Six microsatellite markers were isolated from Fasciola hepatica, a re‐emerging parasite that causes important veterinary and public health problems. In a sample of 52 liver flukes from a region of hyperendemicity (Bolivian Altiplano), five microsatellite were polymorphic. Our results showed that liver flukes present important genetic variability, suggesting a preferential outcrossing reproduction mode for this hermaphroditic parasite.  相似文献

ABSTRACT. We have previously reported that a β-endorphin-like substance inhibits phagocytosis in Tetrahymena perhaps by a mu-like opioid receptor. We now report a further characterization of the elements involved in the signal transduction mechanism of this opioid. Affinity chromatography followed by immunoblots of both intracellular extracts and extracellular medium reveal the presence of two main proteins of 64 and 75 kDa. These molecular weights are much higher than that of any known opioid peptide or precursor protein and suggest that we may be dealing with either a novel opioid or with proteins that by chance cross-react with anti-β-endorphin antibody. Nevertheless, when the biological activity of these proteins was tested it was found that they had an effect similar to that of mammalian β-endorphin, namely inhibition of phagocytosis by a naloxone-reversible mechanism. We have probed a size-selected Tetrahymena library with a pro-opiomelanocortin probe and have obtained several positive clones; the sequencing of their inserts should establish whether we are dealing with a bona fide member of the opioid family. Another aspect we have been studying is the G-proteins which appear to be involved in the modulation of phagocytosis. We have found, by means of Western blotting (using an antibody against the conserved GTP-binding region of the α-subunit), two bands of 51 and 59 kDa; no α-subunit of 59 kDa had been reported previously and may represent a novel G-protein. In spite of these differences, the opioid signal transduction mechanism appears to remarkably resemble that present in more complex organisms.  相似文献

ABSTRACT Serotonin and catecholamines affect the regeneration of cilia in Tetrahymena thermophila in a dose-dependent manner: micromolar concentrations are stimulatory, whereas millimolar concentrations have little or no effect. This conclusion is based on motility measurements in regenerating cells and on ciliary counts in scanning electron micrographs. In addition, the recognition mechanism for each hormone appears to be specific and independent. Our results suggest an evolutionary link with hormonal mechanisms in multicellular eukaryotes.  相似文献

Trade‐offs are thought to be important in constraining evolutionary divergence, as they may limit phenotypic diversification. Limbless animals that burrow head‐first have been suggested to be evolutionarily constrained in the development of a large head size and sexual head shape dimorphism because of potential trade‐offs associated with burrowing. Here we use an acontiine skink (Acontias percivali) to test for the existence of trade‐offs between traits thought to be important in burrowing (speed and force). As head size dimorphism has been shown to be limited in acontiine lizards, thus suggesting constraints on head size and shape, we additionally explore the potential for trade‐offs between burrowing and biting. Our data show that A. percivali uses a burrowing style different from those previously described for caecilians and amphisbaenians, which relies on the use of extensive lateral and dorsoventral head movements. Our data also show that animals use their entire bodies to generate force, as peak force was determined by total length only. Additionally, both bite force and the time needed to burrow into the substrate were principally determined by relative head width, suggesting a trade‐off between biting and burrow speed. Performance data were indeed suggestive of a correlation between bite force and the time needed to burrow, but additional data are needed to confirm this pattern. In summary, our data suggests that trade‐offs may exist, and may have been of crucial importance in shaping the evolution of head shape in A. percivali, and burrowing lizards more generally. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102 , 91–99.  相似文献

Female Aedes albopictus mosquitoes, aged 1 week, were infected with DEN-2 dengue virus. The kinetics of infection in mosquito brain and mesenteron were monitored using DNA probes with polymerase chain reaction (PCR) amplification of target DNA sequences coding for DEN-2 virus envelope protein, compared with the standard immunofluorescence assay technique (IFA). Rates of virus detection in the mesenteron of orally infected mosquitoes rose to 38% by day 4 post-inoculation, then declined until day 8, followed by irregular peaks around days 11-14 and subsequently. In mosquito head squashes, virus was detected from day 4 onwards, reaching 38% positive by day 18. Salivary glands of all the same females were found to be positive for virus by day 8 onwards. Parenterally infected Ae.albopictus females were all positive for DEN-2 in the brain and salivary glands 8 days post-inoculation. In every case, results obtained with the PCR matched those from the IFA. Our DNA probe with PCR procedure can therefore be utilized as a sensitive and reliable method for studies of DEN-2 vectors.  相似文献

We evaluated the validity of the subspecific designation for Schistocerca gregaria gregaria (Forskål) and Schistocerca gregaria flaviventris (Burmeister), isolated in distinct regions along the north–south axis of Africa. Towards this goal, we assessed the variation of multiple morphological and molecular traits within species. We first used elliptic Fourier and landmark‐based relative warps analyses to compare the size and shape of two internal and two external structures of male genitalia. We provide a discriminant function which classified the specimens with 100% accuracy and selected shape elements of the external structures only (cercus and epiproct). We also tested eight molecular markers, and because of either absence of variation or contamination by mitochondrial DNA (mtDNA)‐like sequences, we used a clone‐and‐sequence analysis of the standard cytochrome c oxidase subunit I mitochondrial DNA barcode only. We differentiated 185 true mitochondrial sequences from 66 mitochondrial DNA‐like sequences, most of which were from S. g. gregaria specimens. On the dataset of mitochondrial origin, we identified three characteristic point mutations that diagnosed the two allopatric subspecies with 94% accuracy. Minimum spanning network and parsimony tree analyses identified S. g. flaviventris as a monophyletic lineage distinct from the nominate subspecies. Accordingly, microsatellite data indicate rarely occurring admixture events only, showing that independent evolutionary history is the norm.  相似文献

A short period of heat stress can cause a significant increasein abortion frequency of floral buds and flowers in pea, buta large variability in sensitivity exists among reproductiveorgans (ROs) within a plant or between plants. We have analysedspatial distribution of abortion frequency in plants subjectedor not subjected to heat stress in: (1) four controlled environmentexperiments in which apex temperature was increased to 31 °Cfor 6 h per day for 4 d (moderate stress); (2) one field experimentwith a similar level of stress; and (3) one experiment with2 d with 33/30 °C day/night (severe stress) in additionto treatment (1). Influence of neighbouring organs on sensitivityto heat stress was analysed by partial flower removal. Severeheat stress caused rapid interruption of RO development followedby abscission while, in moderate stress, at leat 4 d elapsedbetween cessation of stress and first symptoms of abortion.In both moderate stress and control treatments, abortion frequencyfollowed a consistent pattern along the stem with no abortionon the lowest reproductive phytomeres, a temperature-dependentabortion frequency on intermediate phytomeres and 100% abortionon apical phytomeres. Regardless of temperature, this patternwas shifted by four positions if ROs were removed on the lowestfour reproductive phytomeres, and no abortion was observed ifyoung pods were sequentially removed. Moderate heat stress increasedthe effect of developing ROs on abortion frequency of youngerROs located above them. We suggest that the effect of mild stressshould be viewed as the acceleration of a programme linked tothe normal termination of phytomere production during the plantcycle, rather than as an abrupt event linked to stress.
