Measurements of the organic carbon inventory, its stable isotopic composition and radiocarbon content were used to deduce vegetation history from two soil profiles in arboreal and grassy savanna ecotones in the Brazilian Pantanal. The Pantanal is a large floodplain area with grass-dominated lowlands subject to seasonal flooding, and arboreal savanna uplands which are only rarely flooded. Organic carbon inventories were lower in the grassy savanna site than in the upland arboreal savanna site, with carbon decreasing exponentially with depth from the surface in both profiles. Changes in 13C of soil organic matter (SOM) with depth differed markedly between the two sites. Differences in surface SOM 13C values reflect the change from C3 to C4 plants between the sites, as confirmed by measurements of 13C of vegetation and the soil surface along a transect between the upland closed-canopy forest and lowland grassy savanna. Changes of 13C in SOM with depth at both sites are larger than the 3–4 per mil increases expected from fractionation associated with organic matter decomposition. We interpret these as recording past changes in the relative abundance of C3 and C4 plants at these sites. Mass balances with 14C and 13C suggest that past vegetational changes from C3 to C4 plants in the grassy savanna, and in the deeper part of the arboreal savanna, occurred between 4600 and 11 400 BP, when major climatic changes were also observed in several places of the South American Continent. The change from C4 to C3, observed only in the upper part of the arboreal savanna, was much more recent (1400 BP), and was probably caused by a local change in the flooding regime.  相似文献

An in vitro evolution model was used to study changes in the genetic diversity of 24 strains of Pichia guilliermondii isolated from the midgut of bark beetles of the genus Dendroctonus . The genetic diversity of P. guilliermondii strains over 400 generations was analysed using multilocus enzyme electrophoresis (MLEE) and random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) markers. Resemblance relationships among strains were observed by cluster analysis. From the MLEE and RAPD markers, it was shown that the effective number of alleles, polymorphism and expected heterozygosity varied over the generations. The average heterozygosity among generations was statistically significant. Both the genetic diversity and the average heterozygosity were statistically significant among generations. The reduction in the population size from 109 to 105 yeast mL−1 associated with each transfer in P. guilliermondii strains and the clonal population structure observed along 400 generations suggest that genetic diversity changes and the observed replacement of genotypes are a consequence of a genetic drift process and not of the reproductive mode.  © 2008 The Linnean Society of London, Biological Journal of the Linnean Society , 2008, 93 , 475–486.  相似文献

1. The evolutionary advantages that have driven the evolution of sex are still very much debated, and a number of benefits of parthenogenesis over sexual reproduction have been proposed. In particular, parthenogenetic individuals are thought to exhibit higher probabilities of establishment following arrival in new, isolated habitats such as islands. 2. One notable example of parthenogenesis occurring in islands is the damselfly Ischnura hastata, an American species that has colonised the Azores archipelago, where the populations consist only of females. This is the only known example of parthenogenesis within the insect order Odonata. 3. Here, two island populations of I. hastata were studied, one in the Galapagos and one in Cuba, to test whether island colonisation is consistently associated with parthenogenesis in this species. Field capture–mark–recapture studies and laboratory rearing of field‐collected eggs were undertaken in both areas. 4. Sex ratios in the field were found to be heavily female‐biased among mature individuals; however, fertility rates of field‐collected eggs were high, and the sex ratios in the laboratory did not differ from 1 : 1. Data from laboratory rearing showed that shorter larval development times and shorter adult life spans in males result in protandry, which might explain the skewed sex ratios in the field. 5. These findings are consistent with sex differences in key demographic parameters which could predispose I. hastata to parthenogenesis. However, the Azores population of I. hastata remains the only documented case of asexual reproduction in this insect group.  相似文献

The role of six Rhizobium strains in the nitrogen metabolismof soybeans (Glycine max Merril) was studied under glasshouseand field conditions. The strains could be divided into twogroups, group I which produced a large nodule mass with relativelylow efficiency and group II which produced less nodule massbut which fixed the same amount of nitrogen. Plants inoculatedwith group I strains remobilized nitrogen faster from leavesbut also lost more nitrogen in senesced leaves. Although thetotal nitrogen transported in the xylem was similar for allstrains, plants inoculated with group I strains contained lessnitrogen in ureides in the xylem sap during the whole growthcycle. This difference was reflected in the nitrogen partitioningwithin the shoot, and smaller nitrogen harvest indexes wereobserved in these plants than in those inoculated with strainsof group II. The role of ureides in the nitrogen partitioningand grain yield was confirmed by the significant correlationbetween mean ureide content in xylem sap and nitrogen partitioningor yield. Further, nodules formed with group I strains evolvedmore hydrogen than those formed with group II strains and thepossible significance of this is discussed. We have developed a new staining procedure that combines the traditional Gram staining for bacteria and the Weber's chromotrope staining method, the standard technique for the detection of microsporidia spores in clinical Specimens. This “Gram-chromotrope” staining technique enhances the staining characteristics of microsporidia spores and facilitates the easy detection and differentiation of spores from other microorganisms that are found in clinical specimens, especially stool samples. This new technique is fast, reliable, and simple to perform, and can be easily adapted for use in clinical laboratories.  相似文献

The effects of propranolol (10−3 mM) on the surface anionic groups of Herpetomonas muscarum muscarum were analysed by cell electrophoresis, by ultrastructural cytochemistry and by identification of sialic acids using paper chromatography. Differentiation of H. muscarum muscarum induced by propranolol treatment caused a significant increase in the net negative surface charge. Binding of cationized ferritin (CF) and colloidal iron hydroxide particles was observed at the cell surface of both untreated and propranolol-treated cells. In cells incubated in the presence of the drug the CF particles were distributed in all membrane regions. However, there were small areas where the particles were absent. In H. muscarum muscarum exposed to propranolol the density of residues of sialic acid per cell was higher, and the agglutinating activity with Sendai virus was more intense. However, the pattern of sialic acid, characterized by the presence of N-acetylneuraminic acid derivative, was not modified upon cell interaction with the drug. Treatment of both control and propranolol-treated protozoa with neuraminidase significantly reduced the surface charge. These findings suggest that sialic acid residues are the major anionogenic groups exposed on the surface of H. muscarum muscarum .  相似文献

ABSTRACT. Drugs that interact with microtubules (colchicine and vinblastine) and microfilaments (cytochalasin B) partially inhibited cell growth and motility of Tritrichomonas foetus. Parasites incubated with these substances became rounded and cell division was blocked. Neither colchicine nor vinblastine disrupted the microtubules that form the peltar-axostylar system. Any one of these drugs interfered with the net negative surface charge of T. foetus as evaluated by determination of the cellular electrophoretic mobility (EPM). The decrease in the EPM of cytochalasin B-treated cells was caused by dimethylsulfoxide, which was used as solvent. Untreated cells as well as cytochalasin B-treated cells showed a uniform distribution of anionic sites on the plasma membrane as seen with cationized ferritin particles. In cells treated with colchicine or vinblastine the anionic sites were distributed in patches. These results are discussed in terms of participation of labile cytoplasmic microtubules and microfilaments in the control of the distribution of anionic sitecontaining macromolecules located on the cell surface of T. foetus.  相似文献

Granules released from human eosinophils upon interaction with Trypanosoma cruzi amastigotes in vitro were seen attached to the surface of non-internalized parasites by electron microscopy. Amastigote damage was preceded by the binding of eosinophil granule material to its membrane, and eosinophil granule major basic protein (MBP) bound to the parasite surface was readily detectable. Additional evidence of eosinophil cytotoxicity for extracellular amastigotes was the observation that amastigotes trapped between two eosinophils, without being ingested by either one, were destroyed at the interface. Amastigotes isolated from the spleens of infected mice or grown in culture were similarly sensitive to the lytic effects of purified MBP. These results demonstrate the ability of human eosinophils to lyse T. cruzi amastigotes extracellularly in the absence of antibody and suggest that MBP may be involved in the effect. Thus, eosinophils, known to be capable of destroying phagocytosed amastigotes, could also contribute to the clearance of these parasites through extracellular killing.  相似文献

The Mexican pseudothelphusid crabs are classified in one subfamily, three tribes, and 13 genera. Up to now, 56 species have been recognized, distributed in a strictly Neotropical pattern, with some of them reaching the state of Sonora on the western slope of Mexico. The tribe Pseudothelphusini is the most diverse, with five genera and 35 species, all of them endemic to Mexico: the two most species‐rich genera are Pseudothelphusa, with 23 species, and Tehuana, with eight species; Epithelphusa includes two species, whereas Disparithelphusa and Smalleyus are monotypic. The Pseudothelphusini lack an updated systematic revision, which could serve as a framework to analyse the monophyletic origin of the group, to clarify the relationships among genera and species, as well as to resolve the taxonomic status of various species complexes. In the present study, an exhaustive morphological revision was conducted using somatic and sexual characters. A phylogenetic analysis was performed using 77 characters and 183 character states, taken from 41 species. Ten trees of the same length were obtained using PAUP 4.0 through a heuristic search. The results show that the tribe as it is actually known constitutes a paraphyletic group, in which the species of Epithelphusa and Pseudothelphusa puntarenas are excluded from the internal group. According to the obtained results, the tribe Pseudothelphusini s.s. includes five genera: Smalleyus, Pseudothelphusa, Tehuana, and two new ones to accommodate Pseudothelphusa galloi and Pseudothelphusa sulcifrons, respectively. This new arrangement considers the provisional suppression of the genus Disparithelphusa, which remained as another species of Pseudothelphusa throughout the cladistic analysis. The phylogenetic results show a strong congruence with the distribution of the species, in several cases grouping species that form morphological clines along a geographical gradient. Abstract. In some insects, the finding of oviposition substrate triggers the uptake into oocytes of yolk proteins that are stored in the fat body during post‐embryonic development. The main host of the bean weevil Zabrotes subfasciatus (Coleoptera; Chrysomelidae; Bruchinae; Amblycerini), in which larval resources are the sole source for future egg maturation, is Phaseolus vulgaris. Despite not feeding as adults, females of this species are able to lay eggs after encountering host seeds but it is not known how females react to changes in the availability of bean seeds. In the present study, the behaviour of Z. subfasciatus facing two very different environments for oviposition is investigated, as well as how this influences offspring fitness. The results obtained show that females of Z. subfasciatus react to variations in the availability of seeds belonging to the same host species by adjusting egg size and number. Females on low bean seed density lay larger and fewer eggs than those on high bean seed density, demonstrating a trade‐off between these reproductive traits. Moreover, females can adjust egg size to changing levels of host availability during the first 4 days of their oviposition period. Although no difference in offspring weight is found, those from small eggs (low competition environment) result in larger adults. No response to selection on these traits after rearing beetles on the same host for 40 generations is observed. This unresponsiveness may indicate that beetle populations behave according to their reaction norm that already allows rapid adaptation to a varying amount of host‐seed availability and better exploitation of the environments of this widespread stored‐seed pest.  相似文献
