Restingas are sandy coastal plains that stand between the sea and the Brazilian Atlantic forest mountains. The predominant restinga vegetation type in northern Rio de Janeiro, Brazil, is characterized by the formation of islands that begins with colonization by some pioneer herbs and/or woody plants. Pioneer plants are stress-resistant and nurse many other less-resistant plant species. Determining the spatiotemporal variation in the dynamics of nurse plants is essential to understand the ecological functioning of restingas as a whole. The goal of this study was to analyze the spatiotemporal variation in population dynamics of the nurse bromeliad Aechmea nudicaulis. We monitored A. nudicaulis ramets in different habitats, microhabitats, and years. We analyzed the spatiotemporal variation in demographic traits and in population growth rate. Results showed young ramet traits were more variable at the microhabitat level, and when variable, vegetative ramet traits varied at all spatiotemporal scales. Overall, λ values indicated that A. nudicaulis basically remained spatiotemporally stable as most of the λ values did not significantly differ from unity. Hence, the stability of A. nudicaulis in different microhabitats and habitats in the restinga may create several settlement opportunities for many other less-resistant species.  相似文献

We describe the synthesis of new perfluorinated dimerizable detergents which contain a tricationic or tetracationic (linear or branched spermine, respectively) polar head, and report on their cmc, their ability to condense DNA into cationic monomolecular DNA nanoparticles as well as on the in vitro transfection efficiency of these nanoparticles. Such cationic nanoparticles were prone to display efficient cell transfection properties as a result of increased contact to the anionic cell surface and internalization by endocytosis, low size compatible with improved intracellular diffusion and nuclear pore crossing, and the presence of amine function of low pK(a) for their endosomal escape. The challenge was to design polymerizable polycationic detergents that display a cmc high enough for the monomer to perform monomolecular DNA condensation (as cationic particles) and low enough for the dimer to form stable nanoparticles capable of efficient cell transfection. Although we succeeded in formulating small-sized cationic monomolecular DNA nanoparticles (<40 nm) with these dimerizable perfluorinated spermine-based detergents for N/P ratios of up to 5 (N=number of detergent amine equivalents/P=number of DNA phosphate equivalents), these small-sized cationic nanoparticles proved to be poor non-specific transfection agents in vitro, even in the presence of chloroquine. Their poor transfection potential could be due more likely to Brownian motion which prevents these very small-sized particles from sedimentation and adsorption onto the adherent cell monolayer, and, consequently, from proteoglycan-triggered endocytosis.  相似文献

The D. seridosuperspecies is a complex mosaic of populations distributed over a vast part of South America and showing various degrees of genetical divergence. We have analyzed its chromosomal constitution in 16 new localities of southeastern and southern Brazil. Both the metaphase and salivary gland chromosomes show a sharp split of these populations in two groups. Four populations, fixed for inversion 2e 8and showing the type I karyotype, represent the southwestern limit of D. seridotype B, which inhabits the Cerrado in central-western Brazil. The remaining populations are homozygous for 2x 7, an inversion also fixed in the Caatinga populations of northeastern Brazil. However, their karyotype, in those populations analyzed, belong to a different type (V) from that of the Caatinga populations. Populations in this second group are polymorphic for five inversions on chromosome 2 plus another on chromosome 5 and show considerable interpopulation differentiation. The breakpoints of chromosome 2 inversions are described and the inversion loops of several heterokaryotypes are presented. Biogeographical information suggests that there are clear ecological differences between the two groups of populations as well as among the populations within the second group. The possible role of host plants in promoting the genetic divergence among the D. seridopopulations is discussed. This study evaluated a newly developed rapid malaria diagnostic test, OptiMAL Assay, to detect "Plasmodium falciparum malaria" and "non Plasmodium falciparum malaria" in blood samples from 139 individuals with a presumptive clinical diagnosis of imported malaria in Italy. OptiMAL Assay utilizes a dipstick coated with monoclonal antibodies against the intracellular metabolic enzyme, plasmodium Lactate Dehydrogenase (pLDH) present in and released from parasite-infected erythrocytes. Blood samples from 56 cases out of 139 were found "Plasmodium falciparum malaria" positive by microscopy; with these samples OptiMAL Assay and the ParaSight-F test, which is a kit detecting the P. falciparum histidin-rich protein 2 (HRP-2), showed an overall sensitivity of 83% and 94%, respectively, in comparison with microscopy. Parasitemia levels tested in the 56 P. falciparum positive blood samples by microscopy ranged from <0.004% to 20%. A correlation between sensitivity and parasitemia was evident and OptiMAL Assay and ParaSight-F test were more sensitive (96-100%; 100%) with samples with 0.1%-20% levels of parasitemia, while proved less sensitive (0-44%; 50-88%) with <0.004-0.01% levels of parasitemia.  相似文献

Leptin and melatonin play an important role in the regulation of body mass and energy balance. Both hormones show a circadian rhythm, with increasing values at night. In addition, melatonin receptors were recently described in adipocytes, where leptin is synthesized. Here, we investigated the influence of melatonin and its interaction with insulin and dexamethasone on leptin expression. Isolated rat adipocytes were incubated with melatonin (1 nM) alone or in combination with insulin (5 nM) and/or dexamethasone (7 nM) for 6 h. Melatonin or insulin alone did not affect leptin expression, but together they increased it by 120%. Dexamethasone increased leptin mRNA content (105%), and this effect was not enhanced by melatonin. Simultaneous treatment with the three hormones provoked a further increase in leptin release (250%) and leptin mRNA (100%). Melatonin prevented the forskolin-induced inhibition (95%) of leptin expression. In addition, melatonin's ability to stimulate leptin release (in the presence of insulin) was completely blocked by pertussis toxin and luzindole. To gain further insight into the molecular basis of melatonin and insulin synergism, the insulin-signaling pathway was investigated. Melatonin increased the insulin-induced insulin receptor-beta tyrosine phosphorylation, which led to an increased serine phosphorylation of the downstream convergent protein Akt. We concluded that melatonin interacts with insulin and upregulates insulin-stimulated leptin expression. These effects are caused by melatonin binding to the pertussis toxin-sensitive G(i) protein-coupled membrane receptor (MT1 subtype) and the cross talk with insulin, since insulin receptor and its convergent target Akt are coactivated by melatonin.  相似文献

Piperazinebenzylamines bearing a small N-(1-methoxy-2-propyl) side chain were found to be potent and selective antagonists of the human melanocortin-4 (MC4) receptor. Compound 7b, having K(i) values of 6.9 and 2800 nM at the human MC4 and MC3 receptors, respectively, has moderate oral bioavailability in mice, which is improved relative to the arylethyl analogues.  相似文献

The melanocortin-4 receptor (MC4R) plays an important role in the regulation of energy homeostasis. Recent studies have shown that blockade of the MC4R reverses tumor-induced weight loss in mice. Herein, we describe the synthesis and identification of potent and selective non-peptide antagonists of the human MC4R from a series of 2-ethoxycarbonylcyclohexyl-piperazines. Compound 12i was found to possess low nanomolar affinity for the MC4R, and exhibit oral bioavailability in rats. More importantly, when administered orally to mice (10 mg/kg), it led to statistically significant increases in food intake over a 24-h period.  相似文献
