A structural gene which codes for an extracellular protease in Aeromonas hydrophilia SO2/2 and D13 was cloned in Escherichia coli C600-1 by using pBR322 as a vector. The gene codes for a temperature-stable protease with a molecular mass of approximately 38,000 daltons. The protein was secreted to the periplasm of E. coli C600-1 and purified by osmotic shock. Cloned protease (P3) was identical in molecular mass and properties to the one purified from A. hydrophila SO2/2 culture supernatant as an extracellular product.  相似文献

Major histocompatibility complex (MHC) genes are central for the adaptive immune response against parasites. Here, we investigated potential associations among MHC‐I alleles and blood parasite infections in a natural breeding population of a passerine bird, the blue tit Cyanistes caeruleus, in central Spain. We screened both infection status (presence/absence of infection) and infection intensity to the pathogenic blood parasites Haemoproteus and Leucocytozoon. Three MHC‐I alleles (UA104, UA108 and UA117) were associated with higher or lower infection intensities by Leucocytozoon. Interestingly, these associations were dependent on age and were found both among young and adult birds. No MHC alleles were associated with infection intensity by Haemoproteus parasites. In addition, no significant relationships were detected between infection status by Haemoproteus and Leucocytozoon infections and MHC alleles. The very high prevalence of these two parasites in our study population (79–100%) poses challenges to identify associations with infection status and also suggests that clearance of infections may be rare. In conclusion, associations between specific MHC‐I alleles and Leucocytozoon parasites were related to either high or low infection intensities, and hence increased susceptibility or resistance to infection.  相似文献

1. The in vitro incorporation of 35S-methionine into actin and total soluble proteins, as well as the levels of actin mRNA, were studied in the hypothalamus and frontal cerebral cortex of adult male rats killed at six different time intervals during a 24-hr cycle. 2. The specific activity of total soluble proteins after labeled methionine incubations did not vary as a function of time of day in any of the examined brain regions. 3. The incorporation of 35S-methionine into a 43-kDa protein, corresponding to the electrophoretic mobility of actin, varied diurnally in the hypothalamus, exhibiting a maximum at 1200 hr. Such a diurnal variation was not found in frontal cerebral cortex. 4. Similar results were obtained when labeled methionine incorporation into actin was assessed in hypothalamus and cerebral cortex by an immunoprecipitation procedure. 5. An increase in actin hypothalamic mRNA levels, quantitated by dot-blot analysis, was found at 0800, 4 hr in advance to the maximum in 35S-methionine incorporation to actin. 6. The levels of actin mRNA did not vary significantly as a function of time of day in the frontal cerebral cortex.  相似文献

Spores of the Phycomyces blakesleeanus strain S440 germinated only for some 4 to 7% when activated with a heat treatment or with ammonium acetate. Contrary to wild type spores, they showed no increase in trehalase activity during or after the activating treatment. This was not due to a variant trehalase or a defective protein kinase but rather to the absence of an increase in cellular cyclic AMP which normally occurs in the wild type. Phosphodiesterase activity in the mutant was comparable to wild type activity and in both strains phosphodiesterase was inactivated by a heat treatment. The phosphodiesterase inhibitor 1-isobutyl, 3-methyl xanthine caused germination and trehalase activation in the wild type but not in the mutant. The results corroborate the importance of cyclic AMP in the breaking of dormancy and the activation of trehalase in this fungus.  相似文献

Thy-1 is a developmentally regulated surface glycoprotein expressed on a number of tissues, including nerve where it is a major surface component of mature neurons. During neural development in the rat and mouse, expression of Thy-1 protein does not necessarily follow appearance of its mRNA, but additionally requires completion of the initial phase of axonal growth. Where there is a substantial lag phase between initial elongation and final axonal outgrowth into a terminal field (e.g. pontine projection to the cerebellum), Thy-1 protein appears at the cell body and dendrites of the neurons, but is excluded from their axons until the terminal phase of axonal growth is completed. In the more complex case of the vestibular ganglion neurons, whose axons project primarily to the vestibular nuclei in the brainstem before birth, and then 1-2 weeks later into the cerebellum, Thy-1 enters the proximal axonal regions where growth is completed, but not the distal growing ends. Thus complex controls govern the initial expression and distribution of Thy-1 so as to exclude it from growing regions of axons.  相似文献

During recent studies in the Santolina rosmarinifolia L. aggregate, nomenclatural problems with the the names S. impressa and S. semidentata published by Hoffmannsegg and Link were revealed. Here, both names are neotypified and S. rosmarinifolia var. cinerea Pau & Merino and S. zamorana Losa are lectotypified. The criteria used to recognise S. impressa and S. semidentata and their diagnostic characters are discussed. Revision of the lectotypes of S. rosmarinifolia var. cinerea Pau & Merino and of S. zamorana Losa suggests that both names are synonyms of S. semidentata. The full synonymy of S. impressa and S. semidentata is provided.  相似文献
