The synthesis of a series of (R)-3-[2-(2-amino)phenethyl]-1-(2,6-difluorobenzyl)-6-methyluracils containing a substituted thiophene or thiazole at C-5 is described. SAR around C-5 of the uracil led to the discovery that a 2-thienyl or (2-phenyl)thiazol-4-yl group is required for optimal receptor binding. The best compound from the series had a binding affinity of 2 nM (K(i)) for the human GnRH receptor. A novel and convenient preparation of N-1-(2,6-difluorobenzyl)-6-methyluracil is also described.  相似文献

The Streptomyces spp. are notorious plant biomass decomposers in soil environments, but only few strains were biochemically and genetically characterized. Here, we employed functional screening along with genomic sequencing for identification of novel lignocellulolytic Streptomyces strains. Streptomyces strains isolated from soil were functional screened based on their cellulolytic and hemicellulolytic capacities by enzymatic plate assays containing carboxymethylcellulose (CMC) and beechwood xylan as sole carbon source. Subsequently, genomes of Streptomyces strains were sequenced, annotated, and interpreted to correlate their genetic contents with biochemical properties. Among the 80 bacterial isolates that were screened for enzymatic activity, two Streptomyces strains (named as F1 and F7) exhiting higher endoglucanase and endoxylanase activities were selected for biochemical and genomic characterization. After cultivation on steam-pretreated sugarcane bagasse-based medium, the supernatant of the strains F1 and F7 exhibited enzymatic activity against different substrates, such as arabinan, rye arabinoxylan, β-glucan, starch, CMC, xylan, and chitin. Furthermore, strain F7 was able to degrade pectin, mannan, and lichenan. The genomic analysis of both strains revealed a diversity of carbohydrate-active enzymes. The F1 and F7 genomes encode 33 and 44 different types of glycosyl hydrolases families, respectively. Moreover, the genomic analysis also identified genes related to degradation of lignin-derived aromatic compounds. Collectively, the study revealed two novel Streptomyces strains and further insights on the degradation capability of lignocellulolytic bacteria, from which a number of technologies can arise, such as saccharification processes.  相似文献

Alien plants invasion has negative impacts on the structure and functionality of ecosystems. Understanding the determinants of this process is fundamental for addressing environmental issues, such as the water availability in South Africa’s catchments. Both environmental and anthropogenic factors determine the invasion of alien species; however, their relative importance has to be quantified. The aim of this paper was to estimate the importance of 32 explanatory variables in predicting the distribution of the major invasive alien plant species (IAPS) of South Africa, through the use of Species Distribution Models. We used data from the National Invasive Alien Plants Survey, delineated at a quaternary catchment level, coupled with climatic, land cover, edaphic, and anthropogenic variables. Using two-part generalized linear models, we compared the accuracy of two different sets of variables in predicting the spatial distribution of IAPS; the first included environmental correlates alone, and the second included both environmental and anthropogenic variables. Using Random Forest, we explored the relative importance of the variables in producing a map of potential distribution of IAPS. Results showed that the inclusion of anthropogenic variables did not significantly improve model predictions. The most important variables influencing the distribution of IAPS appeared to be the climatic ones. The modeled potential distribution was analyzed in relation to provinces, biomes, and species’ minimum residence time.  相似文献

An integrated physicochemical-biotechnological approach for a multipurpose valorization of olive mill wastewaters was studied. More than 60% of the wastewater natural polyphenols were recovered through a solid phase extraction procedure, by employing Amberlite XAD16 resin as the adsorbent and ethanol as the biocompatible desorbing phase. Thereafter, the dephenolized effluent was fed to a mesophilic anaerobic acidogenic packed-bed biofilm reactor for the bioconversion of the organic leftover into volatile fatty acids (VFAs). A VFAs concentration of 19 gCODL(-1) was obtained, representing more than 70% of the COD occurring in the anaerobic effluent. The biotechnological process was assessed by means of bio-molecular analyses, which showed that the reactor packed bed was mostly colonized by bacteria of the Firmicutes phylogenetic group. The biorefinery scheme developed in this study allowed the obtainment of 1.59 g of polyphenols per liter of wastewater treated and 2.72 gCODL(-1) day(-1) of VFAs.  相似文献

Herbaspirillum seropedicae Z67 is a nitrogen-fixing bacterium able to colonize the rhizosphere and the interior of several plants. As iron is a key element for nitrogen fixation, we examined the response of this microorganism to iron deficiency under nitrogen fixing conditions. We identified a H. seropedicae exbD gene that was induced in response to iron limitation and is involved in iron homeostasis. We found that an exbD mutant grown in iron-chelated medium is unable to fix nitrogen. Moreover, we provide evidence that expression of the nifH and nifA genes is iron dependent in a H. seropedicae genetic background.  相似文献

In the denitrification pathway, Pseudomonas aeruginosa cytochrome cd1 nitrite reductase catalyzes the reduction of nitrite to nitric oxide; in vitro, this enzyme is also competent in the reduction of O2 to 2H2O. In this article, we present a comparative kinetic study of the O2 reaction in the wild-type nitrite reductase and in three site-directed mutants (Tyr10-->Phe, His369-->Ala and His327-->Ala/His369-->Ala) of the amino acid residues close to the d1 heme on the distal side. The results clearly indicate that His369 is the key residue in the control of reactivity, as its substitution with Ala, previously shown to affect the reduction of nitrite, also impairs the reaction with O2, affecting both the properties and lifespan of the intermediate species. Our findings allow the presentation of an overall picture for the reactivity of cytochrome cd1 nitrite reductase and extend our previous conclusion that the conserved distal histidines are essential for the binding to reduced d1 heme of different anions, whether a substrate such as nitrite, a ligand such as cyanide, or an intermediate in the O2 reduction. Moreover, we propose that His369 also exerts a protective role against degradation of the d1 heme, by preventing the formation and adverse effects of the reactive O2 species (never present in significant amounts in wild-type cytochrome cd1 nitrite reductase), a finding with clear physiological implications.  相似文献

Circulating levels of the neuro-hypophysial nonapeptide oxytocin increase during sexual arousal and orgasm in both men and women. A few studies have evaluated the effect of the menstrual cycle on plasma oxytocin in normally cycling, sexually active, healthy fertile women using or not using contraceptive pills. In 20 ovulating women and 10 women taking an oral contraceptive (group 1 and group 2, respectively), sexual function, hormonal profile, and plasma oxytocin (OT) were evaluated throughout the menstrual cycle. In group 1, plasma OT was significantly lower during the luteal phase in comparison with both the follicular and ovulatory phases. Plasma oxytocin was significantly correlated with the lubrication domain of the Female Sexual Function Index (FSFI) during the luteal phase and showed a trend towards statistical significance during the follicular phase. In group 2, plasma OT did not show any significant fluctuation throughout the menstrual cycle, even though a significant correlation was evident with both the arousal and the lubrication domain of the FSFI during the assumption of the contraceptive pill. These findings suggest that plasma OT fluctuates throughout the menstrual cycle in normally cycling healthy fertile women with adequate sexual activity but not taking any oral contraceptive pill. Moreover, plasma OT levels significantly relates to the genital lubrication in both women taking and not taking oral contraceptive pill apparently confirming its role in peripheral activation of sexual function.  相似文献

When skeletal muscles are activated and mechanically shortened, the force that is produced by the muscle fibers decreases in two phases, marked by two changes in slope (P1 and P2) that happen at specific lengths (L1 and L2). We tested the hypothesis that these force transients are determined by the amount of myosin cross-bridges attached to actin and by changes in cross-bridge strain due to a changing fraction of cross-bridges in the pre-power-stroke state. Three separate experiments were performed, using skinned muscle fibers that were isolated and subsequently (i) activated at different Ca2+ concentrations (pCa2+ 4.5, 5.0, 5.5, 6.0) (n = 13), (ii) activated in the presence of blebbistatin (n = 16), and (iii) activated in the presence of blebbistatin at varying velocities (n = 5). In all experiments, a ramp shortening was imposed (amplitude 10%Lo, velocity 1 Lo•sarcomere length (SL)•s−1), from an initial SL of 2.5 µm (except by the third group, in which velocities ranged from 0.125 to 2.0 Lo•s−1). The values of P1, P2, L1, and L2 did not change with Ca2+ concentrations. Blebbistatin decreased P1, and it did not alter P2, L1, and L2. We developed a mathematical cross-bridge model comprising a load-dependent power-stroke transition and a pre-power-stroke cross-bridge state. The P1 and P2 critical points as well as the critical lengths L1 and L2 were explained qualitatively by the model, and the effects of blebbistatin inhibition on P1 were also predicted. Furthermore, the results of the model suggest that the mechanism by which blebbistatin inhibits force is by interfering with the closing of the myosin upper binding cleft, biasing cross-bridges into a pre-power-stroke state.  相似文献
