Raspberry bushy dwarf virus (RBDV), recently renamed to Idaeovirus rubi, is one of the most common viruses infecting Rubus species worldwide but there is still a limited number of genome sequences available in the GenBank database and the majority of the sequences include partial sequences of RNA-1 and RNA-2. The distribution and incidence of RBDV in main raspberry and blackberry growing provinces in Turkey were monitored during 2015–2019 and 537 Rubus spp. samples were tested by both DAS-ELISA and RT-PCR. Among the tested samples, 36 samples tested positive for RBDV by DAS-ELISA and 67 samples by RT-PCR. There was relatively low nucleotide diversity among the Turkish isolates. Turkish isolates shared 93%–97.7%, 84.3%–98.9%, and 85%–99.2% nucleotide sequence identities with available sequences in the GenBank, in partial RNA-1, movement protein (MP) and coat protein (CP) genes, respectively. In the phylogenetic tree constructed for RNA-1, MP, and CP sequences, all Turkish raspberry isolates were clustered in a distinct clade. However, the blackberry isolates showed considerable variation in nucleotide sequences and were placed in three distinct groups. The divergent blackberry isolates showed high variability in MP (84.5%–89.3%) and CP (85.5%–89.7%) regions and were placed in a distinct group. The rest of blackberry isolates clustered together with sweet cherry RBDV isolates adjacent to the grapevine clade or together with raspberry isolates. The comparative analysis conducted on three RNA segments of RBDV highlighted the high sequence diversity of Turkish RBDV isolates. This study also emphasizes the importance of regular monitoring of RBDV infections in Turkey, with special regard to those Rubus spp. and grapevine accessions employed in conservation and selection programmes. In particular, the presence of new RBDV genetic variants and infection of Rubus species must be taken into account to choose a correct detection protocol and management strategy. 相似文献

Bovine lactoperoxidase (LPO) was purified from skimmed milk using amberlite CG-50-H+ resin, CM sephadex C-50 ion-exchange chromatography, and sephadex G-100 gel filtration chromatography. Lactoperoxidase was purified 20.45-fold with a yield of 28.8%. Purity of enzyme checked by sodium dodecyl sulphate-polyacrylamide gel electrophoresis method and a single band was observed. K m was 0.25 mM at 20°C, V max value was 7.95 μmol/ml min at 20°C (pH 6.0). Antibacterial study was done by disk diffusion method of Kirby-Bauer using Mueller-Hinton agar medium with slight modification. Bovine LPO showed high antibacterial activity in 100 mM thiocyanate-100 mM H2O2 medium for some bacteria (Brevibacillus centrosaurus, B. choshinensis, B. lyticum, Cedecea davisae, Chryseobacterium indoltheticum, Clavibacter michiganense pv. insidiosum, Kocuria erythromyxa, K. kristinae, K. rosea, K. varians, Paenibacillus validus, Pseudomonas syringae pv. populans, Ralstonia pickettii, Rhodococcus wratislaviensis, Serratia fonticola, Streptomyces violaceusniger, Vibrio cholerae-nonO1) respectively, and compared with well known antibacterial substances (levofloxacin, netilmicin). The carbon balance of a winter wheat crop in Lonzée, Belgium, was assessed from measurements carried out at different spatial and temporal scales between November 2004 and August 2005. From eddy covariance measurements, the net ecosystem exchange was found to be ?0.63 kg C m?2 and resulted from the difference between gross primary productivity (GPP) (?1.58 kg C m?2) and total ecosystem respiration (TER) (0.95 kg C m?2). The impact of the u* threshold value on these fluxes was assessed and found to be very small. GPP assessment was partially validated by comparison with an estimation scaled up from leaf scale assimilation measurements. Soil respiration (SR), extrapolated from chamber measurements, was 0.52 kg C m?2. Net primary productivity, assessed from crop sampling, was ?0.83 kg C m?2. By combining these fluxes, the autotrophic and heterotrophic components of respiration were deduced. Autotrophic respiration dominated both TER and SR. In this study, we aimed to detect the proportion of Candida dubliniensis among yeast strains previously identified as C. albicans by using several phenotypic methods and PCR.
For this purpose, we screened 300 strains by using phenotypic tests suggested for the identification of C. dubliniensis in the literature, but we detected high proportion of false-positive reactions. Only two strains (0.6%) were detected as
true C. dubliniensis by PCR and API ID 32C methods. Moreover, these two strains gave the expected results with all the phenotypic tests, including
modified salt tolerance test for C. dubliniensis.
In conclusion, none of the phenotypic methods, except for the modified salt tolerance test, revealed 100% successful results
in discrimination of C. albicans and C. dubliniensis species. However, in the tobacco agar test, the rate of false positivity was as low as 0.6%. We suggest that in the case
of absence of PCR and other automatized identification systems, these two phenotypic tests can be used in routine laboratories

The inhibitory effect of Cd on nitrification was investigated in a continuous-flow system with enriched nitrifying bacteria.
The maximum specific ammonium utilization rate and the half-saturation constant were found as 671 mg NH4–N/g VSS day and 0.48 mg/l, respectively. In the case of continuous Cd input at 1 and 2.5 mg/l, nitrification was inhibited
by 30% and 47%, respectively. Inhibition ranged from 20% to 40% and no further increase in inhibition was exhibited in new
runs except at 10 mg/l influent Cd. At 10 mg/l influent Cd, specific ammonium utilization and nitrate production rates were
inhibited by 90%. On the contrary, a serious nitrite accumulation was not observed during this period. When Cd feeding was
stopped, recovery from inhibition was observed after 37 day which was seen by the improvement in ammonium utilization and
nitrate production rates. A shift in microbial population from the initial Nitrosomonas sp. to the Cd-tolerant Nitrosospira sp. was observed in the recovery period from severe Cd inhibition. The ability of theRhizobium ciceri, to produce extracellular polysaccharides (EPS) was investigated. Tested carbon and nitrogen sources influced EPS synthesis whenR. ciceri Rc5 was grown in a chemically defined medium. Mannitol was the most efficient carbon source among the six sources tested (sucrose, glucose, arabinose, fructose, xylose and rhamnose) and sodium nitrate was the most efficient nitrogen source among the two tested (ammonium sulphate and glycine). Poly-3-hydroxybutyrate (PHB) film pieces were degraded by sulfate reducing Desulfotomaculum sp. incubated under anaerobic laboratory conditions. Degradation started with adherence of the microbial cells and followed by formation of black colonies on the film surface. Scanning electron microscopic (SEM) observations revealed the presence of bacteria and formation of small holes on the film. After 60 days of incubation at 30°C, 10 % weight loss in polymer and 13 % sulfate reduction in the medium was observed. According to gel permeation chromatography (GPC) analysis, the molecular weight of the PHB decreased after 30 days and did not decrease further at a more extended incubation period.
