The trypanosomatid previously described as Crithidia roitmani is characterized here at the ultrastructural and biochemical levels. The data indicates that the parasite belongs to the Herpetomonas genus, and we therefore suggest the flagellate to be denominated as Herpetomonas roitmani n. comb. Cladistic analysis of isoenzyme data generated by eight different enzymes showed that the parasite presented a distinct banding pattern and could be grouped with some Herpetomonas spp., but not with Crithidia spp., used as reference strains. Accordingly, when the parasites were grown for longer periods in Roitman's defined medium, expontaneous differentiation from promastigotes to opisthomastigotes (typical of the Herpetomonas genus) occurred. Transmission electron microscopy revealed the presence of bacterium-like endosymbionts in the cytoplasm of all evolutive forms of the parasite. All morphological alterations characteristic of endosymbiont-bearing trypanosomatids could be observed. 相似文献

Since the proposition in 1975 of the European Neogene Mammal (MN) scale by Pierre Mein, the amount of taxonomical, stratigraphical and chronological information around Europe has increased exponentially. In this paper, the stratigraphical schemes of three of the best studied areas for the Lower and Middle Miocene, the Aragonian type area in Spain and the Upper Freshwater Molasse from the North Alpine Foreland Basin in Switzerland and Bavaria, are compared. The correlation of their local biostratigraphies are discussed. Sixteen rodent's events are studied and ranked in the three areas according to their local biostratigraphy. This study shows, and quantifies for the first time, the significant asynchronies of the different included rodent events. The MN-system is discussed in the light of those results. In accordance, we propose that it is still useful but only in a biochronological way, as a sequence of time-ordered reference localities allowing coarse long-distance correlations. In order to obtain better temporal resolution, this system has to be combined with local biostratigraphies that are well calibrated to the time scale, implementing the information about synchrony and diachrony of mammal events in different areas. 相似文献

Diabetes is a metabolic disease affecting approximately 300 million people worldwide. Neuropathy is one of its frequent complications, and may affect sensory, motor, and autonomic nerves. Its pathophysiology has not fully been elucidated. Several hypotheses have been proposed, and mitochondria have been suggested to play a significant role. Because of its remoteness and mid-latitude position, New Zealand lacks access to the tropical climates that might have ensured straightforward survival for frost-sensitive species during glacial times. Nevertheless, the New Zealand lowland flora retains a substantial complement of plants sourced in the tropics. While there have been extinction pulses for elements of the frost-sensitive flora under glacial/stadial regimes, the surviving remnants have been able to recolonize large areas of habitat during successive warm climate periods. Refugia for such species in stadial New Zealand are likely to have been localized and ecologically suboptimal. To examine these relationships we have applied chloroplast DNA sequence data to the investigation of phylogeographical pattern for five endemic species of Metrosideros subg. Metrosideros , a wide-ranging group of mostly frost-sensitive woody plants in New Zealand. The results of this research verify the location of two generally mooted stadial refugia for the country and provide support for the existence of a third. A simple pattern of chloroplast haplotype diversity was recorded in extra-refugial areas, compared with a greater complexity in the vicinity of the identified refugia. This pattern was independently repeated in both main islands. The proposed refugia correspond to contemporary localities of high average winter temperatures. The sharing of chloroplast haplotypes between the different species of Metrosideros examined suggests that there has been a history of repeated hybridization and introgression for these plants, possibly initiated by periods of refugial confinement. © 2004 The Linnean Society of London, Biological Journal of the Linnean Society , 2004, 83 , 399–412. 相似文献

Ten microsatellite markers for the flea beetle Phyllotreta nemorum were developed using di‐ and trinucleotide repeat‐enriched libraries. Each of these primer pairs were characterized on 96 individuals. Expected heterozygosities ranged between 0.11 and 0.84 and the number of alleles ranged between two and 14 per locus. Chromogranin A (CgA) is the most abundant granin in gastroenteropancreatic neuroendocrine tumors (GEP-NETs). As a tumor marker is moderately sensitive and nonspecific. Despite the limitations of testing methods, which require careful interpretation, especially in the case of gastrinomas, patients treated with somatostatin analogues, and poorly differentiated tumors, it is the best tumor marker in GEP-NETs and may be of value in other tumors with neuroendocrine differentiation. CgA may be used as a marker in blood or tissue samples through immunohistochemical techniques. CgA levels correlate with tumor burden and extension and may be used for diagnosis and monitoring of GEP-NETs, especially midgut carcinoids and endocrine pancreatic tumors. It is also useful as a prognostic marker for detection of recurrence and monitoring of response to different treatments. 相似文献
