response to global change thereby posing serious threat to freshwater biota and functions. The production of desiccation‐resistant forms is an important adaptation that helps maintain biodiversity in temporary freshwaters by buffering communities from drying, but its potential to mitigate the negative effects of drying in freshwater ecosystems could vary greatly across regions and ecosystem types. We explored this context dependency by quantifying the potential contribution of desiccation‐resistance forms to invertebrate community recovery across levels of regional drying prevalence (defined as the occurrence of drying events in freshwaters in a given region) and ecosystem types (lentic, lotic) in temporary neotropical freshwaters. We first predicted that regional drying prevalence influences the selection of species with desiccation‐resistant forms from the regional species pools and thus increases the ability of communities to recover from drying. Second, we predicted lentic freshwaters harbor higher proportions of species with desiccation‐resistant forms compared to lotic, in response to contrasted hydrologic connectivity. To test these predictions, we used natural experiments to quantify the contribution of desiccation‐resistant forms to benthic invertebrate community recovery in nine intermittent streams and six geographically isolated temporary wetlands from three Bolivian regions differing in drying prevalence. The contribution of desiccation‐resistant forms to community recovery was highest where regional drying prevalence was high, suggesting the species pool was adapted to regional disturbance regimes. The contribution of desiccation‐resistant forms to community recovery was lower in streams than in wetlands, emphasizing the importance of hydrologic connectivity and associated recolonization processes from in‐stream refuges to recovery in lotic systems. In all regions, the majority of functional traits were present in desiccation‐resistant taxa indicating this adaptation may help maintain ecosystem functions by buffering communities from the loss of functional traits. Accounting for regional context and hydrologic connectivity in community recovery processes following drying can help refine predictions of freshwater biodiversity response to global change.  相似文献
