The soil bacterium Bacillus subtilis can get into contact with growth-inhibiting substances, which may be of anthropogenic origin. Glyphosate is such a substance serving as a nonselective herbicide. Glyphosate specifically inhibits the 5-enolpyruvyl-shikimate-3-phosphate (EPSP) synthase, which generates an essential precursor for de novo synthesis of aromatic amino acids in plants, fungi, bacteria and archaea. Inhibition of the EPSP synthase by glyphosate results in depletion of the cellular levels of aromatic amino acids unless the environment provides them. Here, we have assessed the potential of B. subtilis to adapt to glyphosate at the genome level. In contrast to Escherichia coli, which evolves glyphosate resistance by elevating the production and decreasing the glyphosate sensitivity of the EPSP synthase, B. subtilis primarily inactivates the gltT gene encoding the high-affinity glutamate/aspartate symporter GltT. Further adaptation of the gltT mutants to glyphosate led to the inactivation of the gltP gene encoding the glutamate transporter GltP. Metabolome analyses confirmed that GltT is the major entryway of glyphosate into B. subtilis. GltP, the GltT homologue of E. coli also transports glyphosate into B. subtilis. Finally, we found that GltT is involved in uptake of the herbicide glufosinate, which inhibits the glutamine synthetase. 相似文献

The early Late Pliocene (3.6 to ∼3.0 million years ago) is the last extended interval in Earth''s history when atmospheric CO2 concentrations were comparable to today''s and global climate was warmer. Yet a severe global glaciation during marine isotope stage (MIS) M2 interrupted this phase of global warmth ∼3.30 million years ago, and is seen as a premature attempt of the climate system to establish an ice-age world. Here we propose a conceptual model for the glaciation and deglaciation of MIS M2 based on geochemical and palynological records from five marine sediment cores along a Caribbean to eastern North Atlantic transect. Our records show that increased Pacific-to-Atlantic flow via the Central American Seaway weakened the North Atlantic Current and attendant northward heat transport prior to MIS M2. The consequent cooling of the northern high latitude oceans permitted expansion of the continental ice sheets during MIS M2, despite near-modern atmospheric CO2 concentrations. Sea level drop during this glaciation halted the inflow of Pacific water to the Atlantic via the Central American Seaway, allowing the build-up of a Caribbean Warm Pool. Once this warm pool was large enough, the Gulf Stream–North Atlantic Current system was reinvigorated, leading to significant northward heat transport that terminated the glaciation. Before and after MIS M2, heat transport via the North Atlantic Current was crucial in maintaining warm climates comparable to those predicted for the end of this century. 相似文献

Infection by Shiga toxin-producing Escherichia coli causes hemorrhagic colitis, hemolytic uremic syndrome (HUS), acute renal failure, and also central nervous system complications in around 30% of the children affected. Besides, neurological deficits are one of the most unrepairable and untreatable outcomes of HUS. Study of the striatum is relevant because basal ganglia are one of the brain areas most commonly affected in patients that have suffered from HUS and since the deleterious effects of a sub-lethal dose of Shiga toxin have never been studied in the striatum, the purpose of this study was to attempt to simulate an infection by Shiga toxin-producing E. coli in a murine model. To this end, intravenous administration of a sub-lethal dose of Shiga toxin 2 (0.5 ηg per mouse) was used and the correlation between neurological manifestations and ultrastructural changes in striatal brain cells was studied in detail. Neurological manifestations included significant motor behavior abnormalities in spontaneous motor activity, gait, pelvic elevation and hind limb activity eight days after administration of the toxin. Transmission electron microscopy revealed that the toxin caused early perivascular edema two days after administration, as well as significant damage in astrocytes four days after administration and significant damage in neurons and oligodendrocytes eight days after administration. Interrupted synapses and mast cell extravasation were also found eight days after administration of the toxin. We thus conclude that the chronological order of events observed in the striatum could explain the neurological disorders found eight days after administration of the toxin. 相似文献

Aim We searched for evidence of floristic homogenization in widespread oceanic archipelagos. Location Twelve oceanic archipelagos in the Atlantic and Pacific oceans: Ascension, Azores, Canary, Cape Verde, Desventuradas, Easter, Galápagos, Hawaii, Juan Fernández, Madeira, Puerto Rico and Savage. Methods By using Jaccard’s index, we established the floristic similarity between pairs of archipelagos at two stages: original (pre‐European; Jo) and current flora (Jc). Then, we calculated ΔJ = Jc–Jo, where positive differences imply that similarity has increased floristic homogenization. Results We found that floristic similarity increased fourfold on average, from 1.6% to 6.3% for original and current floras, respectively. In fact, we recorded 64 cases in which floristic similarity increased and only two in which it decreased. The importance of invasions exceeds that of extirpations as a driver of biotic change by more than an order of magnitude (2679 versus 142 species, respectively). Main conclusions The vascular floras of these 12 insular oceanic systems have increased in compositional similarity, a phenomenon consistent with the trend towards biotic homogenization. It can be characterized as a teleconnected process that operates across vast geographical distances, driven by the unprecedented capacity of humans for translocation. Trends in biotic homogenization differ depending on the geographical location (i.e. Pacific < Atlantic archipelagos) and phylogenetic groups (island vascular plants > island birds reported in a previous study), emphasizing the complexity of biotic change. 相似文献
