We are constructing high-resolution, chromosomal 'test' maps for the entire pig genome using a 12,000-rad WG-RH panel (IMNpRH2(12,000-rad))to provide a scaffold for the rapid assembly of the porcine genome sequence. Here we present an initial, comparative map of human chromosome (HSA) 11 with pig chromosomes (SSC) 2p and 9p. Two sets of RH mapping vectors were used to construct the RH framework (FW) maps for SSC2p and SSC9p. One set of 590 markers, including 131 microsatellites (MSs), 364 genes/ESTs, and 95 BAC end sequences (BESs) was typed on the IMNpRH2(12,000-rad) panel. A second set of 271 markers (28 MSs, 138 genes/ESTs, and 105 BESs) was typed on the IMpRH(7,000-rad) panel. The two data sets were merged into a single data-set of 655 markers of which 206 markers were typed on both panels. Two large linkage groups of 72 and 194 markers were assigned to SSC2p, and two linkage groups of 84 and 168 markers to SSC9p at a two-point LOD score of 10. A total of 126 and 114 FW markers were ordered with a likelihood ratio of 1000:1 to the SSC2p and SSC9p RH(12,000-rad) FW maps, respectively, with an accumulated map distance of 4046.5 cR(12,000 )and 1355.2 cR(7,000 )for SSC2p, and 4244.1 cR(12,000) and 1802.5 cR(7,000) for SSC9p. The kb/cR ratio in the IMNpRH2(12,000-rad) FW maps was 15.8 for SSC2p, and 15.4 for SSC9p, while the ratio in the IMpRH(7,000-rad) FW maps was 47.1 and 36.3, respectively, or an approximately 3.0-fold increase in map resolution in the IMNpRH(12,000-rad) panel over the IMpRH(7,000-rad) panel. The integrated IMNpRH(12,000-rad) andIMpRH(7,000-rad) maps as well as the genetic and BAC FPC maps provide an inclusive comparative map between SSC2p, SSC9p and HSA11 to close potential gaps between contigs prior to sequencing, and to identify regions where potential problems may arise in sequence assembly.  相似文献

The kinetic parameters, kcat and KM, for the hydrolysis of N-α-tosyl-L-arginine methyl ester (1, TAME) by the wild-type subtilisins Carlsberg and BPN′ as well as the BPN′ mutants Glyl66Ser, GLyl66Asn, and Met222Phe, were determined in the presence of 5 and 15% (v/v) of a selection of water-soluble organic solvents. The goals were to compare and evaluate the solvent effects with a view to expanding their use in organic synthetic applications of the WT and mutant subtilisins. The results showed that subtilisin BPN′ and its mutants were much less affected by organic solvents than subtilisin Carlsberg. The BPN′ mutant Met222Phe demonstrated the greatest resistance to cosolvent inactivation, making it a particularly attractive mutant for peptide synthesis. Dimethyl sulfoxide, acetone, and branched alcohols were found to be the most benign solvents, whereas dioxane, THF, and N-methyl-2-pyrrolidinone seriously reduced catalytic activities, even at low concentrations. The results parallel the solvent-effect data available for other proteinases, including α-chymotrypsin.  相似文献

An age-dependent increase of a cathepsin D-like protease activity that preferentially degrades high molecular weight microtubule-associated proteins (MAPs) has been previously described. Microtubules (MT) purified from rat brain of different ages in the presence of several protease inhibitors retained undegraded MAPs through cycles of polymerization, and revealed several age-dependent changes in the relative amounts of MAPs and MT-associated kinases. MAP2 immunoreactivity was found significantly lower in MT preparations from aged animals in contrast with a relative increase of tau molecules. In addition, the phosphorylation of MAP2 by its associated cyclic AMP-dependent protein kinase was also altered, consecutively to the partial loss of the enzyme during polymerization cycles and an age-dependent decrease in the ability of the cyclic nucleotide to stimulate MAP2-bound kinase activity. The evidence of an unusually high packing density of sedimented MT from old rat brains further suggested the modification with aging of the physical structure of the arm-like projections of MAPs, in addition to a lower amount in high molecular weight MAPs. These results support the hypothesis of a selective alteration with aging of the mechanical and regulatory properties of brain MT, consecutive to a change in the composition and/or the structure of MAPs.  相似文献

Formation of 1-electron oxidation products of aromatic amines in biological systems have been ascertained. The mechanisms of the toxic actions of the aminyl radicals and their corresponding detoxication reactions are much less established. During the studies of reactions of GSH with the N,N,N',N'-tetramethyl-p-phenylenediamine radical cation (TMPD) (Wurster's blue) two pathways were detected: (1) a slow second order reaction (k = 5 M-1.s-1) which gave the parent amine and (ultimately) GSSG, and (2) a fast, complex reaction which yielded 2-(glutathione-S-yl)-N,N,N',N'-tetramethyl-p-phenylenediamine (2-GS-TMPD). From kinetic reasons, this reaction was suggested to be composed of a rapid disproportionation reaction followed by a reductive 1,4-Michael-addition. This reaction pathway prevailed at GSH concentrations below 1 mM. At higher GSH concentrations formation of the thioether was suppressed. This hypothesis was confirmed when the reaction of the highly labile N,N,N',N'-tetramethyl-p-quinonediiminium dication (TMQDI++) with GSH was followed: In this case, thioether formation outweighed clearly reductive mechanisms, the latter yielding ultimately the amine and GSSG. Similar to N,N,N',N'-tetramethyl-p-phenylenediamine (TMPD), 2-GS-TMPD was also capable of producing ferrihemoglobin in a catalytic reaction. Its rate, however, was only 3% that observed with the parent amine. During this reaction the thioether was apparently oxidized to the corresponding quinonediiminium dication, which gave the corresponding quinonemonoimine on acidification.  相似文献

Nitrosobenzene (NOB) formed acid labile conjugates with reduced glutathione (GSH) and hemoglobin within red cells. In vitro, NOB rapidly reacted with GSH with formation of phenylhydroxylamine (PH), oxidized glutathione (GSSG), and a water-soluble compound identified as glutathionesulfinanilide (GSO-AN). Free aniline (AN), aminophenols and azoxybenzene were not detected. The proportion of PH formed increased with increasing GSH concentration and at higher pH values. Spectroscopic analysis revealed the formation of a labile adduct following a second order reaction (K = 5 x 10(3) M-1 . sec-1 at pH 7.4 and 37 degrees). This reaction was reversible because nearly all NOB could be extracted with ether from the labile intermediate. On the other hand, the labile intermediate was transformed into GSO-AN (with increasing rate at lower pH values) or it was cleaved by GSH with formation of GSSG and PH. Intermediate formation of NOB and thiol radicals was ruled out by analysis of the equilibrium data. A tentative scheme is presented for the proposed reaction mechanism.  相似文献

The isolated, electrically-driven, guinea pig left atrium was used to study the ability of two perfluorocarbon emulsions to prevent anaerobic hypofunction in the myocardium. A 20% perfluorodecalin (PFD) emulsion maintained peak tension at 80% of aerated levels for more than 20 minutes. Emulsions of perfluorooctylbromide (PFOB) ranging from 25 to 100% produced a similar result, except that the 25% emulsion could not maintain contractions for the entire time period. Peak tension of atria bathed in K-H solution decreased to less than 50% over the same time period. Both 20% PFD and 100% PFOB maintained myocardial ATP levels at pre-hypoxic levels for at least twenty minutes after aeration was terminated. Unaerated atria, bathed in Krebs-Henseleit solution only, exhibited a significant decline in tissue ATP levels at this time. It appears that perfluorocarbon emulsions may delay oxygen desaturation and thereby protect cardiac tissue from ATP depletion and impaired cardiac function associated with hypoxia. This tissue preparation was found to be very useful for determining the efficacy of potential oxygen carriers.  相似文献
