The pineal hormone melatonin activates two G-protein coupled receptors (MT1 and MT2) to regulate in part biological functions. The MT1 and MT2 melatonin receptors are heterogeneously distributed in the mammalian brain including humans. In the mouse, only a few reports have assessed the expression of the MT1 melatonin receptor expression using 2-iodomelatonin binding, in situ hybridization and/or polymerase chain reaction (PCR). Here, we described a transgenic mouse in which red fluorescence protein (RFP) is expressed under the control of the endogenous MT1 promoter, by inserting RFP cDNA at the start codon of MTNR1a gene within a bacterial artificial chromosome (BAC) and expressing this construct as a transgene. The expression of RFP in the brain of this mouse was examined either directly under a fluorescent microscope or immunohistochemically using an antibody against RFP (RFP-MT1). RFP-MT1 expression was observed in many brain regions including the subcommissural organ, parts of the ependyma lining the lateral and third ventricles, the aqueduct, the hippocampus, the cerebellum, the pars tuberalis, the habenula and the habenula commissure. This RFP-MT1 transgenic model provides a unique tool for studying the distribution of the MT1 receptor in the brain of mice, its cell-specific expression and its function in vivo. 相似文献

Of 17 species of free-living amoebae identified in various samples of salt water, only 1. Acanthamoeba polyphaga , is known to be a potential pathogen. Pregnancy block in mice requires exposure of recently mated females to
urinary pheromones of a strange male, and when working with inbred strains
this invariably requires urine from an outbred line. The pheromones which
induce oestrus and early puberty in mice have been identified as the
brevicomins and dihydrothiazoles. Since the same vomeronasal, neural and
neuroendocrine pathways are also activated in pregnancy block, these
compounds are likely candidates for pregnancy blocking pheromones. However,
these relatively simple chemicals lack the capacity to code for differing
mouse strains. Since large quantities of the polymorphic major urinary
proteins from the lipocalin family found in urine serve as transporters for
the dihydrothiazoles and brevicomins, and differ across strains, then these
proteins must participate in pheromone recognition in the context of
pregnancy block.

ABSTRACT. We demonstrated here the presence of proteins antigenically related to human erythroid spectrin in the parasitic protozoan Toxoplasma gondii . A high molecular weight doublet (M, 245-240,000), present in equimolar ratio, and low molecular weight polypeptides (M, 75,000) were reacted with monoclonal and polyclonal anti-human erythroid spectrin antibodies on electroblotted nitrocellulose sheets. Indirect immunofluorescence assay clearly showed that these proteins were localized in the anterior pole of the organism. Immunogold staining further revealed specific labeling of conoid, rhoptries, micronemes, and dense granules of the apical complex. The presence of the M, 245–240,000 doublet and the M, 75,000 spectrin-like proteins in the anterior pole of T. gondii may probably be consistent with a structural stabilizer function in its organciles which are suspected to be involved in the process of host cell invasion. 相似文献

Bittium reticulatum and Barleeia unifasciata are two of themost abundant microgastropods inhabiting the alga Gelidium latifoliumin the North of Spain. Bittium reticulatum produces one cohortannually, born in August, and living over two years. Barleeiaunifasciata shows two population groups, each one formed bytwo cohorts. There are important differences in density andgrowth rate between the two groups. The two species are separatedspatially on Gelidium. Bittium reticulatum prefers to live atthe base of the plant and Barleeia unifasciata prefers the upperpart. There are different demographic patterns in each population.This fact supports the concept of distribution in compartmentsrelative to intertidal systems. Trade‐offs are thought to be important in constraining evolutionary divergence, as they may limit phenotypic diversification. Limbless animals that burrow head‐first have been suggested to be evolutionarily constrained in the development of a large head size and sexual head shape dimorphism because of potential trade‐offs associated with burrowing. Here we use an acontiine skink (Acontias percivali) to test for the existence of trade‐offs between traits thought to be important in burrowing (speed and force). As head size dimorphism has been shown to be limited in acontiine lizards, thus suggesting constraints on head size and shape, we additionally explore the potential for trade‐offs between burrowing and biting. Our data show that A. percivali uses a burrowing style different from those previously described for caecilians and amphisbaenians, which relies on the use of extensive lateral and dorsoventral head movements. Our data also show that animals use their entire bodies to generate force, as peak force was determined by total length only. Additionally, both bite force and the time needed to burrow into the substrate were principally determined by relative head width, suggesting a trade‐off between biting and burrow speed. Performance data were indeed suggestive of a correlation between bite force and the time needed to burrow, but additional data are needed to confirm this pattern. Neotropical monkeys of the genera Cacajao, Chiropotes, and Pithecia (Pitheciidae) are considered to be highly arboreal, spending most of their time feeding and traveling in the upper canopy. Until now, the use of terrestrial substrates has not been analyzed in detail in this group. Here, we review the frequency of terrestrial use among pitheciin taxa to determine the ecological and social conditions that might lead to such behavior. We collated published and unpublished data from 14 taxa in the three genera. Data were gleaned from 53 published studies (including five on multiple pitheciin genera) and personal communications of unpublished data distributed across 31 localities. Terrestrial activity was reported in 61% of Pithecia field studies (11 of 18), in 34% of Chiropotes studies (10 of 29), and 36% of Cacajao studies (4 of 11). Within Pithecia, terrestrial behavior was more frequently reported in smaller species (e.g. P. pithecia) that are vertical clingers and leapers and make extensive use of the understory than in in the larger bodied canopy dwellers of the western Amazon (e.g. P. irrorata). Terrestrial behavior in Pithecia also occurred more frequently and lasted longer than in Cacajao or Chiropotes. An apparent association was found between flooded habitats and terrestrial activity and there is evidence of the development of a “local pattern” of terrestrial use in some populations. Seasonal fruit availability also may stimulate terrestrial behavior. Individuals also descended to the ground when visiting mineral licks, escaping predators, and responding to accidents such as a dropped infant. Overall, the results of this review emphasize that terrestrial use is rare among the pitheciins in general and is usually associated with the exploitation of specific resources or habitat types. Am.
