Aim We searched for evidence of floristic homogenization in widespread oceanic archipelagos. Location Twelve oceanic archipelagos in the Atlantic and Pacific oceans: Ascension, Azores, Canary, Cape Verde, Desventuradas, Easter, Galápagos, Hawaii, Juan Fernández, Madeira, Puerto Rico and Savage. Methods By using Jaccard’s index, we established the floristic similarity between pairs of archipelagos at two stages: original (pre‐European; Jo) and current flora (Jc). Then, we calculated ΔJ = JcJo, where positive differences imply that similarity has increased floristic homogenization. Results We found that floristic similarity increased fourfold on average, from 1.6% to 6.3% for original and current floras, respectively. In fact, we recorded 64 cases in which floristic similarity increased and only two in which it decreased. The importance of invasions exceeds that of extirpations as a driver of biotic change by more than an order of magnitude (2679 versus 142 species, respectively). Main conclusions The vascular floras of these 12 insular oceanic systems have increased in compositional similarity, a phenomenon consistent with the trend towards biotic homogenization. It can be characterized as a teleconnected process that operates across vast geographical distances, driven by the unprecedented capacity of humans for translocation. Trends in biotic homogenization differ depending on the geographical location (i.e. Pacific < Atlantic archipelagos) and phylogenetic groups (island vascular plants > island birds reported in a previous study), emphasizing the complexity of biotic change.  相似文献

Little is known about the reproductive biology of the Crassulaceae. We studied a population of Echeveria gibbiflora in the Pedregal de San Angel ecological preserve in Mexico City, Mexico. Each flower is open and producing nectar 7 to 8 days. On the days of maximum nectar production (flowers 4–6 days old) an average of 14.5 μl accumulates in a flower per day. The maximum rate of nectar production is between 0700 and 0900 hours. The average sugar concentration in the nectar is 43.7%. In a given flower, pollen is exposed and the stigmas are receptive at the same time. The average natural fruit-set and seed-set are 56.6% and 35.5%, respectively. The pollen-ovule ratio is 124, and the plants are fully self-compatible. The flowers are pollinated by only one species of hummingbird (Cynanthus latirostris) and are never visited by insects. Pollen movement is very limited (mean of pollinator flight distances = 0.72 m, mean distance fluorescent dyes = 0.92 m). Seed dispersion is by gravity and wind, and also is very limited (an average of 1.07 m). The total genetic neighborhood area is 15 m2 to 17 m2, producing a neighborhood effective population size (Nb) of 5.01 to 39.7 individuals. This is a very small Nb, indicating that genetic drift may be a dominant force in the evolution of this species.  相似文献

In butterflies, bees, flies and true bugs specific mouthparts are in close contact or even fused to enable piercing, sucking or sponging of particular food sources. The common phenomenon behind these mouthpart types is a complex composed of several consecutive mouthparts which structurally interact during food uptake. The single mouthparts are thus only functional in conjunction with other adjacent mouthparts, which is fundamentally different to biting–chewing. It is, however, unclear when structural mouthpart interaction (SMI) evolved since this principle obviously occurred multiple times independently in several extant and extinct winged insect groups. Here, we report a new type of SMI in two of the earliest wingless hexapod lineages—Diplura and Collembola. We found that the mandible and maxilla interact with each other via an articulatory stud at the dorsal side of the maxillary stipes, and they are furthermore supported by structures of the hypopharynx and head capsule. These interactions are crucial stabilizing elements during food uptake. The presence of SMI in these ancestrally wingless insects, and its absence in those crustacean groups probably ancestral to insects, indicates that SMI is a groundplan apomorphy of insects. Our results thus contradict the currently established view of insect mouthpart evolution that biting–chewing mouthparts without any form of SMI are the ancestral configuration. Furthermore, SMIs occur in the earliest insects in a high anatomical variety. SMIs in stemgroup representatives of insects may have triggered efficient exploitation and fast adaptation to new terrestrial food sources much earlier than previously supposed.  相似文献

Previous studies have documented the detrimental effects of microgravity on human sensorimotor skills. While that work dealt with simple, laboratory-type skills, we now evaluate the effects of microgravity on a complex, realistic instrument-control skill. Twelve participants controlled a simulated power plant during the short-term microgravity intervals of parabolic flight as well as during level flight. To this end they watched multiple displays, made strategic decisions and used multiple actuators to maximize their virtual earnings from the power plant. We quantified control efficiency as the participants’ net earnings (revenue minus expenses), motor performance as hand kinematics and dynamics, and stress as cortisol level, self-assessed mood and self-assessed workload. We found that compared to normal gravity, control efficiency substantially decreased in microgravity, hand velocity slowed down, and cortisol level and perceived physical strain increased, but other stress and motor scores didn’t change. Furthermore, control efficiency was not correlated with motor and stress scores. From this we conclude that realistic instrument control was degraded in short-term microgravity. This degradation can’t be explained by the motor and/or stress indicators under study, and microgravity affected motor performance differently in our complex, realistic skill than in the simple, laboratory-type skills of earlier studies.  相似文献

In previous studies we characterized arginine transporter genes from Trypanosoma cruzi and Leishmania donovani, the etiological agents of chagas disease and kala azar, respectively, both fatal diseases in humans. Unlike arginine transporters in higher eukaryotes that transport also lysine, these parasite transporters translocate only arginine. This phenomenon prompted us to identify and characterize parasite lysine transporters. Here we demonstrate that LdAAP7 and TcAAP7 encode lysine-specific permeases in L. donovani and T. cruzi, respectively. These two lysine permeases are both members of the large amino acid/auxin permease family and share certain biochemical properties, such as specificity and Km. However, we evidence that LdAAP7 and TcAAP7 differ in their regulation and localization, such differences are likely a reflection of the dissimilar L. donovani and T. cruzi life cycles. Failed attempts to delete both alleles of LdAAP7 support the premise that this is an essential gene that encodes the only lysine permeases expressed in L. donovani promastigotes and T. cruzi epimastigotes, respectively.  相似文献

The radiosynthesis of 3-(4-[(18)F]fluorophenyl)-2-(4-methylsulfonylphenyl)-1H-indole [(18)F]-3 as potential PET radiotracer for functional characterization of cyclooxygenase-2 (COX-2) in vitro and in vivo is described. [(18)F]-3 was prepared by McMurry cyclization of a (18)F-labeled intermediate with low valent titanium and zinc via a two-step procedure in a remote controlled synthesizer unit including HPLC purification and solid phase extraction. In this way [(18)F]-3 was synthesized in 80 min synthesis time in 10% total decay corrected yield from [(18)F]fluoride in radiochemical purity >98% and a specific activity of 74-91 GBq/μmol (EOS). [(18)F]-3 was evaluated in vitro using pro-inflammatory stimulated THP-1 and COX-2 expressing tumor cell lines (FaDu, A2058, HT-29), where the radiotracer uptake was shown to be consistent with up regulated COX-2 expression. The stability of [(18)F]-3 was determined by incubation in rat whole blood and plasma in vitro and by metabolite analysis of arterial blood samples in vivo, showing with 75% of original compound after 60 min an acceptable high metabolic stability. However, no substantial tumor accumulation of [(18)F]-3 could be observed by dynamic small animal PET studies on HT-29 tumor-bearing mice in vivo. This may be due to the only moderate COX-1/COX-2 selectivity of 3 as demonstrated by both cellular and enzymatic cyclooxygenase inhibition assay in vitro. Nevertheless, the new approach first using McMurry cyclization in (18)F-chemistry gives access to (18)F-labeled diarylsubstituted heterocycles that hold promise as radiolabeled COX-2 inhibitors.  相似文献

Methyl NMR spectroscopy is a powerful tool for studying protein structure, dynamics, and interactions. Yet difficulties with resonance assignment and the low abundance of methyl groups can preclude detailed NMR studies, particularly the determination of continuous interaction surfaces. Here we present a straightforward strategy that overcomes these problems. We systematically substituted solvent-exposed residues with reporter methionines in the expected binding site and performed chemical shift perturbation (CSP) experiments using methyl-TROSY spectra. We demonstrate the utility of this approach for the interaction between the HECT domain of the Rsp5p ubiquitin ligase and its cognate E2, Ubc4. Using these mutants, we could instantaneously assign all newly arising reporter methyl signals, determine the Ubc4 interaction surface on a per-residue basis, and investigate the importance of each individual mutation for ligand binding. Our data show that methionine scanning significantly extends the applicability, information content, and spatial resolution of methyl CSP experiments.  相似文献
