Morphological and biochemical properties ofStreptomyces hygroscopicus grown in chemostat

The basic regulation of macromoleoular synthesis in mycelium-forming bacteria is in principle the same as in unicellular bacteria, but modified by morphological peculiarities. The higher the specific growth rate the moreStreptomyces resembles unicellular bacteria.     

THE ANATOMY AND RELATIONSHIPS OF ELACHISINA DALL (GASTROPODA: RISSOACEA)

Species of Elachisina are marine and extend from the Caribbeanand western North and Central America to the Philippines, Hawaiiand New Zealand. The anatomy and other morphological featuresof Elachisina ( = Microdocluis) floridana (Rehder) from theFlorida Keys are described and compared with rissoacean familiesjudged to be most closely allied. This species has a uniquecombination of characters which do not correspond to those character-setsused to define any known rissoacean family. Important featuresinclude a metapodial as well as pallial tentacles, a wide ctenidiumwith low, triangular filaments, an osphradium which is shortrelative to the ctenidium, an S-shaped loop of the intestinewhich extends into the pallial roof, long dorsal folds in themidoesophagus, a rather loosely organised nervous system, anopen prostate gland from which the pallial vas deferens exitsat its anterior extremity, glandular lobes on the penis, a non-glandularfold on the right side of the ventral channel of the capsulegland and a fold on the left side in the posterior end of thechannel, a pair of seminal receptacles lying between the albumenand capsule glands, a bursa which lies alongside the capsulegland and opens posteriorly to the ventral channel and a U-shaped,expanded section of the oviduct proximal to the albumen glandand seminal receptacles. This combination of characters is consideredto be sufficiently distinct to arrant the erection of a newfamily-level taxon to accommodate the small group of speciesjudged to be congeneric with E. floridana. The Elachisinidaeappears to be most closely related to the Rissoidae, Iravadiidaeand Hydrobiidae. (Received 19 October 1983;     

Dynamics and management of some coastal dune woodlands near The Hague,The Netherlands

Changes are reported of Crataegus-Betula dune woodlands between 1950–1980 from Meijendel, a calcareous coastal dune system near the city of The Hague, The Netherlands, which is used as a water catchment area. It concerns both woodlands in degradation stage and, more common, woodlands on the increase. Changes were recorded with help of successive vegetation maps, air photos and permanent plot observations. Woodland increase usually occurs on places with groundwater at or near the surface, either because of natural circumstances or as a result of artificial groundwater recharge through infiltration as part of the water catchment activities. Enzymes of cyclic nucleotide degradation in eye tissues during experimental ocular herpes

Activity of phosphodiesterases disintegrating cAMP and cGMP in the cornea, sclera and ciliary body was investigated in health and in different stages of experimental herpetic keratitis. The problems concerning the role of the cyclase system in the pathogenesis of herpetic keratitis and the possibility of applying some of the drugs to the disease treatment are discussed.     

DETERMINING THE LABELING INDEX IN AUTORADIOGRAPHY

A method is proposed for estimating the labeling index in autoradiographs. It is based on a model for the frequency distribution of grain counts for those cells which have few grains. The method does not require extensive grain counting and provides reasonable estimates even when the background level is high. It is particularly suited for experiments where tumor cells, labeled in vivo, are, autoradiographed after being cultured in vitro.     

Low dose intraarterial cis-platinum/ radiotherapy after local tumour preradiation

Summary The simultaneous use of intraarterial Cis-Platinum and Radiotherapy (CP/RT) was found to be a very effective and relatively little burdened treatment for a palliative treatment concept. This affects life quality as well as the remission - and survival times. The fast and continual remission with low CP/RT concentrations, even in extreme palliative cases, is surprising. CP/RT treatment shows additive and synergistic effects which are not explainable by the single effects of the cis-platinum dose used (60 mg/1.73 m² in our case) or the total irradiation dose (e.g., 5 Gy TD) or the fractionation (e.g., 5 × 1 Gy), especially since the doses of each which were used are by themselves without therapeutic relevance. Only the combination of the modalities with a low dose two-day preradiation program induced the described effects.Dedicated to Prof. L.E. Feinendegen on the occasion of his 60th birthday     

Intrinsic fluorescence of elongation factor Tu in its complexes with GDP and elongation factor Ts

The intrinsic fluorescence properties of elongation factor Tu (EF-Tu) in its complexes with GDP and elongation factor Ts (EF-Ts) have been investigated. The emission spectra for both complexes are dominated by the tyrosine contribution upon excitation at 280 nm whereas excitation at 300 nm leads to exclusive emission from the single tryptophan residue (Trp-184) of EF-Tu. The fluorescence lifetime of this tryptophan residue in both complexes was investigated by using a multifrequency phase fluorometer which achieves a broad range of modulation frequencies utilizing the harmonic content of a mode-locked laser. These results indicated a heterogeneous emission with major components near 4.8 ns for both complexes. Quenching experiments on both complexes indicated limited accessibility of the tryptophan residue to acrylamide and virtually no accessibility to iodide ion. The quenching patterns exhibited by EF-Tu-GDP and EF-Tu X EF-Ts were, however, different; both quenchers were more efficient at quenching the emission from the EF-Tu x EF-Ts complex. Steady-state and dynamic polarization measurements revealed limited local mobility for the tryptophan in the EF-Tu x GDP complex whereas formation of the EF-Tu x EF-Ts complex led to a dramatic increase in this local mobility.     

Quantal sarcomere-length changes in relaxed single myofibrils.

We carried out experiments on single isolated myofibrils in which thin filaments had been functionally removed, leaving the connecting (titin) filaments as the sole agent taking up the length change. With technical advances that gave sub-nanometer detectability we examined the time course of single sarcomere-length change when the myofibril was ramp-released or ramp-stretched by a motor. The sarcomere-length change was stepwise. Step sizes followed a consistent pattern: the smallest was approximately 2.3 nm, and others were integer multiples of that value. The approximately 2.3-nm step quantum is the smallest consistent biomechanical event ever demonstrated. Although the length change must involve the connecting filament, the size of the quantum is an order of magnitude smaller than anticipated from folding of Ig- or fibronectin-like domains, implying either that folding occurs in sub-domain units or that other mechanisms are involved.
