The molluscan radula is a dynamic organ, both in terms of itsuse and production. New rows of teeth are constantly producedat the posterior end of the radula, while older, worn teethare shed anteriorly, producing a dynamic equilibrium. We useda cold-shock to mark the radular ribbon and measure tooth rowproduction rates in two gastropod species, Lacuna vincta (Montagu)and L. vanegata Carpenter. We found that the average tooth rowproduction rate at 10–11°C did not differ betweenthese two species, and was 2.94 (SE = 0.002) rows per day forLacuna vincta and 2.97 (SE = 0 002) for L. vanegata Inter-individualvariability in production rate was very low, and was correlatedwith shell length, smaller individuals had slightly higher productionrates. The total length of the radular nbbon varied greatlyamong individuals, ranging from 47 to 94 (2.57 to 5.68 mm) rowsin L vincta and 53 to 99 rows (2.80 to 7.14 mm) in L vanegata,and was only somewhat correlated with the length of the shelLThis great variability will result in large differences amongindividuals in the time it takes to replace the radula totally,from 14.96 to 35.44 days in L vincta and from 17 43 to 39 69days in L. vanegata. Murine monoclonal antibodies (MAbs) were produced for the detection of faba bean necrotic yellows virus (FBNYV), an isometric ssDNA virus belonging to a new, yet unnamed genus of plant viruses. A total of 19 FBNYV-specific MAbs were obtained from three fusion experiments and characterised by determining their immunoglobulin types and titres as well as their corresponding epitopes. At least six distinct epitopes were revealed on FBNYV particles of different virus isolates. Only two MAbs reacted with SDS-dissociated FBNYV virions in triple antibody sandwich (TAS)-ELISA and with viral capsid protein in Western blots. Almost all MAbs were more sensitive in detecting FBNYV in viruliferous aphids by TAS-ELISA than polyclonal anti-FBNYV IgG by double antibody sandwich ELISA and permitted virus detection in individual aphids even following short acquisition access feeding periods. Coat protein variation among FBNYV isolates and serological relatedness to taxonomically similar viruses was studied by determining the cross reactivity of these MAbs with several field isolates of FBNYV as well as with milk vetch dwarf (MDV), banana bunchy top (BBTV), and subterranean clover stunt (SCSV) viruses. Whereas none of the MAbs reacted with BBTV, only one reacted with SCSV, indicating that FBNYV and SCSV share a common epitope. By contrast, 16 of the 19 MAbs reacted with MDV, suggesting that FBNYV and MDV are serologically closely related and strains of the same virus. When all 19 MAbs produced were tested against a total of 107 samples of FBNYV collected during virus surveys in Egypt, Ethiopia, Jordan, Morocco and Syria, five MAbs showed differential reactions. While the majority of the samples reacted with all 19 MAbs, about 20% of the 107 FBNYV samples did not react with one and/or other of these five MAbs, permitting the differentiation of seven serotypes of FBNYV and suggesting a considerable coat protein variation in FBNYV isolates from the countries surveyed. The MDV isolate from Japan and five FBNYV samples from Ethiopia appeared to be the least closely related to typical FBNYV isolates by not reacting with three and four, respectively, of the five differentiating Mabs.  相似文献

The proteinase activity present in homogenates of trophozoites of Giardia lamblia , active on azocasein and urea-denaturated hemoglobin, was separated into two different enzymes by a series of purification procedures. These procedures included gel filtration on Fractogel TSK HW-55 (F), organomercurial agarose affinity chromatography, and ion exchange chromatography on DEAE-cellulose. By chromatography on Sephadex G-100, two purified enzymes exhibited relative molecular weights of Mr= 95,000 and 35,000 ± 10%, respectively. On the basis of inhibition by thiol reagents and abrogation of this effect by dithiothreitol and cysteine, they were identified as cysteine proteinases. Proteinase I (Mr= 95,000) and proteinase II (Mr= 35,000) were active against the β-chain of insulin releasing characteristic fragments. However, differences in substrate specificities of the two enzymes could be observed by using synthetic peptides that represent sequences 1–6, 8–18, and 20–30 of the insulin β-chain. Furthermore, the synthetic tetrapeptides Arg-Gly-Phe-Phe, Arg-Gly-Leu-Hyp, and Arg-Arg-Phe-Phe were hydrolyzed by the two proteinases releasing Phe-Phe and Leu-Hyp, respectively. Compared with Arg-Gly-Phe-Phe, the rates of hydrolysis of Arg-Gly-Leu-Hyp and Arg-Arg-Phe-Phe at substrate concentrations of 1 mM were 91% and 63% (proteinase I) and 80% and 57% (proteinase II), respectively.  相似文献

Mitochondrial DNA mutations create variation in the efficiency of the oxidative phosphorylation pathway and therefore cellular energy production. Mildly deleterious mutations may reduce the performance of sperm cells in particular, due to their high energy requirements and low number of mitochondria, yet have little or no effect on the viability of somatic cells or ova. Mutations will be maintained in the population, despite the fitness cost for males, because mtDNA is passed down the female line. We looked for this so-called mother's curse effect in our captive colony of European brown hares. Significantly reduced male reproductive success was detected for a divergent haplotype that could be traced back to hares imported from a remote population. Median reproductive success for these hares was 0.17 compared to 0.49 for the indigenous haplotypes (Wilcoxon rank-sum, P  = 0.002). No difference was detected for female reproductive success, nor were we able to find a nuclear DNA component to variation in male fertility. Our data are strong evidence for a mother's curse effect persisting despite multiple crossings over seven generations. These data raise important issues relating to the reproductive fitness of small or intermixing populations and have particular implications for the management of populations for conservation.  相似文献

1. Antipredator behaviour by the facultative planktivorous fish species roach (Rutilus rutilus), perch (Perca fluviatilis) and rudd (Scardinius erythrophthalmus) was studied in a multi‐year whole‐lake experiment to evaluate species‐specific behavioural and numerical responses to the stocking of pikeperch (Sander lucioperca), a predator with different foraging behaviour than the resident predators large perch (P. fluviatilis) and pike (Esox lucius). 2. Behavioural responses to pikeperch varied greatly during the night, ranging from reduced activity (roach and small perch) and a shift in habitat (roach), to no change in the habitat use and activity of rudd. The differing responses of the different planktivorous prey species highlight the potential variation in behavioural response to predation risk from species of similar vulnerability. 3. These differences had profound effects on fitness; the density of species that exhibited an antipredator response declined only slightly (roach) or even increased (small perch), whereas the density of the species that did not exhibit an antipredator response (rudd) decreased dramatically (by more than 80%). 4. The maladaptive behaviour of rudd can be explained by a ‘behavioural syndrome’, i.e. the interdependence of behaviours expressed in different contexts (feeding activity, antipredator) across different situations (different densities of predators). 5. Our study extends previous studies, that have typically been limited to more controlled situations, by illustrating the variability in intensity of phenotypic responses to predators, and the consequences for population density, in a large whole‐lake setting.  相似文献

Female Acheta domesticus L. were tested for tracking model calling songs with different syllable periods on the Kramer treadmill in the dark, under homogenous light, and in the presence of a visually attractive target (black square) positioned either perpendicular to or in front of the invisible acoustical target.
